The Natural Killer Cell: A Historical Perspective and the Use of Supplements to Enhance NKC Activity

The Natural Killer Cell (NKC) is the cell-mediated cornerstone of innate immunity. The purpose of this reviewis to give a historical perspective of the discovery of the Natural Killer Cell (NKC)and to apply the use of supplements in the enhancement of NKC in human cancers for the developmentof human health and *** the discovery of the NKC, as observed by Nomarski optics, scanning (SEM)/transmission electron microscopy (TEM) with cellular numeration and enrichment using bovine serum albumin (BSA) continuous gradients, there have been significant research and clinical studies to increase the effectiveness of NKC in the destruction of cancer cells. Based on significant research and clinical studies, at least 16 components have been identified that enhance or may enhance, based on their immune modulator activity, the NKC. These supplements include Alpha LipoicAcid, Arabinoxylin, Curcumin, Garlic, Genistein, Ginseng, Lentinan, Mistletoe, N-Acetylcysteine, Resveratrol, Selenium, Vitamin B, Vitamin C, Vitamin D3, Vitamin E and zinc.

The formulation for cancer prevention &therapy

Our natural product research over nine years has been to discover effective over-the-counter-supplements, designed to facilitate classical treatments in fighting disease, specifically cancer. These supplements are important in antiangiogenic, immune and antioxidant defense systems. The components include Acetyl L-Carnitine (ALC), Alpha Lipoic Acid (ALA), Coenzyme Q10 (CoQ10), Curcumin with Piperine, Genistein, Lentinan, N-Acetylcysteine (NAC), Resveratrol, selenium, Vitamin B Complex, Vitamin C, Vitamin E and zinc. These supplement components are supported for human cancer by over 15,000 references in the scientific literature withover of these being published clinical trials, not including zinc and the vitamins. These chemical defined components from natural origins have demonstrated, either individually or collectively, to have antioxidant, anti-angiogenesis, and immune stimulation properties. Furthermore, the direct cancer cell cytotoxicity for Curcumin, Genistein and NAC have been shown. Some of the antiangiogenesis components that affect the majority if not all pathways of angiogenesis, such as Curcumin, Genistein and NAC, actually stimulate the in vivo production of natural antiangiogenic compounds that include Angiostatin, Endostatin and Thrombospotin 1. All of the components play a role in serving as either water or lipid soluble (able to cross the blood-brain barrier) antioxidants. Curcumin, Genistein, Resveratrol, Lentinan, NAC, zinc, selenium and vitamins B and C all stimulate the immune system. Except for ALC and CoQ10, the other components show anti-inflammatory activity. ALC, Resveratrol along with the B and C vitamins are helpful in treating fatigue. The components that help protect the brain and promote nerve regeneration include ALA, CoQ10, Resveratrol, NAC, selenium, zinc and the B and C vitamins. In conclusion, effective prevention and treatment for diseases such as cancer, heart disease and immune deficiency will require multiple compounds.

Anticancer Effects of Curcumin, Artemisinin, Genistein, and Resveratrol, and Vitamin C: Free Versus Liposomal Forms

Cancer prevention supplements, which also provide effective treatment with minimal side effects, are urgently needed. An accurate, fast assay system is described that reveals the ability of chemically defined products, such as curcumin, genistein, resveratrol, artemisinin, and vitamin C, to kill K562 Erythroleukemic cells in vitro. In addition, curcumin and vitamin C were encapsulated into fatty acid micelles named NutraNanoSpheresTM (NNS) using all natural products. A unique viability stain, which allows the rapid staining of dead cells by membrane penetration using Propidium Iodide, was used to measure the cell viability by flow cytometry. Cell death by alteration of the cell membranes could be seen within 30 s of exposure to curcumin. The other free components required 0.5 - 70 h to see maximum killing, suggesting a more metabolic and/or apoptotic route of cancer cell destruction. Vitamin C up to 1 × 104 μmol/well did not affect K562 cell viability. The vitamin C-NNS (3.2 nm diameter-60 mg/50 μL) showed an LD50 = 133 μmol/well ± 11 SD (n = 4), which was over 75 times more potent than the free vitamin C. The curcumin-NNS (7.4 nm diameter-25 mg/50 μL) resulted in an LD50 = 41.3 μmol/well ± 5.6 SD (n = 8) and represented a 264 fold increase in activity to destroy the cancer cells. The clinical goal is to develop water-soluble mixtures of anti-cancer compounds in the NNS with their high bioavailability (>90%) and without degradation in the stomach for preventing and curing cancer.
