Background: Individualization of high dose regimen of methotrexate (MTX) in pediatric patients with acute lymphoblastic leukemia based on pharmacokinetic (PK) parameters can help in optimization of the dose and better control of the disease. Building up of a pharmacokinetic model can help dose optimization. Objectives: A NONMEM based population (POP) PK model has been subsequently developed to evaluate the effect of demographics as covariates to address variability in pharmacokinetics of MTX. Method: Forty one patients (24 males & 17 females) with ranges of age, body weight and height of 3 - 15 years, 13 - 54 kg and 100 - 177 cm respectively and diagnosed as acute lymphoblastic leukemia (ALL) were involved in the study. MTX was administered as i.v. infusion at a dose of 2 gm/m2 over a period of two hour and its plasma concentrations were determined primarily at 24 hr post-dose to be utilized in the building-up of PK *** initial/prior estimates of volumes of central (V1) and peripheral compartments (V2) and clearance (CL) and inter-compartmental clearance (Q2) for MTX were extracted from literature. The inter-subject variability was estimated for V1 & CL. The influence of different covariates on model performance and parameter estimates was assessed by evaluating the difference in objective function value. The final POP PK model was validated by bootstrap re-sampling procedures (1000 runs) and the 95% confidence intervals for the estimates were calculated. Results: The POP estimates for CL, V1 and V2 were 2.18 L/h, 5.74 L and 11.2 L respectively. The inter-individual variability for the CL and V1 was 23% and 30% respectively. The covariates analysis showed effect of body surface area and sex on the CL and weight on V1. Conclusions: The POP-PK model developed adequately defines the population PK of MTX in pediatric patients with lymphoblastic leukemia. AIM:To investigate the safety and effectiveness of endoscopic therapy with a paclitaxel-eluting balloon(PEB) for biliary anastomotic stricture(AS) after liver transplantation(LT).METHODS:This prospective pilot study enrolled 13 consecutive eligible patients treated for symptomatic AS after LT at the University Hospital of Münster between January 2011 and March *** patients were treated by endoscopic therapy with a PEB and followed up every 8 wk by endoscopic retrograde cholangiopancreatography(ERCP).In cases of re-stenosis,further balloon dilation with a PEB was ***-up was continued until 24 mo after the last ***:Initial technical feasibility,defined as successful balloon dilation with a PEB during the initial ERCP procedure,was achieved in 100% of ***-term clinical success(LTCS),defined as no need for further endoscopic intervention for at least 24 mo,was achieved in 12 of the 13 patients(92.3%).The mean number of endoscopic interventions required to achieve LTCS was only 1.7 ± *** failure,defined as the need for definitive alternative treatment,occurred in only one patient,who developed recurrent stenosis with increasing bile duct dilatation that required stent ***:Endoscopic therapy with a PEB is very effective for the treatment of AS after LT,and seems to significantly shorten the overall duration of endoscopic treatment by reducing the number of interventions needed to achieve LTCS.

采用柱层析、制备薄层层析等方法从海洋链霉菌S007的发酵液提取物中分离得到了9个化合物,经波谱分析确定为嘧啶(1),吡咯-2-羧酸(2),卡拉霉素(3),3-吲哚丙酸(4),吲哚-3-羧酸(5),N-乙酰基酪胺(6),2'-胸腺嘧啶脱氧核苷(7),N-β-乙酰色胺酸(8)和三乙胺盐酸盐(9)。其中三乙胺盐酸盐是首次从链霉菌中得到;海虾致死实验结果显示:化合物3在40μg/mL浓度下对丰年虾的致死率为86.5%,表明卡拉霉素对丰年虾表现出很强的毒性作用。

Investigation on Lanthanum Fluoride as a Novel Cathode Buffer Material Layer for the Enhancement of Stability and Performance of Organic Solar Cell

This article presents the investigation on very thin Lanthanum Fluoride (LaF3) layer as a new cathode buffer layer (CBL) for organic solar cell (OSC). OSCs were fabricated with poly(3-hexylthiophene) (P3HT) and phenyl-C61-butyric acid methyl ester (PCBM) polymer blend at 1:1 ratio. Electron-beam evaporation at room temperature was used to deposit 3 and 5 nm thick LaF3 layer. A very smooth surface of LaF3 with an average roughness of 0.2 nm has been observed by the Atomic Force Microscope (AFM) that is expected to prevent diffusion of cathode metal ion through it and thereby enhance the lifetime and stability of OSC. Huge enhancement of JSC and VOC was also observed for 3 nm-thick LaF3 CBL. Several excellent features of the LaF3 layer such as, transporting electron through tunneling, blocking of holes to the cathode, minimizing recombination, protecting the photoactive polymer from ambient oxygen, and reducing degradation/oxidation of any low work function layer at the cathode interface, might have contributed to the performance enhancement of OSC. The experimental findings indicate the promise of LaF3 to be an excellent CBL material for OSC.
