目的　研究羊膜培养液对小鼠角膜新生血管的抑制作用及机制。方法　应用cornealmicropocket方法 ,将含 10 0ng碱性成纤维细胞生长因子 (bFGF)和 12 %hydronpolymer的缓释颗粒植入角膜层间 ,制备小鼠角膜新生血管模型。实验分对照组、羊膜组和去上皮羊膜组 ,每组 10只眼 ,收集各组培养液于模型制作当日起 ,每日滴眼 4次至术后 7d摄片观测角膜新生血管长入情况并处死动物。对脐静脉血管内皮细胞 (HUVEC)进行体外培养 ,并应用Boyden小房技术和荧光结合(CyQUANT)实验分别检测羊膜培养液对HUVEC细胞迁移和细胞增殖的影响。利用酶联免疫吸附(ELISA)法检测各组培养液中金属蛋白酶抑制剂 1(TIMP1)和 2 (TIMP2 )的含量。结果　羊膜培养液明显抑制由bFGF诱发的小鼠角膜新生血管的形成 ,对照组、羊膜组和去上皮羊膜组角膜新生血管面积分别是 (2 4 8± 0 76 )mm2 、(0 6 4± 0 5 2 )mm2 和 (1 96± 0 6 5 )mm2 。羊膜培养液明显抑制血管内皮细胞的迁移和增殖 ,而对照组和去上皮羊膜组对HUVEC细胞迁移无作用。羊膜培养液中TIMP2水平明显增高 ,而TIMP1的含量无变化。结论　羊膜培养液中TIMP1的含量未增加。而羊膜上皮产生或释放大量的TIMP2 ,抑制血管内皮细胞的迁移和增殖。

目的　利用兔去上皮角膜模型 ,研究血小板活化因子 (PAF)对角膜伤口愈合的作用及其分子生物学机制。方法　离体角膜上做正中直径 7mm圆形去上皮角膜伤口。去上皮角膜分为 3组 ,即对照、PAF及BN (PAF拮抗剂 )组 ,培养 4 8h后 ,行角膜上皮染色观察伤口愈合状况。分别对兔角膜上皮 (RCE)和角膜基质 (RCK)细胞进行体外传代培养 ,RCE和RCK细胞经PAF和 (或 )BN处理 ,培养 2 4h ,提纯RNA。应用RT PCR及核酸杂交技术分别检测肝细胞生长因子 (HGF)、角质形成生长因子 (KGF)及表皮生长因子 (EGF)基因在RCK和RCE细胞及HGF受体 (HGF R)基因在RCE细胞中的表达强度。分别应用CyQUANT荧光结合和Boyden小房技术检测PAF对RCE细胞黏附、增殖和迁徙的影响。结果　PAF (10 0nmol/L )明显抑制角膜上皮伤口愈合 ,4 8h对照、PAF和BN组角膜上皮未愈合面积经电脑图像分析分别为 :(6 0± 1.5 )U、(5 8 0± 7 0 )U和 (5 0± 1 0 )U。PAF明显增强RCE细胞黏附作用 ,对照、PAF和BN组每 96孔板贴附细胞数荧光光度平均值分别为 :36 96± 372、790 8± 6 71和 3487± 32 4。RT PCR结果显示 :PAF使HGFmRNA在RCK的表达强度降低 4 .1倍 ,同时明显减弱HGF R在RCE细胞中的表达 ,核酸杂交实验证实PCR结果。结论PAF明显增强RCE细胞的黏附作用 ,

Distribution of Candida Species and Molecular Typing of C. albicans Isolates in a Mexico City Tertiary Care Hospital from 2011 to 2013

The incidence of yeast infections has increased significantly over the past few years, constituting a leading cause of morbidity and mortality among hospitalised patients. The rapid identification of candidiasis is important for the clinical management of patients and to facilitate tracing the sources of infections in hospitalized patients. Here, we report a retrospective, single-centre study of Candida spp. distribution and antifungal susceptibility from January 2011 to May 2013 at a hospital in México City, regarding the importance of elucidating the identity of the infection-causing Candida species in order to improve prophylactic measures and treatment. Clinical data were collected from patient medical records and the laboratory database. Isolates were initially identified using standard mycology techniques, and then confirmed by PCR-based system using amplification of intergenic spacers (rDNA ITS) and restriction length polymorphism of PCR products after sequence-specific enzymatic cleavage (PCR-RFLP). We observed no shift from C. albicans to non-albicans Candida species: Candida albicans (73.7%) was the most prevalent species isolated, while C. dubliniensis was not identified in this study. Antifungal susceptibility was determined using FUNGITEST®17.4% of C. albicans isolates were resistant to fluconazole and 21.7% to itraconazole. Multiplex PCR microsatellite analysis of the clinical C. albicans isolates using primers for the CAI, CAIII and CAVI loci identified 29 different alleles for CAI, 8 alleles for CAIII and 31 for CAVI. The combined discriminatory power of these three microsatellites was 0.98, which was considered reliable for molecular typing. Genetic analysis of these isolates revealed a clonal population with a total of 62 genotypes from the examined isolates

Extraction and Characterization of Starch Fractions of Five Phenotypes <i>Pachyrhizus tuberosus</i>(Lam.) Spreng

Pachyrhizus tuberosus is a native plant of short life cycle found in South America riverside, which provides easy starch extraction from its tuberous roots. The aim of this study was to determine the physicochemical, rheological and functionality of the starch granules extracted from the roots of five phenotypes identified as V2, V3, V4, V6 and V7. Protein and ash content of all phenotypes were considerable high when compared to other root sources such as cassava varying from 4.35% to 7.43% and 1.58% to 2.49%, respectively, whereas lipid content was lower, between 0.29 and 0.49%. The starch granules were mostly circular and polygonal with varied sizes. The starch granules structural conformation showed cristallinity A type, normally for cereals. The maximum pasting viscosity at 95℃ ranged from 1644 cP (V7) to 2232 cP (V2). The initial temperature of pasting formation occurred at 69.4℃ for V2, 71.5℃ for V3, 87.9℃ for V4, 69.5℃ for V6 and 71.5℃ for V7. These values showed high variability within the phenotypes and generally high for roots and tubers starches. The maximum viscosity at 95℃ for V2, V3, V4, V6 and V7 were 2232, 2150, 1995, 2214 and 1644 cP, respectively. The viscosity curves showed low tendency to retrogradation. The thermal properties showed that the enthalpy of gelatinization varied from 8.91 J/g (V3) to 11.78 J/g (V2). The initial gelatinization temperature varied from 63.19℃ (V6) to 65.14℃ (V4). The swelling power at 90℃ ranged from 14.7% to 20.1% p/p and solubility from 10.3% (V2) to 27.2% (V7). It is concluded that Pachyrhizus tuberosus starch showed low retrogradation (1320 - 1560 cP) comparable to non-common native waxy starches, a feature which indicates the capability of using this natural and easy extraction starch source as gelling agent in certain manufactured food of undesirable retrogradation.

Microevolution of <i>Candida albicans</i>Isolate from a Patient with Mucocutaneous Candidiasis and HIV Infection

Candidiasis is the most common opportunistic fungal infection in HIV patients, and its presence is ascribed mainly to the persistence of the original infecting strain. The latter might acquire genetic variations during interaction with the host, reflecting the adaptation of the strain. Here, we report the case of a 32-year-old man complaining of asthenia, irregular hyperpyrexia, and dry cough, who was admitted to the emergency unit. Laboratory examination showed positivity for HIV. Dark violet macular lesions and ulcerated lesions with verrucous erosion were observed at the tip of the nose, whereas an ulcer without exudates was noted in the pubic region. Candida albicans was recovered from the skin by scraping these lesions. Cultures from the bronchoalveolar lavage (BAL) were negative for bacteria and opportunistic fungi but were positive for Candida albicans. The isolates from the skin and BAL were typed by PCR-RFLP and Candida albicans was identified. Analysis by microsatellite length polymorphisms, established that the pubic isolate was a genetic variant of the isolate from the nose and mouth. This suggested a microevolutionary event. Despite clinical support, the patient died of multiple organ failure.

Duodenal tuberculosis presenting as gastric outlet obstruction:A case report

Gastric outlet obstruction is commonly associated with malignancies and peptic ulcer disease. However, when no malignancy is seen and the patient is nonresponsive to conventional peptic ulcer treatment, other etiologies need to be explored. We report a case of gastric outlet obstruction due to duodenal tuberculosis. The patient is a 31 year old male who presented with 1 year history of recurrent epigastric pain and an a cute episode of vom iting. Endoscopy revealed duodenal stricture. Computed tomography scan showed pyloro antral thickening. The patient was referred to the surgery service and underwent an exp lor atory laparotomy and gastrojejunostomy. A duodenal mass and calcified lymph nodes were noted on exploration and biopsy revealed a tuberculous origin. The patient was started on anti-tuberculosis medications and had impro ved on discharge. Gastroduodenal tuber culosis is rare and pyloric stenosis resulting from tuber culosis is even rarer. This, however, should be consid ered in patien ts who come from areas where the dise ase is endemic.
