Characterization of heat shock protein 90, 70 and their transcriptional expression patterns on high temperature in adult of Graphofita molesta （Busck）

Grapholita molesta （Busck） is a worldwide insect pest damaging stone and pome fruits. High temperature can significantly affect insect survival, development and fecundity. Heat shock protein （Hsp） genes were speculated to possess a pivotal function in response to high temperature stress. In this study, two full-length Hsp genes, Gmhsp90 and GmhspTO, were cloned from G. molesta using rapid amplification of complementary DNA ends （RACE）. The open reading frames of Gmhsp90 and Gmhsp70 obtained were 2 148 bp and 1 998 bp in length, respectively. Their deduced amino acids showed high homology to Hsp genes of other species. Subsequently, the transcriptional expression of Gmhsp90 and Gmhsp70 in G. molesta adults exposed at various temperatures （26, 29, 32, 35, 38, 41 and 44℃） for 1 h and at 41℃ for various time duration （0, 15, 30, 45, 60, 75, 90 and 105 min） were investigated via real time quantitative polymerase chain reaction （qPCR）. The relative expression levels of Gmhsp90 and Gmhsp70 in G. molesta adults were both up-regulated with the rise of temperature and time duration. In addition, the Gmhsp70 usually showed a higher transcription accumulation than Gmhsp90. Interestingly, Gmhsp90 and Gmhsp 70 in female adults could be induced much earlier than that in males, and the effective induction temperature in females was also lower than that in males. The distinct expression profiles of Gmhsp90 and Gmhsp 70 indicated that Gmhsp90 and Gmhsp 70 may play important roles in G. molesta adults responding to a thermal threat, and there is difference on induction between sexes.

Development of a novel staging classification for Siewert Ⅱ adenocarcinoma of the esophagogastric junction after neoadjuvant chemotherapy

BACKGROUND Stage classification for Siewert Ⅱ adenocarcinoma of the esophagogastric junction(AEG)treated with neoadjuvant chemotherapy(NAC)has not been *** To investigate the optimal stage classification for Siewert Ⅱ AEG with *** A nomogram was established based on Cox regression model that analyzed variables associated with overall survival(OS)and disease-specific survival(DSS).The nomogram performance in terms of discrimination and calibration ability was evaluated using the likelihood-ratio test,Akaike information criterion,Harrell concordance index,time-receiver operating characteristic curve,and decision curve *** Data from 725 patients with Siewert type Ⅱ AEG who underwent neoadjuvant therapy and gastrectomy were obtained from the Surveillance,Epidemiology,and End Results *** and multivariate analyses revealed that sex,marital status,race,ypT stage,and ypN stage were independent prognostic factors of OS,whereas sex,race,ypT stage,and ypN stage were independent prognostic factors for *** factors were incorporated into the OS and DSS *** novel nomogram model performed better in terms of OS and DSS prediction compared to the 8th American Joint Committee of Cancer pathological staging system for esophageal and gastric ***,a user-friendly web application was developed for clinical *** The nomogram established specifically for patients with Siewert type Ⅱ AEG receiving NAC demonstrated good prognostic *** using external data is warranted before its widespread clinical application.

Structured illumination microscopy(SIM)achieves super-resolution(SR)by modulating the high-frequency information of the sample into the passband of the optical system and subsequent image *** traditional Wiener-filtering-based reconstruction algorithm operates in the Fourier domain,it requires prior knowledge of the sinusoidal illumination patterns which makes the time-consuming procedure of parameter estimation to raw datasets necessary,besides,the parameter estimation is sensitive to noise or aberration-induced pattern distortion which leads to reconstruction ***,we propose a spatial-domain image reconstruction method that does not require parameter estimation but calculates patterns from raw datasets,and a reconstructed image can be obtained just by calculating the spatial covariance of differential calculated patterns and differential filtered datasets(the notch filtering operation is performed to the raw datasets for attenuating and compensating the optical transfer function(OTF)).Experiments on reconstructing raw datasets including nonbiological,biological,and simulated samples demonstrate that our method has SR capability,high reconstruction speed,and high robustness to aberration and noise.

Mottled dolomite in the lower Cambrian Longwangmiao formation in the Northern Sichuan Basin, South China

At the end of Early Cambrian time, the Sichuan basin（South China） was located in a wide carbonate platform, with hundreds of meters of carbonate deposited. The Longwangmiao Formation carbonate in Sichuan basin is partially to completely dolomitized, displaying a mottled texture in the northern area of the exposure. The mottled dolomitic limestone developed parallel to bedding, with shape irregular boundaries with limestone that has not been dolomitized. The mottled dolomite is composed of powder crystalline and finely crystalline dolomite, while the matrix limestone is composed of micritic calcite. the isotopic composition of mottled dolomite(δ13C = +0.29‰PDB, δ18O =-1.15‰PDB) is similar to that of micrite calcite(δ13C =-0.49‰PDB, δ18O =-1.45‰PDB). Both isotopic values and trace element data indicate that the dolomitized fluid is originated from sea water. Some beds contain gypsum pseudomorphs and mud cracks, indicating a shallow and evaporative environment with local high salinity during deposition. Dolomitization likely took place early, in part as a result of sea water salinity concentration. Trace fossils thalassinoides horizontalis, thalassinoides callianassa and planolites developed in the Longwangmiao Formation, and the sharp edges of mottled dolomite are similar to these trace fossils. The beds are intensely bioturbated. In the burrow network, the sediments and burrow fill were coarse and loose with little clay, and it is interpreted here as being easier to be dolomitized than the surrounding *** dolomitization is thus interpreted to have occurred in the burrow system, and the degree of dolomitization was related to the degree of bioturbation, which is controlled by the trace-making creatures.

Humic acid(HA)is a typical refractory organicmatter,so it is of great significance to investigate its effect on the performance of Anammox granular *** the dosage ofHA≤50 mg/L,HA promotes the total nitrogen removal rate(NRR)to 1.45 kg/(m^(3)·day).When HA was between 50 and 100 mg/L,the NRR of Anammox was *** this time,the adsorption of HA causes the sludge to gradually turn fromred to brown,but the activities of heme and enzymes showed that its capacitywas not *** HA levels reached 250 mg/L,the NRR dropped to 0.11 kg/(m3·day).Moderate HA levels promoted the release of extracellular polymeric substance(EPS),but excessive HA levels lead to a decrease in EPS *** inhibited Anammox activity,which indirectly hindered the transmission of substrate and accumulated substrate *** HA promoted the increase of heterotrophicmicrobial abundance in Anammox system,the microbial diversity decreased *** the increase of HA concentration,the abundance of Candidatus_Brocadia,the main functional microorganism of Anammox system,decreased gradually,while the abundance of Candidatus_Kuenenia increased gradually.

Data-driven discovery of partial differential equations(PDEs)has recently made tremendous progress,and many canonical PDEs have been discovered successfully for proof of ***,determining the most proper PDE without prior references remains challenging in terms of practical *** this work,a physics-informed information criterion(PIC)is proposed to measure the parsimony and precision of the discovered PDE *** proposed PIC achieves satisfactory robustness to highly noisy and sparse data on 7 canonical PDEs from different physical scenes,which confirms its ability to handle difficult *** PIC is also employed to discover unrevealed macroscale governing equations from microscopic simulation data in an actual physical *** results show that the discovered macroscale PDE is precise and parsimonious and satisfies underlying symmetries,which facilitates understanding and simulation of the physical *** proposition of the PIC enables practical applications of PDE discovery in discovering unrevealed governing equations in broader physical scenes.

The treatment and disposal of radioactive waste are presently facing great *** ion exchange resins have become a focus of attention due to their high production and serious environmental *** this paper,a simplified model of cationic exchange resin is proposed,and the degradation processes of cationic resin monomer initiated by hydroxyl radicals(·OH)are clarified by combining statistical molecular fragmentation(SMF)model and density functional theory(DFT)*** prediction of active sites indicates that the S-O bonds and the C-S bond of the sulfonic group are more likely to react during the *** meta-position of the sulfonic group on the benzene ring is the most active site,and the benzene ring without the sulfonic group has a certain *** C11-C14 and C17-C20 bonds,on the carbon skeleton,are the most easily *** is also found that dihydroxy addition and elimination reactions play a major role in the process of desulfonation,carbon skeleton cleavage and benzene ring *** decomposition mechanisms found through the combination of physical models and chemical calculations,provide theoretical guidance for the treatment of complex polycyclic aromatic hydrocarbons.
