In vitro inhibitory analysis of consensus siRNAs against NS3 gene of hepatitis C virus 1a genotype

Objective: To explore inhibitory effects of genome-specific, chemically synthesized siRNAs(small interference RNA) against NS3 gene of hepatitis C virus(HCV) 1a genotype in stable Huh-7(human hepatoma) cells as well as against viral replication in serum-inoculated Huh-7 cells. Methods: Stable Huh-7 cells persistently expressing NS3 gene were produced under antibiotic gentamycin(G418) selection. The cell clones resistant to 1 000 μg antibiotic concentration(G418) were picked as stable cell clones. The NS3 gene expression in stable cell clone was confirmed by RT-PCR and Western blotting. siRNA cell cytotoxicity was determined by MTT cell proliferation assay. Stable cell lines were transfected with sequence specific siRNAs and their inhibitory effects were determined by RT-PCR, real-time PCR and Western blotting. The viral replication inhibition by siRNAs in serum inoculated Huh-7 cells was determined by real-time PCR. Results: RT-PCR and Western blot analysis confirmed NS3 gene and protein expression in stable cell lines on day 10, 20 and 30 post transfection. MTT cell proliferation assay revealed that at most concentrated dose tested(50 nmol/L), siRNA had no cytotoxic effects on Huh-7 cells and cell proliferation remained unaffected. As demonstrated by the siRNA time-dependent inhibitory analysis, siRNA NS3-is44 showed maximum inhibition of NS3 gene in stable Huh-7 cell clones at 24(80%, P=0.013) and 48 h(75%, P=0.002) post transfection. The impact of siRNAs on virus replication in serum inoculated Huh-7 cells also demonstrated significant decrease in viral copy number, where siRNA NS3-is44 exhibited 70%(Pal RNA reduction as compared to NS3-is33, which showed a 64%(Pal copy number. siRNA synergism(NS3-is33 + NS3-is44) decreased viral load by 84%(Pal inhibition by each siRNA(i.e., 64%–70%(P<0.05) in serum-inoculated cells. Selecting proper sites for underground dam construction using Multi-Attribute Utility Theory in arid and semi-arid regions

although the construction of underground dams is one of the best methods to conserve water resources in arid and semi-arid regions,applying efficient methods for the selection of suitable sites for subsurface dam construction remains a *** to the costly and time-consuming methods of site selection for underground dam construction,this study aimed to present a new method using geographic information systems techniques and decision-making *** exclusionary criteria including fault,slope,hypsometry,land use,soil,stream,geology,and chemical properties of groundwater were selected for site selection of dam construction and inappropriate regions were omitted by integration and scoring layers in ArcGIS based on the Boolean ***,appropriate sites were prioritized using the Multi-Attribute Utility *** to the results of the utility coefficient,seven sites were selected as the region for underground dam construction based on all criteria and experts’*** site of Nazarabad dam was the best location for underground dam construction with a utility coefficient of 0.7137 followed by sites of Akhavan with a utility coefficient of 0.4633 and Mirshamsi with a utility coefficient of *** study proposed a new approach for the construction of the subsurface dam at the proper site and help managers and decision-makers achieve sustainable water resources with limited facilities and capital and avoid wasting national capital.

The bulk viscous string cosmology in an anisotropic universe with late time acceleration

A model of a cloud formed by massive strings is used as a source of Bianchi type II cases. We assume that the expansion （θ） in the model is proportional to the shear （σ）. To get an exact solution, we consider the equation of state of the fluid to be in the stiff form. It is found that the bulk viscosity played a very important role in the history of the universe. In the presence of bulk viscosity the particles dominate over strings whereas in the absence of it, strings dominate over the particles, which is not consistent with recent observations. also we observe that the viscosity causes the expansion of the universe to be accelerating. Our models are evolving from an early decelerating phase to a late time accelerating phase. The physical and geometrical behaviors of these models are discussed.

Interacting viscous dark energy in a Bianchi type-Ⅲ universe

We study the evolution of the equation of state of viscous dark energy in the scope of Bianchi type III space-time. We consider a case where the dark energy is minimally coupled to the perfect fluid, as well as in direct interaction with it. The viscosity and the interaction between the two fluids are parameterized by constants 40 and σ, respectively. We have made a detailed investigation of the cosmological implications of this parametrization. To differentiate between different dark energy models, we have performed a geometrical diagnostic by using the statefinder pair {s, r}.

Power Efficient Battery Charger by Using Constant Current/Constant Voltage Controller

The Battery Charger Specification presents solution for rechargeable batteries used in portable electronic equipment such as laptop computer systems, cellular telephones and video cameras and the demands for low cost battery chargers are rising these days without give attention for the performance. The goal of the paper was combining high efficiency and versatility with low-cost design and this paper includes the design of the snubber cell, and the components of the current/voltage control charge method. This charger applies a relatively constant current and constant voltage to the battery indefinitely regardless of the AC input rang voltage all the equations described in this paper. The design of prototype converter is verified through an experimental result.

Malignant melanoma is characterized by both genetic and molecular alterations that activate phosphoinositide 3-kinase(PI3K),and RAS/BRAF *** this work,through diversity-based high-throughput virtual screening we identified a lead molecule that selectively targets PI3K and BRAF^(V600E) *** screening,Molecular dynamics simulation and MMPBSA calculations were ***3K and BRAF^(V600E) kinase inhibition was done.A375 and G-361 cells were used for in vitro cellular analysis to determine antiproliferative effects,annexin V binding,nuclear fragmentation and cell cycle *** screening of small molecules indicates compound CB-006-3 selectively targets PI3KCG(gamma subunit),PI3KCD(delta subunit)and BRAF^(V600E).Molecular dynamics simulation and MMPBSA bases binding free energy calculations predict a stable binding of CB-006-3 to the active sites of PI3K and BRAF^(V600E).The compound effectively inhibited PI3KCG,PI3KCD and BRAF^(V600E)kinases with respective IC50 values of 75.80,160.10 and 70.84 ***-006-3 controlled the proliferation of A375 and G-361 cells with GI50 values of 223.3 and 143.6 nM,respectively.A dose dependent increase in apoptotic cell population and sub G0/G1 phase of cell cycle were also observed with the compound treatment in addition to observed nuclear fragmentation in these ***,CB-006-3 inhibited BRAF^(V600E),PI3KCD and PI3KCG in both melanoma ***,based on the computational modeling and in vitro validations,we propose CB-006-3 as a lead candidate for selectively targeting PI3K and mutant BRAF^(V600E) to inhibit melanoma cell *** experimental validations,including pharmacokinetic evaluations in mouse models will identify the druggability of the proposed lead candidate for further development as a therapeutic agent for treating melanoma.
