salvianolic acid B modulates the expression of drug-metabolizing enzymes in HepG2 cells

BACKGROUND: Enzymes involved in drug and xenobiotic metabolism have been considered to exist in two groups: phase I and phase II enzymes. Cytochrome P450 isoenzymes (CYPs) are the most important phase I enzymes in the metabolism of xenobiotics. The products of phase I metabolism are then acted upon by phase II enzymes, including glutathione s-transferases (GsTs). Herbs that inhibit CYPs such as CYP3A4 or that induce GsTs may have the potential to protect against chemical carcinogenesis since the mutagenic effects of carcinogens are often mediated through an excess of CYP-generated reactive intermediates. This study was designed to investigate the effects of salvianolic acid B (sal B), a pure compound extracted from Radix salviae Miltiorrhizae, a Chinese herb, on cell proliferation and CYP1A2 and CYP3A4 mRNA expression in the presence or absence of rifampicin, a potent inducer of CYPs and GsT protein expression in HepG2 cells. METHODs: HepG2 cells were incubated with different concentrations of sal B. Cell proliferation was determined by sYTOX-Green nucleic acid staining. CYP3A4 and CYP1A2 mRNA expression was assayed by real-time PCR. GsT protein expression was analyzed by Western blotting. REsULTs: Low concentrations of sal B (0-20 μmol/L) had no significant effects on cell proliferation, while higher concentrations (100-250 μmol/L) significantly inhibited proliferation in a concentration-dependent manner. Ten μmol/L sal B, but not 1 μmol/L, down-regulated CYP3A4 and CYP1A2 mRNA expression after 24 hours of incubation, whereas both 1 and 10 μmol/L sal B down-regulated CYP3A4mRNA expression after 96 hours of incubation; moreover, 1 and 10 μmol/L sal B inhibited CYP3A4 mRNA expression induced by rifampicin. Both 1 μmol/L and 10 μmol/L sal B increased GsT expression. CONCLUsION: sal B inhibits CYP3A4 and CYP1A2 mRNA expression and induces GsT expression in HepG2 cells.

An autonomous microgrid that runs on renewable energy sources is presented in this *** has a supercon-ducting magnetic energy storage(sMEs)device,wind energy-producing devices,and an energy storage ***,because such microgrids are nonlinear and the energy they create varies with time,controlling and managing the energy inside them is a difficult ***-order proportional integral(FOPI)controller is recommended for the current research to enhance a standalone microgrid’s energy management and *** suggested dedicated control for the sMEs comprises two loops:the outer loop,which uses the FOPI to regulate the DC-link voltage,and the inner loop,responsible for regulating the sMEs current,is constructed using the intelligent FOPI(iFOPI).The FOPI+iFOPI parameters are best developed using the dandelion optimizer(DO)approach to achieve the optimum *** suggested FOPI+iFOPI controller’s performance is contrasted with a conventional PI controller for variations in wind speed and microgrid *** optimal FOPI+iFOPI controller manages the voltage and frequency of the *** behavior of the microgrid as a reaction to step changes in load and wind speed was measured using the proposed *** simulations were used to evaluate the recommended system’s *** results of the simulations showed that throughout all interruptions,the recommended microgrid provided the load with AC power with a constant amplitude and *** addition,the required load demand was accurately ***,the microgrid functioned incredibly well despite sMEs and varying wind *** obtained under identical conditions were compared with and without the best FOPI+iFOPI *** utilizing the optimal FOPI+iFOPI controller with sMEs,it was found that the microgrid performed better than the microgrid without sMEs.

Geode, boudinage, and undulation structures are widely distributed in the siliceous beds of the Upper Cretaceous/Tertiary rocks in Jordan. Their formation was attributed to tectonic forces, syngenetic processes, organic disintegration processes, subaquatic gliding, compaction and settlement, and meteoritic impacts. In this work, the structural features in the siliceous beds of Jordan are attributed to an interplay of load and directed pressures, and mineralogical transformation processes (opal-A to opal-CT to quartz), governed by pH changes. Tectonic directed pressure was acting in an EsE-WsW direction and is common in the silicified limestone of Upper Cretaceous.

The network switches in the data plane of software Defined Networking (sDN) are empowered by an elementary process, in which enormous number of packets which resemble big volumes of data are classified into specific flows by matching them against a set of dynamic rules. This basic process accelerates the processing of data, so that instead of processing singular packets repeatedly, corresponding actions are performed on corresponding flows of packets. In this paper, first, we address limitations on a typical packet classification algorithm like Tuple space search (Tss). Then, we present a set of different scenarios to parallelize it on different parallel processing platforms, including Graphics Processing Units (GPUs), clusters of Central Processing Units (CPUs), and hybrid clusters. Experimental results show that the hybrid cluster provides the best platform for parallelizing packet classification algorithms, which promises the average throughput rate of 4.2 Million packets per second (Mpps). That is, the hybrid cluster produced by the integration of Compute Unified Device Architecture (CUDA), Message Passing Interface (MPI), and OpenMP programming model could classify 0.24 million packets per second more than the GPU cluster scheme. such a packet classifier satisfies the required processing speed in the programmable network systems that would be used to communicate big medical data.
