The differential effects of prostate stromal cells derived from different zones on prostate cancer epithelial cells under the action of sex hormones

It is well known that prostate cancer （PCa） occurs predominantly in the peripheral zone （PZ）, whereas benign prostatic hyperplasia （BPH） typically develops in the transition zone. To identify possible mechanisms underlying zonal differences, we compared the effects of prostate stromal cells derived from the peripheral zone （PZsc） and the transition zone （TZsc） on a PCa epithelial cell line （PC3） in the presence of sex hormones. First, we observed that androgen receptor （AR） mRNA was more highly expressed in PZsc than TZsc when the cells were treated with dihydrotestosterone （DHT） and β-oestradiol （E2）（P〈0.05）. By ELISA, we looked for differences in the secretion of peptide growth factors from PZsc and TZsc. We found that keratinocyte growth factor （KGF） secretion increased with increasing concentrations of DHT （P〉0.01） and was higher in PZsc than TZsc. Under treatment with DHT plus E2, PZsc secreted more transforming growth factor-β1 （TGF-β1） than TZsc, but this pattern was reversed when the cells were treated with E2 only. With increasing concentrations of DHT, insulin-like growth factor-1 （IGF-1） secretion increased in PZsc but decreased in TZsc. To further characterize the effects of PZsc and TZsc on PC3 cells, we developed a coculture model and performed MTT assays, Western blot analysis and real-time RT-PCR. We found that PZsc promoted PC3 cell proliferation and progression better than TZsc, particularly when treated with 10 nmoll^- 1 DHT plus 10 nmoll^-1 E2. In conclusion, our data suggest that PZsc may have a greater capacity to induce PCa development and progression than TZsc via growth factors regulated by sex hormones. These findings provide possible mechanisms underlying zonal differences in prostate diseases, which may aid the search for novel therapeutic targets for PCa.

It showed high selectivity and sensitivity for Hg2＋ in acetonitrile-H20 （1 ：1, V ： V） solution. Dynamic light scatter-ing measurements were conducted to verify that the addition of Hg2 changed the particle size and induced fluorescence quenching.

Objective The Lincang granitic batholith, extending over 370 km from north to south with an area of more than 10000 km2, is the major part of the Sanjiang region in western Yunnan, SW China, and is one key area to study the evolution of the Tethys. Previous studies all agreed that the Lincang batholith was formed in the Triassic, and widely considered its formation to be the result of Paleo- Tethyan orogeny （Dong et al., 2013）. However, the early Ordovician syenogranite has been discovered recently for the first tirne in the Lincang batholith, and its petrologic and geochronological results are presented here to provide evidence in support of its association with Proto-Tethyan orogeny. This new discovery has important significance lbr understanding the geodynamic history of the Lincang batholith and tectonic evolution of the Sanjiang region.

BACKGROUND To date,the optimal timing for percutaneous transhepatic gallbladder drainage(PTGBD),particularly for patients who have missed the optimal window for emergency laparoscopic cholecystectomy(LC)(within 72 hours of symptom onset)has not been *** To study the effects of LC timing on outcomes of grade II/III acute cholecystitis(AC)in patients with delayed *** Data of patients diagnosed with Tokyo Guidelines 2018 grade II or III AC who underwent delayed PTGBD followed by LC at a single hospital between 2018 and 2022 were retrospectively *** to the interval between gallbladder drainage and cholecystectomy,the patients were divided into early and delayed LC *** including surgery time,postoperative complications and hospital stay,and patient satisfaction were analyzed and compared between the two groups using t-andχ^(2) *** There were no significant differences between the two groups in intraoperative blood loss,postoperative abdominal drainage tube placement time,pain index,or total disease duration(all P>0.05).Compared with those of the early LC group,the delayed group showed significant decreases in the length of procedure(surgery time),conversion rate to open surgery,degree of adhesions,surgical complications,postoperative hospital stay,and total treatment costs,and increased patient satisfaction despite a longer interval before PTGBD(all P<0.05).CONCLUSION For patients with grade II/III AC with delayed PTGBD,LC should be performed 2 weeks after PTGBD to decrease postoperative complications and hospital stays and improve patient satisfaction.

Introduction:This study aimed to evaluate the effectiveness and optimal course time of electro-acupuncture(EA)combined with conventional antipsychotics in the treatment of schizophrenia(SZ)and to provide a basis for its clinical ***:Relevant databases and authoritative websites were retrieved,and references were screened up to May *** quality evaluation and data extraction were carried out according to the requirements of Cochrane version 5.1.0,and meta-analysis was conducted using Rav Man5.4 ***:In total,16 randomized controlled trails comprising 1352 patients were ***-analysis revealed that the observation group showed greater improvements than the control group in total clinical efficacy(P0.05).In the 4-6 weeks following treatment,a significant improvement in all the four components of the PANSS scores was observed in the observation ***:The efficacy of EA combined with conventional antipsychotics for SZ is better than that of psychiatric drugs *** addition,the effect of a 4-to 6-week treatment course on each outcome index is more significant,and the efficacy is higher.

Two coordination polymers(CPs) [Zn(PTA)(DTP)(HO)]·(DMF)(CP-1) and [Zn(BTC)(DTP)]·(CHCN)·(HO)(CP-2) with one-and two-dimensional architectures were synthesized from Zn(Ⅱ) ion and different organic linkers like terephthalic acid(HPTA), benzene-1,3,5-tricarboxylic acid(HBTC), and 3,5-di(1,2,4-triazol-1-yl) pyridine(DTP). The fluorescent sensing experiments showed that the two CPs displayed effective, sensitive, and selective abilities towards Feand CrO. For sensing the pesticides, CP-1 outperforms in sensing of metamitron(MMT) and CP-2 is ultrasensitive towards imidacloprid(IMI). The possible mechanisms involved in the quenching of the fluorescence intensity include the inner filter effect(IFE) and the fluorescence resonance energy transfer(FRET) effect.
