Expression of Notch pathway components (Numb, Itch, and Siah-1) in colorectal tumors: A clinicopathological study

BACKGROUND The role of the Notch pathway in carcinogenesis and tumor progression has been demonstrated in many organs,including the ***,studies aimed at developing therapies targeting this pathway in various cancers require the identification of several factors that may play a role in regulating Notch-1 *** Numb,Itch,and seven in absentia homolog-1(Siah-1)have been shown to contribute to the regulation of Notch signaling,their role in colorectal carcinogenesis and tumor progression has not been fully elucidated to *** To evaluate Numb,Itch,and Siah-1 expression in colorectal tumors to clarify their relationship with Notch-1 expression and their role in carcinogenesis and tumor *** Expression of Notch-1,Numb,Itch,and Siah-1 was investigated in 50 colorectal carcinomas,30 adenomas,and 20 healthy colonic tissues by immunohistochemistry and quantitative real-time polymerase chain reaction(PCR)*** In contrast to Notch-1,which is expressed at higher levels in tumor tissues and adenomas,expression of Numb,Itch,and Siah-1 was stronger and more frequent in normal mucosa(P<0.01).There was a positive correlation between Notch-1 expression and high histological grade,the presence of lymph node metastasis,and advanced-stage tumors,whereas expression of Numb,Itch,and Siah-1 was absent or reduced in tumors with these clinicopathological parameters(P<0.05).In survival analysis,expression of Notch was related to poor prognosis but that of Numb,Itch,and Siah-1 correlated with improved survival(P<0.05).Multivariate analysis revealed Notch-1 expression and loss of Numb expression to be independent prognostic parameters together with lymph node metastasis(P<0.05).CONCLUSION Our findings support the role of Notch-1 in colorectal carcinoma and indicate that loss of Numb,Itch,and Siah-1 expression is associated with *** data also suggest that these three proteins might be involved in the Notch-1 pathway during colo

Effect of growth hormone on small intestinal homeostasis relation to cellular mediators IGF-I and IGFBP-3

AIM:To evaluate the effects of growth hormone(GH) on the histology of small intestines which might be related to the role of insulin like growth factor(IGF)-I, IGF-binding protein 3(IGFBP-3)and its receptors. METHODS:Twelve week-old adult male Wistar albino rats were divided into two *** study group(n =10),received recombinant human growth hormone (rGH)at a dose of 2 mg/kg per day subcutaneously for 14 d and the control group(n=10)received physiologic *** sections of jejunum were stained with periodic acid shift(PAS)and hematoxylin and eosin(HE) for light *** were also examined for IGF-I, IGFBP-3 and IGF-receptor *** intensity was graded semi-quantitatively using the HS- CORE. RESULTS:Goblet cells and the cells in crypt epitheliawere significantly increased in the study group compared to that of the control *** have demonstrated an increase of IGF-I and IGFBP-3 immunoreactivities in surface epithelium of the small intestine by GH ***-I receptor immunoreactivities of crypt,villous columnar cells,enteroendocrine cells and muscularis mucosae were also more strongly positive in the study group compared to those of in the control group. CONCLUSION:These findings confirm the important trophic and protective role of GH in the homeostasis of the small *** trophic effect is mediated by an increase in IGF-I synthesis in the small intestine, but the protective effect is not related to IGF-I.

Relationship between pinguecula formation and exposure to tandoor ovens in a hospital-based study

AIM: To investigate the relationship between pinguecula and the use of tandoor ***: A total of 539 women, ranging in age from20 to 86 y who attended an outpatient clinic were enrolled. All the patients were asked whether they used tandoor ovens. Women exposed to tandoor ovens(n =286) were accepted as participants in the study group and they were compared with participants in the control group(n =253). The age, presence of pinguecula,duration of exposure to tandoor ovens as years and occupations were recorded for all the ***: Mean duration for exposure to tandoor was20.26y(range 1-62y) in the study group. The rate of pinguecula in the study group was 82.2%(235/286), and the rate in the control group was 37.5%(95/253); this difference was statistically significant(P <0.05).Pinguecula was seen in 61.2%(330/539) of all the ***: Pinguecula is strongly associated with exposure to tandoor ovens.

Premature newborn with intracardiac fungus balls and endocarditis which is curable only with anti-fungal therapy: Case presentation

Invasive fungal infections often caused by Candida species are infections which have high morbidity and mortality rates in newborn infants. 5% of the cases have also concomitant endocarditis and this causes mortality rates over 60%. Thus it has vital importance to diagnose and treat these patients with endocarditis in early stages. Most preferred treatment option is a surgical incision in addition to systemic anti-fungal therapy. Here we presented an extremely low birth weight, premature newborn with endocarditis caused by Candida tropicalis and cured solely by systemic anti-fungal treatment without the need for surgical intervention.

Diagnosis of asymptomatic atrial septal aneurysms using two-dimensional color Doppler and contrast transthoracic echocardiography

Objective To evaluate the dimensions of atrial septal aneurysm (ASA), the presence and characteristics of interatrial shunt, the movement of the wall of the aneurysm, and correlation between these findings and sign and/or symptoms suggesting embolism in Manisa, a district of a western Anatolian city of Turkey. Methods Two thousand five hundred cases were examined by routine transthoracic echocardiography (TTE) in both pediatric and adult cardiology outpatient clinics. ASA was detected in 20 cases and evaluated by two-dimensional color Doppler echocardiography (CDE). The length of the base, the maximum radius and the maximum displacement of ASA were measured. The shunt between the atria was examined by CDE. In cases where a shunt could not be found, galactose and palmitic acid was injected. Standard 12-lead electrocardiogram (ECG) and exercise stress test were also performed. Results No clinical signs or symptoms were found, suggesting a systemic or cerebral embolism. The maximum displacement of ASA was between 2 and 5 mm. All of the aneurysms were localized in the right atrium, and the walls of the aneurysm did not move beyond the base of the left atrium during the maximum displacement. Interatrial shunt was detected in 14 of 20 patients (70%) by CDE and in the remaining six cases by contrast TTE. Frequent ventricular ectopic beats were observed in one patient. Conclusions During routine TTE we observed 0.8% asymptomatic ASA in our population. The use of a contrast agent was found to be a valuable additional method in patients with ASA when the shunt could not be detected by CDE. The risk for embolism is not high when the maximum displacement of the wall of ASA was 5 mm or less and no bulge into the left atrium was observed. Based on our experience with this method, TTE is easy to perform, well-tolerated and acceptable.

Acute rheumatic fever (ARF) is an inflammatory disease that occurs following an infection with certain strains of group A beta-hemolytic streptococci. Despite dramatic falls in the incidence, Acute Rheumatic Fever remains a major cause of morbidity and mortality associated with acquired heart disease in developing countries. Infective endocarditis (IE) is defined as a microbial infection of the endothelial surface of the heart. IE occurs most frequently in patients with chronic rheumatic heart disease. As far as we know, the simultaneous occurrence of both conditions has not been previously reported. References to our outpatient clinic with complaints of fever and joint pain, clinical and laboratory findings suggestive of infective endocarditis fits both with both the ARF and remitted entirely with appropriate treatment, as far as we know, we think that this is not a case of our similar event in the literature. Here we present a case of a fifteen-year-old girl with concurrent acute rheumatic fever and infective endocarditis.

Alveolar Ridge Preservation of an Extraction Socket of Fractured Maxillary Lateral Incisor

Background: Alveolar ridge resorption still continues to be a problem in oral surgery. Cause of bone resorption is including tooth extraction, periodontal disease and inflammatory periapical pathologies. Various methods and materials have been suggested to minimize this resorption. Aim: Goal of this case report is to present alveolar ridge preservation (ARP) following horizontally fractured maxillary lateral incisor with allograft in the aesthetic zone. Case presentation: 30-year-old female patient with fractured tooth was treated by grafting and insertion dental implant. Fractured tooth extraction was performed and extraction socket augmentation was performed by allograft and covered with collagen membrane. Augmented area was treated with bone-level implant. Definitive prosthesis single-tooth porcelain-fused-to-zirconia restorations were fabricated. Conclusions: Before implant insertion, extraction and grafting socket procedure is appropriate treatment for of fractured teeth with granulation tissue.

Correction of Mandibular Prognathism in Combination with Polydiastema Using Rectangular Body Ostectomy: Literature Review and Case Report

Objective: Mandibular prognathism is inadaptability between skull and mandible and is one phenotype of class III malocculsion. Polydiastema may be present together with class III malocclusion. Skeletal class III malocclusion with mandibular prognathism can be diagnosed by cephalometric parameters. Study Design: A 22-year-old man complaining about difficult mastication and speech was referred to Department of Oral and Maxillofacial Surgery. According to the cephalometric analysis of the patient in sagittal plane, maxilla was 3 mm behind and mandible was 1 mm ahead from the skull base. During presurgical orthodontic treatment, polydiastema was treated in maxilla and mandible. Result and Conclusions: Skeletal class III malocclusion requires an orthognathic surgical procedure including some techniques. One of these techniques is mandibular body ostectomy, which is performed often as a surgical procedure for skeletal class III malocclusion.
