Constitutively Active Soluble Form of Erythropoietin Receptor Suppresses Growth and Angiogenesis of Xenografts of Transfected Cancer Cell Lines

We examined the effect of blocking the erythropoietin (Epo) signaling using an anti-Epo antibody, soluble form of Epo receptor (sEpoR) capable of binding to Epo or EpoR antagonist, and proved to be effective against xenografts of female reproductive organ malignancies and of cancer cell lines in nude mice. We transfected seven cancer cell lines of various origins to express constitutively active sEpoR, and examined their tumorigenesis in nude mice. Suppression of the tumor growth, decrease in viable and proliferating cells and reduction of vascular density were seen individually in all xenografts of transfected cell lines compared with the controls. Quantitative RT-PCR analyses showed that expression levels of Epo, EpoR, ?1A-adrenaline receptor (?1A-ADR) and muscalinic acetylcholine receptor subunit 3 mRNAs (m3-AchR) were higher in the majority of the wild-type xenografts than in the corresponding cell lines except for A549. In some of the transfected xenografts, EpoR, ?1A-ADR and m3-AchR mRNAs were down-regulated. Western blot analyses revealed that the constitutively activated ERK1/2MAPK was discernible in the majority of non-transfected cell lines and was reduced in the transfected cell lines. However, it was regained after exposure to acetylcholine and/or noradrenaline. These findings suggest that constitutively active sEpoR can effectively destroy the xenografts but signals from the autonomic neurotransmitters of the host produced under stress may interfere with this antitumor activity.

AIM To perform automatic gastric cancer risk classificationusing photofluorography for realizing effective mass screening as a preliminary study. METHODS We used data for 2100 subjects including X-ray images, pepsinogen Ⅰ and Ⅱ levels, PGⅠ/PGⅡ ratio, Helicobacter pylori (H. pylori) antibody, H. pylori eradication history and interview sheets. We performed two-stage classification with our system. In the first stage, H. pylori infection status classification was performed, and H. pylori-infected subjects were automatically detected. In the second stage, we performed atrophic level classification to validate the effectiveness of our *** Sensitivity, specificity and Youden index(YI) of H. pylori infection status classification were 0.884, 0.895 and 0.779, respectively, in the first stage. In the second stage, sensitivity, specificity and YI of atrophic level classification for H. pylori-infected subjects were 0.777, 0.824 and 0.601, respectively. CONCLUSION Although further improvements of the system are needed, experimental results indicated the effectiveness of machine learning techniques for estimation of gastric cancer risk.

Fossil woods from the Upper Cretaceous to Paleocene of Heilongjang(Amur)River area of China and Russia

Fossil woods were collected from the Upper Cretaceous to Paleocene beds distributed around Heilong- jang （Amur） River area of China and Russia. A total of 43 specimens were collected from Baishantou, Long- gushan, and Yong＇ancun in Jiayin, China, and 85 specimens from Zeya-Bureya Basin lying southeast of Blagoveshchensk in Russia. Selected specimens were sectioned and studied anatomically. The source deposits of almost all identifiable specimens belong to the Upper Cretaceous （Campanian） Taipinglinchang Formation and the Paleocene Wuyun Formation in China, and the Paleocene Upper Tsagayan Formation in Russia. Al- though the results obtained are still far from complete, the taxonomic composition of the fossil woods show marked stratigraphic differences between the two horizons beyond the K/T boundary in China. There is also a strong similarity in wood flora between the Wuyun Formation of China and the Upper Tsagayan Formation of Russia. All identified specimens from the Taipinglinchang Formation are Xenoxylon latiporosum, This is one of the stratigraphically younger records of Xenoxylon, which is a conifer morphogenus of uncertain affinity most common in the Triassic to Early Cretaceous. Only one specimen with possible derivation from either the Wuyun or the Maastriehtian Furao Formation in China was identified as Taxodioxylon sp. The Wuyun Formation yielded both conifers and dicotyledonous woods. The conifers include Taxodioxylon sequoianum of the Taxodiaceae, and Protopiceoxylon amurense, which is attributable to the extant genera Keteleeria or Nothotsuga of the Pinaeeac, both are at present endemic to China. At least two morphotypes of dicotyledonous woods were recognized, with one identified as el. Hamamelidoxylon , and the other diffuse porous wood is characterized by numerous heterogeneous rays. Effects of Advanced Oxidation Processes on the Decomposition Properties of Organic Compounds with Different Molecular Structures in Water

Studies to decompose persistent organic pollutants in wastewater from chemical factories by using Advanced Oxidation Processes (AOPs) have recently been performed. Oxidation reactions involving ozone and •OH radicals and cleavage caused by UV are the main decomposition reactions that occur in AOPs using ozone and UV. The mechanisms through which organic compounds are decomposed in AOPs are complicated and difficult to understand because various decomposition reactions occur simultaneously. The Total Organic Carbon (TOC) removal efficiencies achieved in several different AOPs were evaluated in this study. The TOC removal efficiencies were different for organic compounds with different chemical structures. The TOC was more effectively removed when aromatic compounds were treated using the O3-UV-TiO2 process than when using the other AOPs, and the TOC was removed more effectively by the O3-UV process than by the UV-TiO2 process. However, the TOC was removed more effectively when open-chain compounds were treated using the UV-TiO2 process than using the O3-UV process, and the UV-TiO2 and O3-UV-TiO2 processes resulted in similar TOC removal efficiencies. Therefore, it is necessary to use the O3-UV-TiO2 process to decompose aromatic compounds as quickly as possible. On the other hand, the UV-TiO2 process degraded the open-chain compounds most effectively, and the O3-UV-TiO2 process did not need to decompose open-chain compounds. Moreover, the TOC of aromatic compounds was removed more slowly than that of open-chain compounds. The TOC removal efficiency increased with decreasing the number of carbon atoms in the molecule. The reconstruction after pylorus preserving pancreatoduodenectomy: pancreatogastrostomy with Roux-en-Y reconstruction versus conventional Traverso type reconstruction

Objective: To compare the result of pancreatogastrostomy (PG) with Roux-en-Y reconstruction versus pancreatojejunostomy (PJ) with conventional Traverso-type reconstruction following pylorus-preserving pancreatectomy (PP- PD) in a retrospective study. PPPD has been accepted as a radical surgical treatment for malignant periampullary neoplasms. However, the reconstruction of this surgery is technically complicated. The leakage of pancreatic juice and the delayed gastric emptying are the major complications of PPPD and may be fatal. To solve these problems, we have performed PG with Roux-en-Y anastomosis. There are several techniques proposed the reconstruction after PPPD, however there have been no previous study describing the result of PG with Roux- en-Y anastomosis. Since 2002 we have perfor- med 32 cases of PPPD. They were divided into two groups according to the reconstruction procedures: PG with Roux-en-Y reconstruction (group PG-RY) (17 cases) and pancreatojejunostomy with Traverso-type reconstruction (group PJ-T) (15 cases). Results: Patient age, gender, and underlying disease were comparable among the groups. Two groups showed no differences in intraoperative bleeding amount, and the time of surgery. The occurrence of the pancreatic leakage was significantly reduced in the group PG-RY compared with the group PJ-T. There was no case of the delayed gastric emptying in the group PGRY. Conclusions: Pancreatogastrosotmy with Roux-en-Y anastomosis can reduce the occurrence of the pancreatic leakage and delayed gastric emptying following pylorus-preserving pancreatoduodenectomy.
