Aim:To explore the intracellular mechanisms underlying the survival/differentia-tion effect of the glial cell line-derived neurotrophic factor(GDNF)on dopamine(DA)***:Midbrain slice culture and primary cell culture wereestablished,and the cultures were divided into 3 groups:control group,GDNFgroup,and the phosphatidylinositol 3-kinase/Akt(PI3-K/Akt)*** the expression of tyrosine hydroxylase(TH)was detected byimmunostaining as well as Western ***:GDNF treatment inducedan increase in the number of TH-immunoreactive(ir)cells and the neurite numberof TH-ir cells,as well as in the level of TH expression in cultures(Number of TH-ir cells in the slice culture:control group,8.76±0.75;GDNF group,18.63±*** of TH-ir cells and neurite number of TH-ir cells in cell culture:controlgroup,3.65±0.88 and 2.49±0.42;GDNF group,6.01±0.43 and 4.89±0.46).Meanwhile,the stimulation of cultured cells with GDNF increased the phosphorylation of Akt,which is a downstream effector of PI3-K/*** effects of GDNF were specifi-cally blocked by the inhibitor of the PI3-K/Akt pathway,wortmannin(Number ofTH-ir cells in slice culture:PI3-K/Akt pathway-inhibited group,6.98±***-ber of TH-ir cells and neurite number of TH-ir cells in cell culture:PI3-K/Aktpathway-inhibited group,3.79±0.62 and 2.50±0.25,respectively).Conclusion:The PI3-K/Akt pathway mediates the survival/differentiation effect of GDNF onDA cells.

This paper is going to review the state-of-the-art of the high-speed 850/940-nm vertical cavity surface emitting laser（VCSEL）, discussing the structural design, mode control and the related data transmission performance. InGaAs/AlGaAsmultiple quantum well （MQW） was used to increase the differential gain and photon density in VCSEL. The multiple oxidelayers and oxide-confined aperture were well designed in VCSEL to decrease the parasitic capacitance and generate single mode （SM） VCSEL. The maximal modulation bandwidth of 30 GHz was achieved with well-designed VCSEL structure. At the end of the paper, other applications of the near-infrared VCSELs are discussed.

At least six major genotypes of Hepatitis C virus （HCV） cause liver diseases worldwide. The efficacy rates with current standard of care are about 50% against genotype 1, the most prevalent strain in the United States, Europe and Japan. Therefore more effective pan-genotypic therapies are needed. HCV RNA replication provides a number of validated targets for virus-specific and potentially pan-genotypic inhibitors. In vitro assays capturing the different steps of RNA synthesis are needed not only to identify new inhibitors, but also to examine their mechanisms of action. This review attempts to provide a comprehensive summary of the biochemical, cell-based and animal model systems to assess HCV polymerase activity and HCV RNA replication that should be useful for both basic research and applied studies.

Glutathione S-transferases (GSTs) play an important role in the detoxification of polycyclic aromatic hydrocarbons and aromatic amines, the toxic substances contained in cigarettes. GST Omega 1 (GSTO1) not only utilizes glutathione in conjugation reaction but also contributes to the biotransformation of several xenobiotics. A single nucleotide polymorphism (Ala-140Asp) of GSTO1 gene causing variations in enzyme activity may influence individual susceptibility to bladder cancer (BC). It is hypothesized that genetic polymorphism of GSTO1 gene has an effect on BC risk in particular by interacting with cigarette smoking. A total of histopathologically confirmed 300 BC patients and 300 cancer-free controls were recruited from February 2002 to February 2009. Genotyping of the GSTO1 Ala140Asp polymorphism was determined using a polymerase chain reaction-restricted fragment length polymorphism (PCR-RFLP) method. The odds ratio (OR) and 95% confidence interval (CI) were calculated as a measure of the combined effect of cigarette smoking and the GSTO1 Ala140Asp polymorphism on BC risk. We found that study subjects with the GSTO1 Ala/Ala genotype have a significantly increased BC risk (OR = 1.5;95% CI = 1.1 - 2.7). A statistically significant increased BC risk was also found in ever smokers with the GSTO1 Ala/Ala genotype (OR = 4.9;95%CI = 2.8 - 9.7). In conclusion, this study provides an epidemiologic evidence of a significantly increased BC risk among ever smokers with the GSTO1 Ala/Ala genotype.

以菊科植物牛蒡(A rctium lapp a L.)的等外根为材料,经热水浸提、木瓜蛋白酶-Sevag法脱蛋白、乙醇沉淀和SephadexG-50凝胶柱层析分离纯化,得到水溶性的牛蒡寡糖(BO S-2).采用化学分析方法研究了BO S-2的理化性质,并通过TLC,GC-M S,HP-GPC,UV,IR,1H NM R及13C NM R等对BO S-2的纯度、结构和组成进行表征.结果表明,BO S-2由果糖和葡萄糖组成,摩尔比约为12∶1,纯度较高,为均一性组分;由12个呋喃型的果糖以β(2→1)糖苷键相连,1个吡喃型的葡萄糖以α(1→2)糖苷键连接在果糖末端的线性直链结构,是一种菊糖构型的低聚果糖.

Intraoperative confirmation of negative resection margins is an essential component of soft tissue sarcoma *** section examination of samples from the resection bed after excision of sarcomas is the gold standard for intraoperative assessment of margin ***,it takes time to complete histologic examination of these samples,and the technique does not provide real-time diagnosis in the operating room(OR),which delays completion of the *** paper presents a study and development of sensing technology using Raman spectroscopy that could be used for detection and classification of the tumor after resection with negative sarcoma margins in real *** acquired Raman spectra from samples of sarcoma and surrounding benign muscle,fat,and dermis during surgery and developed(i)a quantitative method(QM)and(ii)a machine learning method(MLM)to assess the spectral patterns and determine if they could accurately identify these tissue types when compared to findings in adjacent H&E-stained frozen *** classification accuracy(>85%)was achieved with both methods,indicating that these four types of tissue can be identified using the analytical methodology.A hand-held Raman probe could be employed to further develop the methodology to obtain spectra in the OR to provide real-time in vivo capability for the assessment of sarcoma resection margin status.

Located at the collision boundary between the Philippine Sea Plate and the Eurasian Continental Plate,the island of Taiwan is generally recognized as an important example in the MARGINS Program Science Plan and "source-to-sink" research because of its high tectonic activity,heavy rainfall and unique *** suspended sediment loads are transported to the adjacent ocean by Taiwan Residents rivers every year,making Taiwan an important source of sediments into the adjacent seas and a natural laboratory for studying the systemic movement of fluvial sediments from source to sink.A detailed study on the clay mineral composition of surface sediments collected from the drainage basins of 12 Taiwan Residents rivers using X-ray diffraction methods was *** results indicated that the clay mineral assemblages consisted dominantly of illite(approximately 73%) and chlorite(approximately 24%),with lesser abundances of kaolinite(approximately 3%) and even lower levels of smectite from the Danshuei River sediments in northwestern *** Jhuoshuei River sediments from western Taiwan contained clay mineral assemblages that consisted of illite(approximately 75%) and chlorite(approximately 25%),but they lacked kaolinite and *** southwestern Taiwan,the clay mineral assemblages were dominated by illite(approximately 75%) and chlorite(approximately 23%),but had a low abundance of kaolinite(generally kaolinite and smectite being scarce,and these trends are different from those in China's mainland *** clay miner
