Expression and reconstitution of the bioluminescent Ca2+reporter aequorin in human embryonic stem cells,and exploration of the presence of functional IP3 and ryanodine receptors during the early stages of their differentiation into cardiomyocytes

In order to develop a novel method of visualizing possible Ca2＋ signaling during the early differentiation of hEsCs into cardi- omyocytes and avoid some of the inherent problems associated with using fluorescent reporters, we expressed the biolumines- cent Ca2＋ reporter, apo-aequorin, in HEs2 cells and then reconstituted active holo-aequorin by incubation withf-coelenterazine. The temporal nature of the Ca2＋ signals generated by the holo-f-aequorin-expressing HEs2 cells during the earliest stages of differentiation into cardiomyocytes was then investigated. Our data show that no endogenous Ca2＋ transients （generated by re- lease from intracellular stores） were detected in 1-12-day-old cardiospheres but transients were generated in cardiospheres following stimulation with KC1 or CaC12, indicating that holo-f-aequorin was functional in these cells. Furthermore, following the addition of exogenous ATP, an inositol trisphosphate receptor （IP3R） agonist, small Ca2＋transients were generated from day 1 onward. That ATP was inducing Ca2＋ release from functional IP3Rs was demonstrated by treatment with 2-APB, a known IP3R antagonist. In contrast, following treatment with caffeine, a ryanodine receptor （RyR） agonist, a minima/Ca2＋ response was observed at day 8 of differentiation only. Thus, our data indicate that unlike RyRs, IP3Rs are present and continually functional at these early stages of cardiomyocyte differentiation.

In order to develop a novel method of visualizing possible Ca signaling during the early differentiation of h EsCs into cardiomyocytes and avoid some of the inherent problems associated with using fluorescent reporters, we expressed the bioluminescent Ca reporter, apoaequorin, in HEs2 cells. Active holo-aequorin was then reconstituted in the apo-aequorinexpressing cells, by incubation with f-coelenterazine, one of a variety of prosthetic co-factors of the holo-aequorin complex. Holo-f-aequorin is a Ca-sensitive bioluminescent protein complex that emits light at 70 nm on binding with Ca and it is 9 times brighter than heterogeneous natural aequorin. Being bioluminescent, aequorin does not require excitation light;it remains in the cells that it is injected into or in the cell compartments where it is targeted to;and it has minimal buffering capacity. The temporal nature of the Ca signals generated by the holo-f-aequorin-expressing HEs2 cells during the earliest stages of differentiation into h EsC-v CMs(i.e., in cardiospheres during the first 12 days) was then investigated using a luminometer. Our data show that no endogenous Ca transients(generated by release from intracellular stores) could be detected in 1-12 day old cardiospheres but that Ca transients were generated in cardiospheres following stimulation with KCl or CaCl, indicating that holof-aequorin was functional in these cells during this developmental time-window. In addition, following the addition of exogenous ATP, an agonist of the inositol trisphosphate receptor(IP3R), Ca transients were generated from day 4 onward. That ATP was inducing Ca release from functional IP3 Rs was demonstrated by treatment with 2-APB, a known IP3 R antagonist. On the other hand, following treatment with caffeine, an agonist of the ryanodine receptor(Ry R), a minimal Ca response was observed at day 8 of differentiation only. Feasibility of achieving net-zero energy performance in high-rise buildings using solar energy

As part of a broad strategy to reach net-zero greenhouse gas emissions and limit global warming,many countries are requiring all new buildings to have net-zero energy *** requires that on-site energy use not exceed on-site generation of renewable energy(taken here to be solar energy),or equivalently,that the building Energy Use Intensity(EUI,kWh/m^(2)a)not exceed the supply of on-site solar energy(electricity and heat)per m^(2)of floor area per *** this basis,we find that achieving net-zero energy performance in an archetype 40-story square building in 16 different cities of North America requires EUI of 17–24 kWh/m^(2)a using PV panels,and 19–28 kWh/m^(2)a using PVT *** building orientation to a non-square floor shape can improve maximum permitted EUI by up to 50%in PV and 60%in PVT ***,the best-performing residential and commercial buildings have EUIs of 50–75 kWh/m^(2)*** if building heights are limited to 5–10 floors does the available solar energy,and thus the permitted EUI,reach 50–75 kWh/m^(2)***,we recommend that policymakers not require high-rise buildings to be net-zero energy,unless they are prepared to limit building heights to 5–10 floors.

An efficient method for unfolding kinetic pressure driven VIsAR data

Velocity Interferometer system for Any Reflector(VIsAR) [Barker and Hollenbach, J. Appl. Phys. 43, 4669(1972)]is a well-known diagnostic that is employed on many shock physics and pulsed-power experiments. With the VIsAR diagnostic, the velocity on the surface of any metal flyer can be found. For most experiments employing VIsAR, either a kinetic pressure [Grady, Mech. Mater. 29, 181(1998)] or a magnetic pressure [Lemke et al., Intl J. Impact Eng. 38,480(2011)] drives the motion of the flyer. Moreover, reliable prediction of the time-dependent pressure is often a critical component to understanding the physics of these experiments. Although VIsAR can provide a precise measurement of a flyer's surface velocity, the real challenge of this diagnostic implementation is using this velocity to unfold the timedependent pressure. The purpose of this paper is to elucidate a new method for quickly and reliably unfolding VIsAR data.

Global sensitivity analysis of a single-cell HBV model for viral dynamics in the liver
