BACKGROUND The challenges for inflammatory bowel disease(IBD)diagnostics are to discriminate it from gut conditions with similar symptoms such as irritable bowel syndrome(IBS),to distinguish IBD subtypes,to predict disease progression,and to establish the risk to develop colorectal cancer(CRC).Alterations in gut microbiota have been proposed as a source of information to assist in IBD *** prausnitzii(***),its phylogroups,and Escherichia coli(***)have been reported as potential biomarkers,but their performance in challenging IBD diagnostic situations remains *** hypothesize that bacterial biomarkers based in these species may help to discriminate these conditions of complex *** To evaluate the usefulness of indices calculated from the quantification of these species as biomarkers to aid in IBD *** A retrospective study of 131 subj.cts(31 controls(H);45 Crohn’s disease(CD),25 ulcerative colitis(UC),10 IBS,and 20 CRC patients)was performed to assess the usefulness of bacterial biomarkers in ***,the performance of biomarkers in faeces was studied in 29 stool samples(19 CD,10 UC).Relative abundances of total ***(FP),its phylogroups(PHGI and PHGII),and ***(E)quantification were determined by *** were combined to calculate the FP-E index,the PHGI–E index and the PHGII-E *** accuracy to discriminate among conditions was measured by the area under the receiver operating characteristic curve(AUC).RESULTS In biopsies,FP-E index was good for discriminating IBS from CD(AUC=0.752)while PHGII-E index was suitable for discriminating IBS from UC(AUC=0.632).The FP-E index would be the choice to discriminate IBD from CRC,especially from all UC subtypes(AUC≥0.875),regardless of the activity status of the *** between UC patients that had the longest disease duration and those with CRC featured slightly lower AUC *** differentiation in IB

Headache is one of the commonest complaints that doctors need to address in clinical *** genetic mechanisms of different types of headache are not well understood while it has been suggested that self-reported headache and self-reported migraine were genetically *** this study,we performed a meta-analysis of genome-wide association studies(GWAS)on the self-reported headache phenotype from the UK Biobank and the self-reported migraine phenotype from the 23andMe using the Unified Score-based Association Test(metaUSAT)software for genetically correlated phenotypes(N=397,385).We identified 38 loci for headaches,of which 34 loci have been reported before and four loci were newly *** LDL receptor related protein 1(LRP1)-Signal Transducer and Activator of Transcription 6(STAT6)-Short chain Dehydrogenase/Reductase family 9C member 7(SDR9C7)region in chromosome 12 was the most significantly associated locus with a leading p value of 1.24×10^(-62)of *** One Cut homeobox 2(ONECUT2)gene locus in chromosome 18 was the strongest signal among the four new loci with a p value of 1.29×10^(-9)of *** study demonstrated that the genetically correlated phenotypes of self-reported headache and self-reported migraine can be meta-analysed together in theory and in practice to boost study power to identify more variants for *** study has paved way for a large GWAS meta-analysis involving cohorts of different while genetically correlated headache phenotypes.

Obj.ctives: Recent findings have further highlighted the role of the thyroid system in the pathophysiology of depression and revealed new physiologically relevant elements of the thyroid system. Our previous study showed an antidepressant-like effect of 3,5-diiodo-L-thyronine(T2), which was previously considered to be a physiologically inactive molecule, in mice. Here, we aimed to investigate the antidepressant-like effects of T2 further. Methods: We studied the effects of bolus inj.ctions of T2 to C57Bl6 j.mice at doses of 0.25 or 0.75 mg/kg with the tail suspension and forced swim models. The effects of the higher dose were investigated in CD1 mice in the forced swim test. Potential behavioral effects of these treatments were also studied using the novel cage and dark-light box ***: A reduction of depressive-like behavior was found in mice treated with 0.75 mg/kg of T2 in the tail suspension test, but not in the forced swim test. Locomotion and anxiety variables were unaltered following treatment with T2. There were no significant changes after bolus administration of 0.25 mg/kg T2 in either test for depressive-like behavior. Thus, bolus inj.ction of T2 at the dose 0.75 mg/kg can induce antidepressant-like effects without affecting other behaviors. Conclusions: A discrepant result in the forced swim test may be due to its different sensitivity to T2 compared with the tail suspension paradigm. Furthermore, the development of procedural modifications of this model can be useful in its application in pre-clinical studies.

AIM: To ascertain fine needle aspiration(FNA) tech-niques by endosonographers with varying levels of ex-perience and ***: A survey study was performed on United States based endosonographers. The subj.cts complet-ed an anonymous online electronic survey. The main outcome measurements were differences in needle choice, FNA technique, and clinical decision making among endosonographers and how this relates to years in practice, volume of EUS-FNA procedures, and prac-tice ***: A total of 210(30.8%) endosonographers completed the survey. j.st over half(51.4%) identified themselves as academic/university-based practitioners. The vast maj.rity of respondents(77.1%) identified themselves as high-volume endoscopic ultrasound(EUS)(> 150 EUS/year) and high-volume FNA(> 75 FNA/year) performers(73.3). If final cytology is non-diagnostic, high-volume EUS physicians were more likely than low volume physicians to repeat FNA with a core needle(60.5% vs 31.2%; P = 0.0004), and low volume physicians were more likely to refer patients for either surgical or percutaneous biopsy,(33.4% vs 4.9%, P < 0.0001). Academic physicians were more likely to repeat FNA with a core needle(66.7%) compared to community physicians(40.2%, P < 0.001). CONCLUSION: There is significant variation in EUS-FNA practices among United States endosonographers. Differences appear to be related to EUS volume and practice environment.
