Promoter silencing by ectopic?de novo?meth.lation of tumor suppressor genes h.s been proposed as comparable or equivalent to inactivating mutations as a factor in carcinogenesis. h.wever, th.s h.poth.ses h.d not previously been tested by h.gh.resolution,?h.gh.coverage wh.le-genome meth.lation profiling in primary carcinomas. We h.ve determined th. genomic meth.lation status of a series of primary mammary carcinomas and match.d control tissues by examination of more th.n 2.7 billion CpG dinucleotides. Most of th. tumors sh.wed variable losses of DNA meth.lation?from all sequence compartments, but increases in promoter meth.lation were infrequent, very small in extent, and were observed largely at CpG-poor promoters. De novo meth.lation at th. promoters?of proto-oncogenes and tumor suppressor genes occurred at approximately th. same frequency. Th. findings indicate th.t tumor suppressor silencing by?de novo?meth.lation is much.less common th.n currently believed. We put forward a h.poth.sis under wh.ch.th. demeth.lation commonly observed in carcinomas is a manifestation of a defensive system th.t kills incipient cancer cells.
