Delivery of dsRNA for RNAi in insects： an overview and future directions

RNA interference （RNAi） refers to the process of exogenous double-stranded RNA （dsRNA） silencing the complementary endogenous messenger RNA. RNAi has been widely used in entomological research for functional genomics in a variety of insects and its potential for RNAi-based pest control has been increasingly emphasized mainly because of its high specificity. This review focuses on the approaches of introducing dsRNA into insect cells or insect bodies to induce effective RNAi. The three most common delivery methods, namely, microinjection, ingestion, and soaking, are illustrated in details and their advantages and limitations are summarized for purpose of feasible RNAi research. In this review, we also briefly introduce the two possible dsRNA uptake machineries, other dsRNA delivery methods and the history of RNAi in entomology. Factors that influence the specificity and efficiency of RNAi such as transfection reagents, selection of dsRNA region, length, and stability of dsRNA in RNAi research are discussed for further studies.

Background: Caregivers experience problems when caring for people with intellectual disabilities who are terminally ill. Aim: The aim of this study is to gain a deeper understanding of the experience of the caregiver when caring for a person with intellectual disabilities who is terminally ill and what influences this experience. Design: A qualitative research was carried out following the principles of Grounded Theory (GT) method, through fifteen interviews with caregivers in a living facility with continuous care. Results: The results show how caregivers strive to create meaningful moments for their client during the last period of his life. Since very few of the clients can clearly express their wishes and feelings, continuous observation is required of the caregivers for the interpretation of small signals of their clients. In that way, the appropriateness of their actions can be made certain. The results show that because of their involvement, caregivers are able to recognize clients’ signals. When a client is dying, the special atmosphere created on the ward contributes to coping with the situation and a positive experience. Conclusion: Direct-caregivers caring for a client with an intellectual disability who is terminally ill, experience an intense period of insecurity and also a special period meaningful when caring a dying client with intellectual disabilities. Their relationship with the client makes them to be more able to strive for client’s comfort. The results also show that some factors such as enough staff, clear and open communication with the physician and support of the manager contribute to the quality of palliative care on the ward.

Metformin is a worldwide used and effective antihyperglycemic agent. It is generally well tolerated but contraindicated in severe renal dysfunction. Metformin does not cause hypoglycaemia, however this case describes hypoglycaemia after an overdose. Another well-known, but rare and potentially life-threatening complication of metformin is the occurrence of LA, the so-called metformin-associated lactic acidosis (MALA). The severe consequences of an overdose of metformin, as well as the pathophysiologic mechanisms and the clinical consequences of LA in combination with severe hypoglycaemia are described in this report. Early recognition and swift clinical intervention is of paramount importance in case of auto-intoxication with metformin to prevent a lethal outcome.

Traumatic Brain Injury in Prisoners: Relation to Risky Decision-Making, Aggression and Criminal Behavior

The objective of this cross-sectional study was to investigate if Traumatic Brain Injury (TBI) is related to decision-making deficits as well as to heightened levels of (reactive) aggression and criminal behavior in a group of Dutch male prisoners. Characteristics of aggression were assessed using three self-report questionnaires as well as systematic staff observations, complemented with data from criminal records. A semi-structured interview was conducted to rate lifetime prevalence of TBI and the Iowa Gambling Task (IGT) was completed to assess risky decision-making. Out of the total of 133 participants, 74 (55.6%) reported having sustained one or more incidents with potential TBI. Statistical comparisons between offenders with and without TBI indicated that the offenders with TBI reported significantly higher levels of (reactive) aggression on two out of three questionnaires than offenders without TBI (p-values ranging from 0.003 to 0.008), but no significant differences were found on staff observations of aggression and on the IGT. The higher conviction rates in offenders with TBI compared to the non-TBI group showed a trend. The confirmed relationship between TBI and higher aggressive tendencies is especially relevant for forensic populations due to the high prevalence rates of TBI in offenders. How this relates to specific cognitive processes remains unclear. The lack of a statistical difference between offenders with and without TBI on the IGT is in contrast with literature in non-offender samples, but seems to be in line with other studies in forensic populations.
