＜正＞ We use the acetylcholine binding protein （AChBP） from mollusks as a soluble surrogate for the extracellular ligand binding domain of nicotinic acetylcholine receptors （ nAChR）. Ligand binding in the nAChR extracellular domain induces conformational states that allosterically open an ion channel. nAChR states have not been studied at atomic resolution. We have solved X- ray crystal structures of receptor agonists, antagonists and non- competitive ligand bound to AChBP. These structures reveal large conformation changes in the ligand binding pocket and distinct interface binding surfaces. Conformational changes in loop C reveal a general mechanism for agonism and partial agonism. X- ray structures of noncompetitivereceptor ligands galanthamine. cocaine, and thienyl - cylohexylpiperidine（TCP） in complex with AChBP reveal valuable information for accurately describing non- competitive receptor modulation. A crystal structure of apo AChBP is presented and compared to that of bound agonists, lobeline and epibatidine. and antagonists, alpha-conotoxin Iml and methyllycaconitine.

A High-Performing and Cost-Effective SNP Genotyping Method Using rhPCR and Universal Reporters

We have developed a novel dual enzyme chemistry called rhAmp®SNP genotyping based on RNase H2-dependent PCR (rhPCR) that provides high signal and specificity for SNP analysis. rhAmp SNP genotyping combines a unique two-enzyme system with 3’ end blocked DNA-RNA hybrid primers to interrogate SNP loci. Activation of the blocked primers occurs upon hybridization to its perfectly matched target, which eliminates or greatly reduces primer dimers. A thermostable hot-start RNase H2 cleaves the primer immediately 5’ of the ribose sugar, releasing the blocking group and allowing primer extension. PCR specificity is further improved with the use of a mutant Taq DNA polymerase, resulting in improved allelic discrimination. Signal generation is obtained using a universal reporter system which requires only two reporter probes for any bi-allelic SNP. 1000 randomly selected SNPs were chosen to validate the 95% design rate of the design pipeline. A subsampling of 130 human SNP targets was tested and achieved a 98% call rate, and 99% call accuracy. rhAmp SNP genotyping assays are compatible with various qPCR instruments including QuantStudioTM 7 Flex, CFX384TM, IntelliQube®, and Biomark HDTM. In comparison to TaqMan®, rhAmp SNP genotyping assays show higher signal (Rn) and greater cluster separation, resulting in more reliable SNP genotyping performance. The rhAmp SNP genotyping solution is suited for high-throughput SNP genotyping applications in humans and plants.

Estimation of alfalfa fall dormancy using spaced plant and sward trials across multiple environments

Background:Alfalfa is a globally important forage crop.Cultivars are characterized by fall dormancy(FD).FD affects biomass yield and winter survival and is used to identify appropriate growing regions of cultivars.It has historically been assessed by measuring the natural height of regrowth in autumn of spaced plants in the field.Because commercial alfalfa is normally grown as a solid planted sward,FD could be different if plants were grown under real production conditions.The objective of this experiment was to assess whether FD ratings obtained from swards were similar to those obtained on spaced plants.Methods:We evaluated 20 alfalfa cultivars in field trials established in 2015 at five locations in the United States.We harvested the trials in early autumn and measured regrowth plant height approximately 3 weeks later in 2015 and 2016.Results:Autumn plant height responded as expected for the established check cultivars in all locations.Fall dormancy ratings for cultivars under both management systems were highly correlated.Conclusions:Estimating FD from spaced plant height measurements in autumn is very robust,and using height data from sward plots gives equivalent results to that measured in spaced plant nurseries.This finding has many practical advantages.

The cryptic lifestyle of most fungi necessitates molecular identification of the guild in environmental studies.Over the past decades,rapid development and affordability of molecular tools have tremendously improved insights of the fungal diversity in all ecosystems and habitats.Yet,in spite of the progress of molecular methods,knowledge about functional properties of the fungal taxa is vague and interpretation of environmental studies in an ecologically meaningful manner remains challenging.In order to facilitate functional assignments and ecological interpretation of environmental studies we introduce a user friendly traits and character database FungalTraits operating at genus and species hypothesis levels.Combining the information from previous efforts such as FUNGuild and FunFun together with involvement of expert knowledge,we reannotated 10,210 and 151 fungal and Stramenopila genera,respectively.This resulted in a stand-alone spreadsheet dataset covering 17 lifestyle related traits of fungal and Stramenopila genera,designed for rapid functional assignments of environmental stud-ies.In order to assign the trait states to fungal species hypotheses,the scientific community of experts manually categorised and assigned available trait information to 697,413 fungal ITS sequences.On the basis of those sequences we were able to summarise trait and host information into 92,623 fungal species hypotheses at 1%dissimilarity threshold.
