Identification of TWIST-interacting genes in prostate cancer

Prostate cancer is one of the most common cancers in men worldwide, and the number of diagnosed patients has dramatically increased in recent years. Currently, the clinical parameters used to diagnose prostate cancer, such as Gleason score, pathological tumor staging, and prostate-specific antigen(PSA) expression level, are considered insufficient to inform recommendation to guide clinical practice. Thus, identification of a novel biomarker is necessary. TWIST is one of the well-studied targets and is correlated with cancer invasion and metastasis in several human cancers. We have investigated two largest prostate cancer patient cohorts available in GEO database and found that TWIST expression is positive correlated with Gleason score and associated with poorer survival. By using a prostate cancer cohort and a prostate cancer cell line dataset, we have identified three potential downstream targets of TWIST, PPM1 A, SRP72 and TBCB. TWIST's prognostic capacity is lost when the gene is mutated. Further investigation in the prostate cancer cohort revealed that gene expression of SERPINA, STX7, PDIA2, FMP5, GP1 BB, VGLL4,KCNMA1, SHMT2, SAA4 and DIDO1 influence the prognostic significance of TWIST and vice versa. Importantly, eight out of these ten genes are prognostic indicator by itself. In conclusion, our study has further confirmed that TWIST is a prognostic marker in prostate cancer, identified its potential downstream targets and genes that could possibly give additional prognostic value to predict TWIST-mediated prostate cancer progression.

Antibody research targeting Cathepsin S for cancer therapy

Cathepsin S is a cysteine protease highly expressed in many type of cancers including colorectal, prostate, breast, and glioblastoma’s. It is involved in tumor progression through extracellular matrix remodeling. In recent years, antibody research specifically targeting Cathepsin S to block/inhibit tumorigenic effects were generated some positive preclinical data. This antagonistic antibody was demonstrating efficacy in multiple in vitro/in vivo cancer models both as a monotherapy and in combination with approved agents. This mini-review provides an overview of therapeutic antibody targeting Cathepsin S strategies in the last half decade, focusing on the rationale of cell-surface Cathepsin S targeted and their potential clinical application.

A prospective cohort of men with localized prostate cancer on active surveillance protocol in Hong Kong,China:what did we learn?

This study aimed to report the outcomes of active surveillance(AS)in the management of low-risk prostate cancer(PCa).It recruited87 men who were prospectively followed up according to the Prostate Cancer Research International Active Surveillance(PRIAS)protocol with local adaptation at SH Ho Urology Centre,Prince of Wales Hospital,Hong Kong,*** investigated the predictorsof disease progression and found that baseline prostate-specific antigen density(PSAD)and the presence of the highest ProstateImaging-Reporting and Data System(PI-RADS)score 5 lesion on magnetic resonance imaging(MRI)are significantly correlatedwith disease ***,men with PSAD>0.2 ng ml^(−2)or PI-RADS 4 or 5 lesions had significantly worse upgradingfree survival compared to those with PSAD≤0.2 ng ml−2 and PI-RADS 2 or 3 *** study concludes that AS is a safe andeffective management strategy for selected patients to defer radical treatment and that most disease progression can be detectedafter the first repeated *** combination of PSAD>0.2 ng ml^(−2)and PI-RADS 4 or 5 lesions may serve as a useful predictorof early disease progression and provide a guide to optimize follow-up protocols for men in different risk groups.

牛结核病发生率持续增长给农业生产造成巨大的经济损失。目前用于检测和控制牛结核病的方法(皮肤试验和γ-干扰素)缺乏预期敏感性和特异性,因此急需开发一种快速可靠的牛结核病血清学检测方法。用检测针对特效分枝杆菌抗原抗体的方法可以解决特异性和敏感性问题。用不同分枝杆菌对兔模型进行攻毒试验,通过体液免疫应答筛选分枝杆菌抗体,攻毒后2周可在血清中检测到针对抗原MTB40、ESAT6和CFP10的抗体,攻毒后8～11周可检测到针对Rv3870和Rv1580c的抗体。所选抗原与禽结核菌素无交叉反应,仅与牛结核菌素有轻微阳性反应。试验结果表明,特效分枝杆菌抗原反应板可用于血清样本牛分枝杆菌的鉴定,多种抗原组合将有利于牛结核病的诊断。

Transurethral resection of prostate for acute urinary retention is linked to shorter survival in younger men

It is largely unknown whether lower urinary tract symptoms(LUTS)or acute retention of urine(AROU)is linked to shorter life expectancy in *** conducted a multicenter,retrospective database analysis of patients undergoing transurethral resection of prostate(TURP)to study their *** Cox regression analysis and Kaplan-Meier analysis with stratification to age and indication of TURP were *** further performed an age-and sex-matched survival analysis with the general population using data from the Census and Statistics Department of the Hong Kong Special Administrative Region(Hong Kong,China).From January 2002 to December 2012,3496 patients undergoing TURP were included in our study,with 1764 patients in the LUTS group and 1732 patients in the AROU *** age,ischemic heart disease,cerebrovascular accident,and AROU were risk factors of *** aged80 *** to the general population,younger patients in the LUTS group appeared to have better survival(80 years,survival was similar with the general population in both the LUTS and AROU *** conclusion,AROU at young age was associated with mortality,while early detection and management of LUTS may improve survival.

目的:通过比较不同高雄激素血症(HA)表型下多囊卵巢综合征(PCOS)患者的一般临床特征和生化指标,中医证候的分布以及针刺联合枸橼酸克罗米芬促排卵后的妊娠结局,以求探索临床最敏感的雄激素指标。方法:选择“针刺联合克罗米芬对PCOS不孕妇女活产的影响临床试验(PCOSACt)”948例受试者数据,根据总睾酮(TT)值和游离雄激素指数(fai)值将患者分为:单TT升高为A组(143例)、单fai升高为B组(160例)、两者均升高为C组(295例)、均不升高为D组(350例)。结果:A组BMI、WC、WHR、黑棘皮评分、fai、FINS、HOMA-IR、TG、APOB、IR、高血脂症和脾虚痰湿型的发生率显著低于B、C组;SHBG、HDL-c和APOA1和肾虚肝郁型发生率显著高于B、C组(Pfai在各组最低;肾虚肝郁型发生率显著高于B、C组(Pfai升高常伴有更严重的肥胖和糖脂代谢紊乱;二者同时升高并不会比单fai升高时的代谢紊乱严重,但其排卵率、受孕率、临床妊娠率、活产率最低。
