Intrahepatic Cd8^+ lymphocyte trapping during tolerance induction using mushroom derived formulations:A possible role for liver in tolerance induction

AIM: To determine the immunomodulatory effect of Shiitake (a mushroom extract),we tested its effect on liver-mediated immune regulation in a model of immune-mediated colitis. METHOdS: Four groups of mice were studied. Colitis was induced by intracolonic instillation of TNBS in groups A and B. Groups A and C were treated daily with Shiitake extract,while groups B and d received bovine serum albumin. Mice were evaluated for development of macroscopic and microscopic. The immune effects of Shiitake were determined by FACS analysis of intra-hepatic and intrasplenic lymphocytes and IFN-γ ELISPOT assay. RESULTS: Administration of Shiitake resulted in marked alleviation of colitis,manifested by signifi cant improvement in the macroscopic and microscopic scores,and by reduction in IFN-γ-producing colonies in group A,compared to group B mice (1.5 pfu/mL vs 3.7 pfu/mL,respectively). This beneficial effect was associated with a significant increase in the intra-hepatic Cd8+ lymphocyte trapping,demonstratedby an increased intrasplenic/intrahepatic Cd4/Cd8 lymphocyte ratio. These effects were accompanied by a 17% increase in the number of intrahepatic natural killer T (NKT) cells. A similar effect was observed when Shiitake was administered to animals without disease induction. CONCLUSION: Shiitake extract affected liver-mediated immune regulation by altering the NKT lymphocyte distribution and increasing intrahepatic Cd8+ T lymphocyte trapping,thereby leading to alleviation of immune-mediated colitis.

Considerable progress has been made in advanced imaging techniques,such as micro CT（μCT） and micro magnetic resonance imaging（μMRI）,for noninvasive and/or nondestructive assessment of trabeculae in OVX induced osteopenia and in various treatments in rats in vivo and in *** a small highefficiency coil in high-field imager,uMRI can be performed at resolutions sufficient to discriminate individual trabeculae,and is capable of demonstrating differences in rat trabeculae following OVX that are not detected by *** they are equipment and technique demanding,the advanced imaging methods are able to measure 3d structure and connectivity in arbitrary orientations in a highly automated, objective,non-user-specific *** can have a large sample size and therefore less sampling error. They are non-destructive which allows multiple tests on the same *** most important aim of any therapy for osteoporosis is to improve biomechanical competence,as an increase in quantity of bone mineral is not necessarily followed by an increase in bone quality,such as treatment of low dose long-term sodium fluoride in OVX *** of the long bones and compression of the vertebral body are among commonly used biomechanical testing methods.

Objective:To identify bacteria isolated from diseased Southern flounder and determine whether they are virulent to channel catfish and Nile ***:Gram-negative bacteria isolates were recovered from five tissues of diseased Southern flounder(Paralichthys lethostigma).The isolates were subjected to biochemical and molecular identification followed by virulence study in ***:Based on biochemical analysis,the 25 isolates were found to share homologies with either Edwardsiella tarda(***)or Aeromonas hydrophila(***).Based on sequencing results of partial 16S rRNA gene,15 isolates shared 100%identities with the 16S rRNA sequence of previously identified *** strain TX1,whereas the other 10 isolates shared 100%identities with the 16S rRNA sequence of previously identified *** strain *** healthy fish were exposed to flounder isolate by intracoelomic injection,the Ld50 values of flounder isolate *** to channel catfish or Nile tilapia[(10±2)g]were 6.1×10^(4)and 1.1×10^(7)CFU/fish,respectively,whereas that of flounder isolate *** to channel catfish and Nile tilapia were 1.4×10^(7)and 5.6×10^(7)CFU/fish,***:This is the first report that *** and *** isolated from diseased Southern flounder are virulent to catfish and tilapia.
