为了验证小分子RNA let-7a与其预测靶基因CC趋化因子受体7(CCR7)之间的关系,采用生物信息学软件预测CCR7的3′非编码区(3′UTR)是否存在与let-7a的靶向结合位点;利用PCR扩增出包含靶向结合位点区域的CCR7-3′UTR,并构建其荧光素酶表达载体;将验证后的阳性重组子瞬时共转染人胚胎肾细胞293(293T),进行双荧光素酶报告基因检测;通过将合成的let-7a模拟物和let-7a抑制剂分别转染奶牛乳腺上皮细胞MAC-T,利用qRT-PCR和ELISA检测let-7a与CCR7基因mRNA和蛋白表达水平的变化,验证两者之间的靶向调控关系。菌液PCR和双酶切结果表明,成功构建了重组双荧光素酶报告质粒pMIR-REPORTTM-CCR7-3′UTR。双荧光素酶报告基因检测结果表明,let-7a与CCR7基因之间确实存在靶向调控关系。qRT-PCR和ELISA进一步证实了过表达let-7a能显著下调MAC-T细胞中CCR7基因mRNA和蛋白的表达量(P<0.05),两者存在负调控关系。本研究结果证实了let-7a与CCR7基因之间存在靶向调控关系,为进一步探究其分子机制提供参考。

Enhancement of oxygen reduction activity and stability via introducing acid-resistant refractory Mo and regulating the near-surface Pt content

Although Pt Ni catalyst possesses good oxygen reduction activity, its poor stability is the main obstacle for the commercialization of proton exchange membrane fuel cells(PEMFCs). In this work, we introduce the acid-resistant refractory Mo to enhance the structure stability and modify the electronic structure of Pt in the prepared PtNi catalyst, improving the catalytic activity for oxygen reduction reaction(ORR). In addition, near-surface Pt content in the nanoparticle is also optimized to balance the ORR activity and stability. The electrochemical results show that the alloy formed by Mo and Pt Ni is obviously more stable than the PtNi alloy alone, because the acid-resistant Mo and its oxides effectively prevent the dissolution of Pt. Especially, the Pt3 Ni3 MoN/C exhibits the optimal ORR catalytic performance in O2-saturated 0.1 mol L^(-1) HClO4 aqueous solutions, with mass activity(MA) of 900 m A mg^(-1) Pt at 0.90 V vs. RHE, which is 3.75 times enhancement compared with the commercial Pt/C(240 mA mg^(-1) Pt). After 30 k accelerated durability tests, its MA(690 m A mg^(-1) Pt) is still 2.88 times higher than the pristine Pt/C. This study thus provides a valuable method to design stable ORR catalysts with high efficiency and has great significance for the commercialization of PEMFCs.

AIM:To estimate the prevalence of the lactase non-persistent genotype(C/C-13910)in a northern Russian population in accordance with ethnicity,and to evaluate self-reported milk consumption depending on lactase activity. METHODS:Blood samples for genotyping lactase activity,defining the C/T-13910 variant by polymerase chain reaction,and direct sequencing were taken from 231 medical students of Russian origin aged 17-26 years. We analyzed milk product consumption by questionnaire which was specially designed for the estimation of milk consumption and abdominal complaints. RESULTS:We found that the prevalence of the C/C-13190 genotype in the northern Russian population was 35.6%.The other genotypes nearby C/T-13910 and associated with lactase activity were not present in the study *** consumption of milk among people with the non-persistent genotype tended to be lower than among the lactose tolerant subjects,but was not statistically ***:An investigation of the lactase persistent genotype in a northern Russian population has not been performed *** genotype did not affect the consumption of milk products in this population which could be explained by low consumption of milk products among the entire study population.

Inflammatory diseases such as rheumatoid arthritis (RA) can lead to chronic pain and joint deformity. First line treatment includes disease-modifying antirheumatic drugs (DMARDs) to slow disease progression, but RA patients are often left with debilitating symptoms. This article explores alternative topical and naturally based options to treat RA symptoms, with supporting evidence for their use. Considered options include Aloe vera, Perillyl Alcohol (POH), Aleurites moluccana, Calendula officinalis extract (quercetin), Achyranthes species, Guizhi Shaoyao Zhimu, Curcumin (turmeric), Wutou, Radix paeoniae extract (Paeoniflorin), Tripterygium wilfordii Hook F (TWHF), Capsaicin, Cannabinoids (CBD), and Berberine. Many of these products show promising data for supplemental treatment of RA through various mechanisms, with CBD and TWHF having the most robust evidence supporting their use.

BACKGROUND： The possibility of dietary ginger to enhance oxidative stress resistance and to extend life span was studied on Drosophila melanogaster. METHODS： Oxidative stress was induced by a reducing agent dithiothreitol. Experimental groups of male D. melanogaster were cultured on media containing： 1） no additive; 2） dithiothreitol, added into the nutritional mixture to the final concentration of 10 mM; 3） 25 mg of ginger powder g^-1 of the nutritional mixture; and 4） 10 mM ofdithiothreitol and 25 mg of ginger powder g l of the nutritional mixture. The number of alive fruit flies was inspected daily, and mean life span was determined for each experimental group. RESULTS： The addition of dithiothreitol to D. melanogaster nutritional mixture was established to result in an increase in concentration of two markers of oxidative stress conditions （thiobarbituric acid reactive substances as products of lipid peroxidation and carbonylated proteins as products of protein oxidation） in fly tissues. It was followed by significant reduction of mean life span and maximum life span of the last 10% of flies. Plant preparation, being added simultaneously with dithiothreitol, significantly diminished the negative effects of this xenobiotic. In conditions of additional stress load induced by hydrogen peroxide or high temperature, survival of insects treated with dithiothreitol on the background of ginger powder was the highest. CONCLUSIONS： Thus, the presented data give the evidence that ginger preparations can reduce oxidative stress outcomes and significantly increase the life expectancy of fruit flies in stress conditions.

Objective:To investigate the anti-inflammatory effects of Sanguisorbae Radix on contact dermatitis(CD).Methods:Mice were sensitized by painting 30μL of 1-fluoro-2,4-dinitrofluorobenzene(DNFB)onto each ear for 3 *** days later,mice were challenged by painting with 50μL of DNFB onto the shaved dorsum every 2 *** Radix methanol extract(MESR)was applied onto the shaved dorsum every 2 *** effects of MESR on skin thickness,skin weights,histopathological changes,skin lesions and cytokine production in DNFB-induced CD mice were investigated,as well as its effects on body weights and spleen/body weight ***:Topical application of MESR effectively inhibited enlargement of skin thickness and weight(P<0.05).MESR treatment also inhibited hyperplasia,spongiosis and immune cell infiltration induced by DNFB in inflamed tissues and improved lesions on dorsum skin in CD ***,treatment with MESR suppressed the increase in the levels of tumor necrosis factor α(TNF-α,P<0.01)and interferonγ(IFN-γ,P<0.05),***,MESR had no effect on body weight gain or spleen/body weight ***:These data suggest that MESR acts as an anti-inflammatory agent that decreases the production of TNF-α and IFN-γ,resulting in reductions of skin lesions and histopathological changes in inflamed skin tissues.
