RPLP0/TbP are the most stable reference genes for human dental pulp stem cells under osteogenic differentiation

bACKGROUND Validation of the reference gene(RG)stability during experimental analyses is essential for correct quantitative real-time polymerase chain reaction(RT-qPCR)data ***,in an unreliable way,several studies use genes involved in essential cellular functions[glyceraldehyde-3-phosphate dehydro-genase(GAPDH),18S rRNA,andβ-actin]without paying attention to whether they are suitable for such experimental conditions or the reason for choosing such ***,such studies use only one gene when Minimum Information for Publication of Quantitative Real-Time PCR Experiments guidelines recom-mend two or more *** impacts the credibility of these studies and causes dis-tortions in the gene expression *** tissue engineering,the accuracy of gene expression drives the best experimental or therapeutical *** cultivated DPSCs under two conditions:Undifferentiated and osteogenic dif-ferentiation,both for 35 *** evaluated the gene expression of 10 candidates for RGs[ribosomal protein,large,P0(RPLP0),TATA-binding protein(TbP),GAPDH,actin beta(ACTb),tubulin(TUb),aminolevulinic acid synthase 1(ALAS1),tyro-sine 3-monooxygenase/tryptophan 5-monooxygenase activation protein,zeta(YWHAZ),eukaryotic translational elongation factor 1 alpha(EF1a),succinate dehydrogenase complex,subunit A,flavoprotein(SDHA),and beta-2-micro-globulin(b2M)]every 7 d(1,7,14,21,28,and 35 d)by *** data were analysed by the four main algorithms,ΔCt method,geNorm,NormFinder,and bestKeeper and ranked by the RefFinder *** subdivided the samples into eight *** All of the data sets from clonogenic and osteogenic samples were analysed using the RefFinder *** final ranking showed RPLP0/TbP as the two most stable RGs and TUb/b2M as the two least stable *** theΔCt method or NormFinder analysis showed TbP/RPLP0 as the two most stable ***,geNorm analysis showed RPLP0/EF1αin the first *** algorithms’two least stab

Anti-ganglioside antibodies are associated with delayed/poor clinical recovery in Guillain-barrèsyndrome,mostly related to halted axon ***-linking of cell surface gangliosides by anti-ganglioside antibodies triggers inhibition of nerve repair in in vitro and in vivo paradigms of axon *** effects involve the activation of the small GTPase Rho A/ROCK signaling pathways,which negatively modulate growth cone cytoskeleton,similarly to well stablished inhibitors of axon regeneration described so *** aim of this work was to perform a proof of concept study to demonstrate the effectiveness of Y-27632,a selective pharmacological inhibitor of ROCK,in a mouse model of axon regeneration of peripheral nerves,where the passive immunization with a monoclonal antibody targeting gangliosides GD1a and GT1b was previously reported to exert a potent inhibitory effect on regeneration of both myelinated and unmyelinated *** results demonstrate a differential sensitivity of myelinated and unmyelinated axons to the pro-regenerative effect of *** with a total dosage of 9 mg/kg of Y-27632 resulted in a complete prevention of anti-GD1a/GT1b monoclonal antibody-mediated inhibition of axon regeneration of unmyelinated fibers to skin and the functional recovery of mechanical cutaneous *** contrast,the same dose showed toxic effects on the regeneration of myelinated ***,scale down of the dosage of Y-27632 to 5 mg/kg resulted in a significant although not complete recovery of regenerated myelinated axons exposed to anti-GD1a/GT1b monoclonal antibody in the absence of toxicity in animals exposed to only ***,these findings confirm the in vivo participation of Rho A/ROCK signaling pathways in the molecular mechanisms associated with the inhibition of axon regeneration induced by anti-GD1a/GT1b monoclonal *** findings open the possibility of therapeutic pharmacological intervention targeting Rho A/R

Chinese urban residential buildings face serious indoor air quality *** purification systems(e.g.,air purifiers,fresh air systems)that supply fresh air or purify indoor air are widely used in residential buildings to improve indoor air *** the challenges of energy conservation and emission reduction,it is urgent to guide residents on choosing appropriate air purification *** key problem is how to identify the best air purification system with the lowest energy consumption or operating *** of air purification systems have rarely been conducted on the premise that they should deliver air such that the main indoor pollutant concentrations all meet air quality *** ignored in comparisons are the increased filtration resistance caused by dust accumulation on the filter and the cost of filter *** this study,we propose a method to compare residential air purification systems based on energy consumption and total cost,and we provide methods to select air purification systems in China's different climatic *** used a standard two-person bedroom as the example,and determined the most suitable(minimum cost or energy consumption)air purification systems for the capital cities of the provinces in Chinese mainland.

Astrophysical S-factor and reaction rate for 15N(p,γ)16O within the modified potential cluster model

We study radiative p^(15)N capture on the ground state of ^(16)O at stellar energies within the framework of a modified potential cluster model(MPCM)with forbidden states,including low-lying *** investigation of the ^(15)N(p,γ0)^(16)O reaction includes the consideration of ^(3)S_(1) resonances due to E1 transitions and the contribution of the ^(3)P_(1) scattering wave in the p+^(15)N channel due to the ^(3)P_(1)→^(3)P_(0)M1 *** calculated the astrophysical low-energy S-factor,and the extrapolated S(0)turned out to be within 34.7−40.4 keV·*** important role of the asymptotic constant(AC)for the ^(15)N(p,γ0)16O process with interfering ^(3)S_(1)(312)and ^(3)S_(1)(962)resonances is elucidated.A comparison of our calculation for the S-factor with existing experimental and theoretical data is addressed,and a reasonable agreement is *** reaction rate is calculated and compared with the existing *** has negligible dependence on the variation of AC but shows a strong impact of the interference of ^(3)S_(1)(312)and ^(3)S_(1)(962)resonances in reference to the CNO Gamow windows,especially at low *** estimate the contribution of cascade transitions to the reaction rate based on the exclusive experimental data from ***.C.85,065810(2012).The reaction rate enhancement due to the cascade transitions is observed from T_(9)>0.3 and reaches the maximum factor~1.3 at T_(9)=*** present the Gamow energy window and a comparison of rates for radiative proton capture reactions ^(12)N(p,γ)^(13)O,^(13)N(p,γ)^(14)O,^(14)N(p,γ)^(15)O,and ^(15)N(p,γ)^(16)O obtained in the framework of the MPCM and provide the temperature windows,prevalence,and significance of each process.

The rate of deadwood decomposition processes in tree stand gaps resulting from bark beetle infestation spots in mountain forests

Decaying wood is an essential element of forest ecosystems and it affects its other *** aim of our research was to determine the decomposition rate of deadwood in various humidity and thermal conditions in the gaps formed in the montane forest *** research was carried out in the babiog orski National *** research plots were marked out in the gaps of the stands,which were formed as a result of bark beetle *** plots were located in undisturbed *** research covered wood of two species–spruce and beech in the form of cubes with dimensions of 50 mm×50 mm×22 *** samples were placed directly on the soil surface and subjected to laboratory analysis after 36 months.A significant influence of the wood species and the study plot type on the physicochemical properties of the tested wood samples was *** characteristics strongly correlated with soil moisture.A significantly higher mass decline of wood samples was recorded on the reference study plots,which were characterized by more stable moisture *** decomposition of wood in the gaps regardless of the species is related to lower *** wood species covered by the study differed in the decomposition *** wood samples were characterized by a significantly higher decomposition rate compared to beech wood *** research has confirmed that disturbances that lead to the formation of gaps have a direct impact on the decomposition process of deadwood.

Compressive property and energy absorption characteristic of interconnected porous Mg-Zn-Y alloys with adjusting Y addition

In this study,interconnected porous Mg-2Zn-xY alloys with different phase compositions were prepared by various Y additions(x=0.4,3,and 6 wt.%)to adjust the compressive properties and energy absorption *** characterization methods were then applied to identify the microstructure of the porous Mg-Zn-Y and describe the details of the second *** tests were performed at room temperature(RT),200℃,and 300℃to study the impact of the Y addition and testing temperature on the compressive properties of the porous *** experimental results showed that a high Y content promotes a microstructure refinement and increases the volume fraction of the second *** the Y content increases,different Mg-Zn-Y ternary phases appear:I-phase(Mg_(3)Zn_(6)Y),W-phase(Mg_(3)Zn_(3)Y_(2)),and LPSO phase(Mg_(12)ZnY).When the Y content ranges between 0.4%and 6%,the compressive strength increases from 6.30MPa to 9.23 MPa,and the energy absorption capacity increases from 7.33 MJ/m^(3)to 10.97 MJ/m^(3)at RT,which is mainly attributed to the phase composition and volume fraction of the second ***,the average energy absorption efficiency is independent of the Y *** addition,the compressive deformation behaviors of the porous Mg-Zn-Y are altered by the testing *** compressive strength and energy absorption capacity of the porous Mg-Zn-Y decrease due to the softening effect of the high temperature on the *** deformation behaviors at different temperatures are finally observed to reflect the failure mechanisms of the struts.

Probing the bardeen-Kiselev black hole with the cosmological constant caused by Einstein equations coupled with nonlinear electrodynamics using quasinormal modes and greybody bounds

In this work,we investigate a static and spherically symmetric bardeen-Kiselev black hole(bH)with the cosmological constant,which is a solution of the Einstein-non-linear Maxwell field *** compute the quasinormal frequencies for the bardeen-Kiselev bH with the cosmological constant due to electromagnetic and gravitational *** varying the bH parameters,we discuss the behavior of both real and imaginary parts of the bH quasinormal frequencies and compare these frequencies with the Reissner-Nordström-de Sitter bH surrounded by quintessence(RN-dSQ).Interestingly,it is shown that the responses of the bardeen-Kiselev bH with the cosmological constant and the RN-dSQ under electromagnetic perturbations are different when the charge parameter q,the state parameter w and the normalization factor c are varied;however,for the gravitational perturbations,the responses of the bardeen-Kiselev bH with the cosmological constant and the RN-dSQ are different only when the charge parameter q is ***,compared with the gravitational perturbations,the electromagnetic perturbations can be used to understand nonlinear and linear electromagnetic fields in curved spacetime *** interesting observation is that,due to the presence of Kiselev quintessence,the electromagnetic perturbations around the bardeen-Kiselev bH with the cosmological constant damps faster and oscillates slowly;for the gravitational perturbations,the quasinormal mode decays slowly and oscillates *** also study the reflection and transmission coefficients along with the absorption cross section in the bardeen-Kiselev bH with the cosmological constant;it is shown that the transmission coefficients will increase due to the presence of Kiselev quintessence.

Radiative capture of proton through the ^(14)N(p,γ)^(15)O reaction at low energy

The CNO cycle is the main source of energy in stars more massive than our *** process defines the energy production,the duration of which can be used to determine the lifetime of massive *** cycle is an important tool for determining the age of globular *** proton capture via p+^(14)N→^(15)O+γ,at energies of astrophysical interest,is an important process in the CNO *** this project,we apply a potential model to describe both non-resonant and resonant reactions in the channels where radiative capture occurs through electric E1 *** employed the R-matrix method to describe the ongoing reactions via M1 resonant transitions,when it was not possible to correctly reproduce the experimental data using the potential *** partial components of the astrophysical S-factor are calculated for all possible electric and magnetic dipole transitions in ^(15)*** linear extrapolated S-factor at zero energy(S(0))agrees well with earlier reported values for all transition types considered in this *** on the value of the total astrophysical S-factor,depending on the collision energy,we calculate the nuclear reaction rates for p+^(14)N→^(15)O+γ.The computed rates agree well with the results reported in the NACRE II Collaboration and most recent existing measurements.
