SBL-JP-0004:A promising dual inhibitor of JAK2 and PI3KCD against gastric cancer

Background:Gastric cancer(GC)remains a global health burden and is often characterized by heterogeneous molecular profiles and resistance to conventional *** phosphoinositide 3-kinase and PI3K and Janus kinase(JAK)signal transducer and activator of transcription(JAK-STAT)pathways play pivotal roles in GC progression,making them attractive targets for therapeutic ***:This study applied a computational and molecular dynamics simulation approach to identify and characterize SBL-JP-0004 as a potential dual inhibitor of JAK2 and PI3KCD *** and SNU-5 GC cells were used for in vitro ***:SBL-JP-0004 exhibited a robust binding affinity for JAK2 and PI3KCD kinases,as evidenced by molecular docking scores and molecular dynamics *** interactions and Gibbs binding free energy estimates confirmed stable and favorable interactions with target ***-JP-0004 displayed an half-maximal inhibitory concentration(IC_(50))value of 118.9 nm against JAK2 kinase and 200.9 nm against PI3KCD ***-JP-0004 exhibited potent inhibition of cell proliferation in KATOIII and SNU-5 cells,with half-maximal growth inhibitory concentration(GI50)values of 250.8 and 516.3 nm,respectively.A significant elevation in the early phase apoptosis(28.53%in KATOIII cells and 26.85%in SNU-5 cells)and late phase apoptosis(17.37%in KATOIII cells and 10.05%in SNU-5 cells)were observed with SBL-JP-0004 treatment compared to 2.1%and 2.83%in their respective ***:The results highlight SBL-JP-0004 as a promising dual inhibitor targeting JAK2 and PI3KCD kinases for treating GC and warrant further preclinical and clinical investigations to validate its utility in clinical settings.

Spinal cord injury remains a major cause of disability in young adults,and beyond acute decompression and rehabilitation,there are no pharmacological treatments to limit the progression of injury and optimize recovery in this *** the thorough investigation of the complement system in triggering and propagating cerebral neuroinflammation,a similar role for complement in spinal neuroinflammation is a focus of ongoing *** this work,we survey the current literature investigating the role of complement in spinal cord injury including the sources of complement proteins,triggers of complement activation,and role of effector functions in the *** study relevant data demonstrating the different triggers of complement activation after spinal cord injury including direct binding to cellular debris,and or activation via antibody binding to damage-associated molecular *** effector functions of complement have been implicated in spinal cord injury,and we critically evaluate recent studies on the dual role of complement anaphylatoxins in spinal cord injury while emphasizing the lack of pathophysiological understanding of the role of opsonins in spinal cord *** this pathophysiological review,we systematically review the different translational approaches used in preclinical models of spinal cord injury and discuss the challenges for future translation into human *** review emphasizes the need for future studies to dissect the roles of different complement pathways in the pathology of spinal cord injury,to evaluate the phases of involvement of opsonins and anaphylatoxins,and to study the role of complement in white matter degeneration and regeneration using translational strategies to supplement genetic models.

Three Keggin-type heteropolyanions, namely H3Pmo12O40-13H2O, （NH4）3Pmo12O40·4H2O and H3PW12O40·13H2O were prepared and tested in the ring-opening polymerization reaction of tetrahydrofuran. The effects of the counter-cation （H＋, NH4＋） and the peripheral atoms （mo, W） on the polymerization were investigated. It has been found that when the protons of H3Pmo12O40·13H2O were replaced by the ammonium cations the polymerization rate decreased dramatically. Whereas, when the peripheral atoms （mo） were replaced by their homologous （W）, the polymerization rate increased twofold. As for the viscosity average molecular weight （my） of polymer products, it was found that the high molecular weight （7930） was obtained by using H3PW12O40·13H2O. The molecular weight （my） obtained by H3Pmo12O40·13H2O and （NH4）H3Pmo12O40·13H2O was 6470 and 6810, respectively.

Oral Health Status and Gingival Response to Three Different Restorative materials among Saudi Patients: A Clinical &Histopathological Study

Background: The correlation between oral health and dental restoration is fundamental. For the gingival and periodontal tissues to stay healthy, dental restoration should be in regularity with the surrounding tissues. This study aims to assess the oral health status and histopathological gingival response to three different restorative materials among Saudi patients. methods: The study groups consist of 240 patients (50% males and 50% females), aged 18 - 45, with inclusion and exclusion criteria in the study. Participants are divided into three equal groups: those with composite resin restorations, those with amalgam restorations and those with glass ionomer restorations. Biopsies were taken from adjacent gingival tissues. Clinical parameters were determined by: plaque index (PLI), gingival index (GI) and clinical attachment loss (Cal). all data were collected and evaluated by through statistical analysis. Results: The clinical findings of the current study revealed that amalgam restorations produce a higher means of PLI, GI and Cal compared with composite resin restorations and glass ionomer restorations, but not insignificant levels, except Cal (p = 0.004*). As for histopathological findings, there were significant differences in gingival tissue response to amalgam restorations, composite resin restorations and glass ionomer cement fillings, where there were statistically significant differences in numbers of chronic inflammatory cells (p Conclusion: At the end of the present study, we concluded that the amalgam restorations are less biocompatible compared to composite resin restorations and glass ionomer restorations.

In the attempt to reduce surface free energy of silica to improve interaction of silica with silver, silica was doped by different amounts of low surface energy lanthanum, La_2O_3, through impregnation. The doped and undoped silica were used as supports for preparation of Na/Ag/mo/La_2O_3-SiO_2 catalysts. Catalytic performances of the catalysts were evaluated in direct epoxidation of propylene(DPO) using molecular oxygen under atmospheric pressure and without adding hydrogen. Adding 5 wt%La to the Na/Ag/mo/SiO_2 catalyst improves both the catalysts electivity in DPO and its stability over 20h of *** characterization results show that La_2O_3 species interact strongly with silver particles on the silica surface which result in significant improvement in the dispersion profile of silver and marked decrease in the size of silver nanoparticles(AgNPs). The estimated mean diameter of AgNPs is ca. 4.0 nm in Na/Ag/mo/5 wt%La_2O_3-SiO_2, which is smaller than that(53.9 nm) found in Na/Ag/SiO_2. The presence of subnanometer AgNPs on Ag/La_2O_3-SiO_2 prior addition of moO_3 and NaCl to the sample can enhance the mutual electronic synergism between Ag, moO_3 and Na for selective production of propylene oxide.

Rapid sequence induction(RSI) in trauma patients: Insights from healthcare providers

BACKGROUND: We aimed to describe the current practice of emergency physicians and anaesthesiologists in the selection of drugs for rapid-sequence induction(RSI) among trauma ***: A prospective survey audit was conducted based on a self-administered questionnaire among two intubating specialties. The preferred type and dose of hypnotics, opioids, and muscle relaxants used for RSI in trauma patients were sought in the questionnaire. Data were compared for the use of induction agent, opioid use and muscle relaxant among stable and unstable trauma patients by the intubating ***: A total of 102 participants were included; 47 were anaesthetists and 55 were emergency physicians. Propofol(74.5%) and Etomidate(50.0%) were the most frequently used induction agents. Significantly higher proportion of anesthesiologist used Propofol whereas, Etomidate was commonly used by emergency physicians in stable patients(P=0.001). Emergency physicians preferred Etomidate(63.6%) and Ketamine(20.0%) in unstable patients. The two groups were comparable for opioid use for stable patients. In unstable patients, use of opioid differed significantly by intubating specialties. The relation between rocuronium and suxamethonium use did change among the anaesthetists. Emergency physicians used more suxamethonium(55.6% vs. 27.7%, P=0.01) in stable as well as unstable(43.4 % vs. 27.7%, P=0.08) ***: There is variability in the use of drugs for RSI in trauma patients amongst emergency physicians and anaesthesiologists. There is a need to develop an RSI protocol using standardized types and dose of these agents to deliver an effective airway management for trauma patients.

A thermal lens technique is adopted using a single modulated continuous wave （cw） 532-nm laser beam to evaluate the nonlinear refractive index n2, and the thermo-optic coefficient dn/dT, in polymer Poly （1-naphthyl methacrylate）（P-1-Nm） dissolved in chloroform, tetrahydrofuran （THF）, and dimethyl sulfoxide （DmSO） solvents. The results are compared with Z-scan and diffraction ring techniques. The comparison reveals the effectiveness and the simplicity of the TTL modulation technique. The physical origin is discussed for the obtained results.

Pollen morphology of the family Cistaceae in Egypt and its systematic significance

The pollen morphology of 11 species (including two subspecies and two varieties) belonging to two genera (Helianthemum and Fumana) of the family Cistaceae in Egypt was studied using light and scanning electron *** grains of the studied taxa were found to be radially symmetrical and *** size,shape,apertures,and exine ornamentation characteristics were valuable parameters among the studied *** largest pollen size was recorded in *** and the smallest one observed in *** ***- *** shape in the Egyptian taxa varied from (sub-)prolate to prolate spheroidal,but *** is different in having sub-oblate *** pollen data confirm that *** and *** are very closely related *** sculpture was useful in distinguishing between *** *** and *** *** main pollen types of exine ornamentation were recognized: retipilate;reticulate to verrucate;and *** on palynological data,a key for the studied taxa is suggested.
