Overexpression of inhibinα(1-32)fusion protein promotes apoptosis and cell cycle arrest in a cervical cancer cell model(Hela cells)

Inhibins play important roles in the reproductive system. To evaluate whether inhibin α (1-32) fusion protein plays a role in cervical cancer growth, the plasmid pVAX-inhα was constructed and its effect on proliferation and apoptosis of the human cervical cancer cell line (Hela) was checked by flow cytometry and real-time PCR. The expression and localization of inhibin α protein were detected by RT-PCR and confocal mi- croscopy which showed that inhibin α protein was expressed and localized in the nucleus of Hela cells. Over ex- pression of inhibin α gene significantly induced cell apoptosis and ceased S phase of cell cycle. Furthermore, cell proliferation was significantly suppressed 96 h post-transfection and mRNA level of anti-apoptosis genes (Bcl-2, NFκB) were decreased but pro-apoptosis genes (Bax, wild type p53) and inhibin co receptor (TGFβR3) were increased, indicating that inhibin, through its co-receptor, might activate apoptotic and cell growth cascades which regulate proliferation and apoptosis in Hela cells. These results suggest that inhibin α (1-32) fusion pro- tein, located in the cell nucleus, can regulate Hela cells growth and apoptosis by induction of apoptotic pathways such as NFκB, Bcl-2 and p53 families. These findings may have a significant impact on future research regarding cervical cancer cell lines.

The Sentinel-2 A satellite having embedded advantage of red edge spectral bands offers multispectral imageries with improved spatial,spectral and temporal resolutions as compared to the other contemporary satellites providing medium resolution *** study was aimed at exploring the potential of Sentinel-2 A imagery to estimate Above Ground Biomass(AGB) of Subtropical Pine Forest in Pakistan administered *** developed an AGB predictive model using field inventory and Sentinel 2 A based spectral and textural parameters along with topographic features derived from ALOS Digital Elevation Model(DEM).Field inventory data was collected from 108 randomly distributed plots(0.1 ha each) across the study *** stepwise linear regression method was employed to investigate the potential relationship between field data and corresponding satellite *** and carbon mapping of the study area was carried out through established AGB estimation model with R(o.86),R2(0.74),adjusted R2(0.72) and RMSE value of 33 t/*** results showed that first order textures(mean,standard deviation and variance) significantly contributed in AGB predictive modeling while only one spectral band ratio made contribution from spectral *** study leads to the conclusion that Sentinel-2 A optical data is a potential source for AGB estimation in subtropical pine forest of the area of interest with added benefit of its free of cost availability,higher quality data and long-term continuity that can be utilized for biomass carbon distribution mapping in the resource constraint study area for sustainable forest management.

Synthesis and <i>in Vitro</i>Biological Activities of 4,5-Disubstituted 1,2,4-Triazole-3-Thiols

Purpose: The triazole nucleus is an important part of the therapeutically interesting drug candidate as antimicrobial, analgesic, anticancer, anticonvulsant and anti-inflammatory agents. Methods: Therefore, in this study, twelve 4,5-disubstituted-1,2,4-triazole-3-thiols were synthesized by the reaction of substituted isothiocyanates and hydrazides using the common method of base catalysed intramolecular dehydrative cyclization of substituted thiosemicarbazides 3(a-f) and 4(a-f). The structures of these compounds were characterized by means of FT-IR, 1H-NMR, and elemental analysis data. All these compounds were screened for antibacterial, antioxidant, antitumor and cytotoxic activities. Results: Among these compounds: 5c, 5f and 6f were found active against gram positive cocci, the compounds 5a, 5b, 5d, 6a and 6f showed 85% free radical scavenging effect at 3 ppm when tested for antioxidant activity, 75% tumors inhibition was recorded using 5c, 5d and 6a and brine shrimps lethality assay declared 5a, 5b and 6d was 129.62 μg/ml, 161.577 μg/ml and 81.56 μg/ml respectively. Conclusion: Compounds carrying significant bioactivity can be further studied using animal models to establish their safety profile prior to initiating clinical trials.

Mosquito-borne diseases,particularly dengue and chikungunya have become global threats,infecting millions of people worldwide,including developing countries of Southeast Asia and Latin ***,like many other developing countries,is experiencing frequent dengue *** article,therefore,critically discussed the current status of dengue disease,vector control approaches,and the need for Wolbachia-mediated intervention in Bangladesh and other dengue-endemic developing *** this narrative review study,rel-evant literature was searched from major databases and search engines such as PubMed,BanglaJol,World Health Organization(WHO)/European Centre for Disease Prevention and Control(ECDC)and Google *** the selection criteria,our search strategies finally involved 55 related literature for further *** showed that current vector control strategies could not render protection for an extended period,and the disease burden of arboviruses is *** impoverished outbreak preparedness,urbanization,climate change,and less efficacy of existing control methods have made people susceptible to vector-borne ***,Wolbachia,a naturally occurring endosymbiont of many mosquito species that can potentially limit virus transmission through several host genetic alterations,would be a potential alterna-tive for dengue *** also critically discussed the challenges and prospects of Wolbachia-based den-gue control in developing *** evidence supporting the efficacy and safety of this intervention and its mechanism have also been *** evidence suggests that this introgression method could be an eco-friendly and long-lasting dengue control *** review would help the policymakers and health experts devise a scheme of Wolbachia-based dengue control that can control mosquito-borne diseases,partic-ularly dengue in Bangladesh and other developing countries.
