To observe the effects of simulated microgravity on beta islet cell culture, we have compared the survival rates and the insulin levels of the isolated rat islet cells cultured at micro- and normal gravity conditions. The survival rates of the cells cultured were determined by acridine orange-propidium iodide double-staining on day 3, 7 and 14. The morphology of the cells was observed by electron microscopy. Insulin levels were measured by radio immuno assays. Our results show that the cell number cultured under the microgravity condition is significantly higher than that under the routine condition (P<0.01). Some tubular structure shown by transmission electron microscopy, possibly for the transport of nutrients, were formed intercellularly in the microgravity cultured group on day 7. There were also abundant secretion particles and mitochondria in the cytoplasm of the cells. Equine infectious anemia virus strain L (EIAV-L) is the parental virulent virus of equine infectious anemia donkey leukocyte attenuated vaccine (DLA EIAV). In this study, peripheral blood leukocytes(PBL) were collected from a horse infected with *** PBL DNAs were *** EIAV-L proviral DNA was amplified in four parts covering the entire proviral genomic sequence by polymerase chain reaction (PCR). Each of the four parts was cloned into the plasmid pBluescript SK, and the recombinant plasmids were designated as p2.8, p2.4. p3.1, and p1.2 respectively. After identification with restriction digestion, the inserts within the four plasmids were sequenced. The complete nucleotide sequence of EIAV-L provirus was determined by analyzing each of the four parts and connecting them as a whole. The genome of EIAV-L is 8235 bp in length, and G + C content is 38%. The comparison analysis by the computer software DNASIS showed that the sequence of EIAV-L shares 98.4% and 96.9% identities with that of D-A EIAV and DLA EIAV respectively. The high homology between these strains showed that they were genetically related. The homology between EIAV-L and D-A EIAV is higher than that between EIAV-L and DLA EIAV, and this is consistent with the derivation progress of DLA EIAV. At both ends of EIAV-L provirus, there is an identical long terminal repeat (LTR) sequence of 316bp in length. The LTR consists of U3, R, and U5 regions. The genome of EIAV-L provirus has three long open reading frames(ORF) corresponding to gag, pol and env genes respectively. The gag gene is 1200bp and located at position 613-1912nt. The pol gene is 3402bp and located at position 1708-5109nt. There is a termination codon within the env dividing it into two parts, envl of 699bp (position 5305-6003nt)and env2 of 1827bp (position 6073-7899nt). 应用ELISA方法快速检测IHNV—B病毒

A reverse transcription-polymerase chain reaction (RT-PCR) procedure for the detection of avian, duck and goose reovirus (ARV, DRV and GRV) RNA from cell culture supernatant and clinical samples was established. The RNA of the virus was purified by the Trizol reagent and reverse transcription made by using random hexamers. The amplified, cloned, and sequenced 598-base pairs products on the σA-encoding gene fragment from 24 isolates (5 DRV, 3 GRV, and 8 ARV ranging more than 30 years) indicated that the primer regions were well conserved. The sensitivity of this method was determined to be 10-2 PFU. Specificity of the RT-PCR method was determined by testing specimens containing avian influenza A viruses, newcastle disease virus, and Infectious bronchitis virus, all of which yielded negative results with (?)discernible background. The efficiency of the system for detection of ARV, DRV, and GRV directly in 71/83 clinical samples was confirmed. The nucleotide sequence analysis indicates that all the duck and goose reoviruses isolated from China different locale and year were closely related, showing 97.4-100 % identity to each other, but only with 86.7-88.5 % identity to French MDRV 89026. The homology of amplified σA-encoding gene and amino acids sequences in this study and GenBank were varied from 74.2-78.4% and 87.4-92.5%, respectively, between duck/geese and chicken species. Reoviruses σA-encoding gene segment shared 88.6-100% and 87.3-100% nucleotide identities, respectively, within duck/geese and chicken species, Phylogenetic analysis based on σA-encoding gene indicated that GRV with DRV segregated into the same specified genogroup within orthoreoviruses genus subgroup 11 and more related to ARV than to Nelson Bay virus (NB V). Overall, this study developed a sensitive and specific technique for the identification ARV, DRV and GRV and sequencing analysis enhanced our understudying of ARV, DRV, and GRV evolution relationship.

采用酶联免疫吸附试验（ELISA）快速诊断病毒性疾病,由于具有灵敏度高、特异性强和简便等优点,已在医学和兽医学上得到了广泛的应用。在鱼类病毒检测方面,Dixon,P.F.和Hill,B.J.用ELI-SA方法（1983）快速检测传染性胰脏坏死症病毒（IPNV）和（1984）几种鱼类弹状病毒,江育林等（1990）曾报道用此技术快速检测IPN病毒,1990年在我国本溪虹鳟鱼苗中分离出一株弹状病毒,暂称之为传染性造血器官坏死症病毒中国本溪株（IHNV-B）,由于该病毒对虹鳟（rainbow trout）鱼苗危害极为严重,为预防和控制其蔓延,本文报道了应用ELISA方法对感染的鱼苗、鱼卵和细胞培养物中的IHNV-B病毒进行快速检测,可用目测或酶标检测仪来判定结果。

CELO病毒的DNA长43.8kb,具有末端倒转重复序列(ITR),基因结构与人腺病毒的比较,无可识别的E1、E3和E4区,但存在可替代的新读码框。2个fiber基因是病毒毒力基因,表达2个不同的蛋白,在病毒复制扩散中起作用,缺失fiber1导致柯萨奇病毒和腺病毒受体(CAR)的特异性转导作用消失,使CELO病毒只能感染禽类细胞,不能感染哺乳类动物细胞。在基因组的某些区段可以删除并插入外源基因不影响病毒复制。CELO病毒及其载体可在廉价的鸡胚中得到大量复制。CELO病毒的这些特性为其发展成新型基因工程栽体提供了条件。插入表达外源基因制备疫苗成为可能,新型的无病毒基因的载体也可以应用ITR及一些调控元件、包装信号共同构建形成。CELO病毒与人腺病毒属于同一属,CELO病毒作为基因工程载体有同样的特性,并且具有种属特异性和安全性,在动物基因工程疫苗的研制方面有特殊而广阔的研究和应用前景。

Based on the introduction of the status of animal vaccine industry in China,the strength,weakness,opportunity and threat of animal vaccine industry are analyzed by SWOT. Among them,strength is mainly reflected in the broad market,and the favorable industrial policy and development environment. Weakness is the irrational structure of product,the nonstandard production inspection,and the irregular market. Opportunity is mainly reflected in improving government's emphasis on animal security,expanding the scope of compulsory immunization,increasing the proportion of scale breeding,upgrading animal vaccine in China,speeding up industry consolidation and improving market concentration. Threat is manifested mainly in the poor market consciousness due to the long-term planned economy. And animal vaccine enterprises in China are at a relatively disadvantage position during the competition with international companies. Based on these,countermeasures to promote the development of animal vaccine enterprises in China are put forward from the aspects of both government and enterprise. On the one hand,government should improve the laws and regulations and promote the healthy development of vaccine industry,strengthen the manufacturers' consciousness of quality and biological safety,and create a favorable environment for market competition. On the other hand,enterprises should adjust the product structure and strengthen research and innovation,improve the brand competitiveness,take the road of common development,enhance the management capacity of the enterprises,and promote customer service.
