AIM To conduct a prospective assessment of anti-hepatitis E virus(HEV) Ig G seroprevalence in the Western Cape Province of South Africa in conjunction with evaluating risk factors for *** Consenting participants attending clinics and wards of Groote Schuur, Red Cross Children's Hospital and their affiliated teaching hospitals in Cape Town, South Africa, were sampled. Healthy adults attending blood donor clinics were also recruited. Patients with known liver disease were excluded and all major ethnic/race groups were included to broadly represent local demographics. Relevant demographic data was captured at the time of sampling using an interviewer-administered confidential questionnaire. Human immunodeficiency virus(HIV) status was self-disclosed. HEV Ig G testing was performed using the Wantai? *** HEV is endemic in the region with a seroprevalence of 27.9%(n = 324/1161) 95%CI: 25.3%-30.5%(21.9% when age-adjusted) with no significant differences between ethnic groups or HIV status. Seroprevalence in children is low but rapidly increases in early adulthood. With univariate analysis, age ≥ 30 years old, pork and bacon/ham consumption suggested risk. In the multivariate analysis, the highest risk factor for HEV Ig G seropositivity(OR = 7.679, 95%CI: 5.38-10.96, P der followed by pork consumption(OR = 2.052, 95%CI: 1.39-3.03, P demonstrates that HEV genotype 3 may be currently circulating in the Western *** Hepatitis E seroprevalence was considerably higher than previously thought suggesting that hepatitis E warrants consideration in any patient presenting with an unexplained hepatitis in the Western Cape, irrespective of travel history, age or ethnicity.

背景:骨质疏松涉及破骨细胞分化成熟和成骨类细胞凋亡两方面,肿瘤坏死因子α能诱导多种细胞凋亡且效率差异很大,骨保护素能抑制破骨细胞成熟,但对成骨类细胞凋亡的作用未见报道。目的:探讨肿瘤坏死因子α能否诱导成骨类细胞凋亡及骨保护素能否抑制其凋亡。设计:以成骨细胞和MG63细胞作为实验对象、以鼠WEHI164细胞系用于阳性对照的观察对比分析。单位:解放军总医院全军口腔医学研究所、美国匹茨堡医学院退伍军人医疗中心。材料:本实验于2001-01/2003-12在***实验室和口腔研究所实验室完成。选择商品化细胞系,RPMI1640培养液和各种相关蛋白用于实验。方法:用人间充质干细胞在分化培养液中培养21d使之成为成骨细胞,与MG63一起用于凋亡实验。AnnexinV和原位末端标记法检测肿瘤坏死因子α诱导成骨类细胞凋亡。主要观察指标:全部细胞数及凋亡细胞数。结果:和FasL诱导细胞凋亡一样,肿瘤坏死因子α能引起MG63骨肉瘤细胞,间充质干细胞和成骨细胞的凋亡,并表现为明显的浓度依赖和时间依赖。低浓度肿瘤坏死因子α(170～500pmol/L)作用于细胞2～4h就显示了较明显的凋亡,而骨保护素在0.45～1.5nmol/L浓度时,几乎完全抑制了500pmol/L肿瘤坏死因子α诱导的凋亡。成骨细胞分泌骨保护素,而破骨细胞及破骨前体细胞产生肿瘤坏死因子α,它们相互作用降低成骨细胞的凋亡。结论:肿瘤坏死因子α能诱导成骨类细胞凋亡,骨保护素能够抑制肿瘤坏死因子α的凋亡诱导作用,从抑制破骨细胞成熟和成骨细胞凋亡两方面表现骨保护素防治骨质疏松的作用机制。

果实风味品质备受关注,然而近年来消费者常常抱怨"越来越没有以前的味儿了"。以产量和抗性为目标的现代育种策略是果实风味品质降低的重要原因之一。另外,过度追求采后货架寿命为目标的采后贮藏物流技术进一步导致了果实风味品质的下降。芳香物质是风味品质的重要组成,对于采后低温贮藏敏感。采收后置于4℃低温贮藏7 d然后转货架1-3 d,贮藏后番茄的风味与新鲜采收果实相差甚远。桃果实的研究进一步表明,芳香物质含量减少是不适宜低温贮藏后果实风味品质下降的主要原因。同时,采收成熟度也是导致贮藏后果实芳香等风味品质下降的重要因子。结合代谢组学、转录组学和表观组学等技术,发现参与芳香物质合成的结构基因以及转录因子受到果实采后贮藏物流措施影响,dNA甲基化与上述基因表达具有相关性。研究从采后生物学角度阐述了果实芳香品质的调控机制,为进一步完善贮藏物流技术研发提供了依据。

BACKGROUNd Acute gastrointestinal(GI)graft-vs-host disease(aGVHd)is the most complication of hematopoietic stem cell transplant(HSCT)in patients with hematologic *** data exists on endoscopic evaluation of GVHd in post-HSCT patients with differing GI ***,the diagnostic value of gross endoscopic findings as well as the safety of endoscopy in this commonly thrombocytopenic and neutropenic patient population remains *** To understand the diagnostic value of symptoms and gross endoscopic findings as well as safety of endoscopy in aGVHd *** We analyzed 195 endoscopies performed at City of Hope in patients who underwent allogeneic HSCT less than 100 d prior for hematologic malignancy and were subsequently evaluated for aGVHd via *** yield,sensitivity,and specificity of diagnosing aGVHd were calculated for upper and lower endoscopy,various GI tract locations,and presenting *** Combined esophagogastroduodenoscopy(EGd)and flexible sigmoidoscopy(FS)demonstrated a greater diagnostic yield for aGVHd(83.1%)compared to EGd(66.7%)or FS(77.2%)alone with any presenting *** upper and lower GI tract demonstrated similar yields regardless of whether patients presented with diarrhea(95.7%vs 99.1%)or nausea/vomiting(97.5%vs 96.8%).Normalappearing mucosa was generally as specific(91.3%)as abnormal mucosa(58.7%-97.8%)for the presence of *** events such as bleeding(1.0%),infection(1.0%),and perforation(0.5%)only occurred in a small proportion of patients,with no significant differences in those with underlying thrombocytopenia(P=1.000)and neutropenia(P=0.425).CONCLUSION Combined EGd and FS with biopsies of normal and inflamed mucosa demonstrated the greatest diagnostic yield regardless of presenting symptom and appears to be safe in this population of patients.

雪貂是一种极适合流感病毒研究的理想动物。它感染流感病毒后,发病过程和机体反应与人体相似,表现为非致死性感染。主要受感染的部位为上呼吸道,部分动物可有轻度病毒血症和肺部侵袭,但损害很轻。长期以来,我们利用这一动物模型对流感病毒的致病能力,强、弱毒株的毒力差别,以及机体反应等方面进行了研究,以期阐明人类流感病毒的致病机理。我们的研究选用了PR/8、64d二个弱毒株,和7a、64c二个强毒株,从EBId50（50%批量感染量）、上呼吸道感染、发热、下呼吸道感染等四个方面进行探讨。

Objective: To define the prevalence of auto antibodies (Ab) to different organs in young patients with type 1 diabetes (type 1dM). Methods: Ab to thyroid, celiac and adrenal disease was analyzed in 831 type 1 dM patients. Results: Hundred twenty-three (14.8%) had positive thyroid Ab. The risk of developing thyroid Ab was increased in girls (HR 2.3;95% CI 1.6 - 3.2, p d positive endomysium Ab (3.1% in girls and 3.5% in boys, p = NS). Adrenal Ab was detected in 5 patients. The dQA1*0301-dQB1*0301 haplotype was more prevalent in patients with thyroid Ab (p = 0.0281);dQA1*301-dQB1*302 and dQA1*501-dQB1*201 in patients with endo- mysium Ab (p = 0.0251 and p drenal Ab were dQA1*301-dQB1*302 positive. Conclusions: Type 1d patients should be screened annually for thyroid autoimmunity and celiac disease. The dQA1*0301-dQB1*0301 haplotype seems to confer susceptibility to thyroid autoimmunity, dQA1*301-dQB1*302 and dQA1*501-dQB1*201 to celiac disease and dQA1*301-dQB1*302 to adrenal autoimmunity.
