Seasonal changes in gut microbiota of sea cucumber over natural aestivation cycle

Sea cucumber Apostichopus japonicus is an ideal model organism for marine invertebrate aestivation;it annually enters a“sleeping state”for nearly 3 months when ambient water temperature range is 25–30℃.The natural fasting is accompanied by remodeling the intestinal biota and function,which is a part of host biology and could affect the gut microbiota.We investigatesd the impact of annual aestivation on gut microbiota using high-throughput sequencing of 16S rRNA amplicons.Results reveal a notable alteration in the composition of gut bacteria during aestivation during which various indigenous families and genera that exhibit a preference for dietary glycans(e.g.,family Rhodobacteraceae and Flavobacteriaceae)decreased,while the taxa capable of utilizing substrates derived from the host(e.g.,genus Akkermansia and Prevotella)increased,and so did certain opportunistic pathogenic bacteria.Moreover,the investigation delved into the gut morphology and immunity response of A.japonicus and reveal that the intestine of A.japonicus undergoes substantial atrophy and degeneration during aestivation.However,there was an observed augmentation in the levels of acid and neutral mucin within mucous cells,as well as an enhanced immune defense response(as evidenced by increased gene expression of AjTLR3,LITAF,Ajp105,and LYZ).These results imply that the composition of sea cucumber gut microbiota differed between aestivation and active periods,which potentially affects the intestinal functions of the host and the symbiotic relationship between host and its microbiota over the activeaestivation cycle.

This paper introduces an intelligent computational approach for extracting salient objects fromimages and estimatingtheir distance information with PTZ (Pan-Tilt-Zoom) cameras. PTZ cameras have found wide applications innumerous public places, serving various purposes such as public securitymanagement, natural disastermonitoring,and crisis alarms, particularly with the rapid development of Artificial Intelligence and global infrastructuralprojects. In this paper, we combine Gauss optical principles with the PTZ camera’s capabilities of horizontal andpitch rotation, as well as optical zoom, to estimate the distance of the object.We present a novel monocular objectdistance estimation model based on the Focal Length-Target Pixel Size (FLTPS) relationship, achieving an accuracyrate of over 95% for objects within a 5 km range. The salient object extraction is achieved through a simplifiedconvolution kernel and the utilization of the object’s RGB features, which offer significantly faster computingspeeds compared to Convolutional Neural Networks (CNNs). Additionally, we introduce the dark channel beforethe fog removal algorithm, resulting in a 20 dB increase in image definition, which significantly benefits distanceestimation. Our system offers the advantages of stability and low device load, making it an asset for public securityaffairs and providing a reference point for future developments in surveillance hardware.

Exosomes,nanoscale extracellular vesicles(EVs)derived from the invagination of the endosomal membrane,are secreted by a majority of cell types.As carriers of DNA,mRNA,proteins,and microRNAs,exosomes are implicated in regulating biological activities under physiological and pathological conditions.Kidney-derived exosomes,which vary in origin and function,may either contribute to the pathogenesis of disease or represent a potential therapeutic resource.Membranous nephropathy(MN),an autoimmune kidney disease characterized by glomerular damage,is a predominant cause of nephrotic syndrome.Notably,MN,especially idiopathic membranous nephropathy(IMN),often results in end-stage renal disease(ESRD),affecting approximately 30%of patients and posing a considerable economic challenge to healthcare systems.Despite substantial research,therapeutic options remain ineffective at halting IMN progression,underscoring the urgent need for innovative strategies.Emerging evidence has implicated exosomes in IMN’s pathophysiology;Providing a fresh perspective for the discovery of novel biomarkers and therapeutic strategies.This review aims to scrutinize recent developments in exosome-related mechanisms in IMN and evaluate their potential as promising therapeutic targets and diagnostic biomarkers,with the hope of catalyzing further investigations into the utility of exosomes in MN,particularly IMN,ultimately contributing to improved patient outcomes in these challenging disease settings.

The performance degradation and even damage of the e-textiles caused by sweat,water,or submersion during all-weather health monitoring are the main reasons that e-textiles have not been commercialized and routinized so far.Herein,we developed an amphibious,high-performance,air-permeable,and comfortable all-textile triboelectric sensor for continuous and precise measurement of epidermal pulse waves during full-day activities.Based on the principle of preparing gas by acid-base neutralization reaction,a one-piece preparation process of amphibious conductive yarn(ACY)with densely porous structures is proposed.An innovative three-dimensional(3D)interlocking fabric knitted from ACYs(0.6 mm in diameter)and polytetrafluoroethylene yarns exhibit high sensitivity(0.433 V·kPa^(-1)),wide bandwidth(up to 10 Hz),and stability(>30,000 cycles).With these benefits,98.8%agreement was achieved between wrist pulse waves acquired by the sensor and a high-precision laser vibrometer.Furthermore,the polytetrafluoroethylene yarn with good compression resilience provides sufficient mechanical support for the contact separation of the ACYs.Meanwhile,the unique skeletonized design of the 3D interlocking structure can effectively relieve the water pressure on the sensor surface to obtain stable and accurate pulse waves(underwater depth of 5 cm).This achievement represents an important step in improving the practicality of e-textiles and early diagnosis of cardiovascular diseases.

Associations between Social Media Use and Sleep Quality in China:Exploring the Mediating Role of Social Media Addiction

Sleep quality is closely linked to people’s health,and during the COVID-19 pandemic,the sleep patterns of residents in China were notably poor.The lockdown in China led to an increase in social media use,prompting questions about its impact on sleep.Therefore,this study investigates the association between social media use and sleep quality among Chinese residents during the COVID-19 outbreak,highlighting the potential mediating role of social media addiction.Data were collected via questionnaires through a cross-sectional survey with 779 valid responses.Variance analysis was used to test for differences in social media use among different demographic variables.Bivariate correlation analysis was employed to explore the relationships between variables,while regression analysis investigated the correlations between various media factors and sleep quality.Additionally,Bootstrap sampling was utilized to analyze the potential mediating influence of social media addiction in the relationship between social media use and sleep.The study's findings reveal a significant correlation between social media use,particularly before bedtime,and sleep quality(p<0.01),with pre-sleep activity notably linked to poorer overall sleep scores(β=0.141,p=0.004).Although the daily use of social media did not directly impact most individuals’sleep quality,specific platforms like news apps,short video apps,dating apps,and content community platforms were associated with higher levels of social media addiction,subsequently negatively affecting sleep quality.Specifically,the use of news apps(B=0.068,95%CI[0.000,0.019]),short video apps(B=0.112,95%CI[0.001,0.031]),dating apps(B=0.147,95%CI[0.000,0.028]),and content community platforms(B=0.106,95%CI[0.001,0.028])was found to increase the risk of social media addiction,subsequently leading to adverse effects on sleep quality.The study underscores a notable link between social media use and sleep quality,suggesting that mindful social media habits,particularly before bedtime,and reducing addiction

Bitterness,one of the 5“basic tastes”,is usually undesired by humans.However,abundant literature reported that bitter fruits and vegetables have beneficial health effects due to their bitter contributors.This review provided an updated overview of the main bitter contributors of typical bitter fruits and vegetables and their health benefits.The main bitter contributors,including phenolics,terpenoids,alkaloids,amino acids,nucleosides and purines,were summarized.The bioactivities and wide range of beneficial effects of them on anti-cancers,anti-inflammations,anti-microbes,neuroprotection,inhibiting chronic and acute injury in organs,as well as regulating behavior performance and metabolism were reported.Furthermore,not only did the bitter taste receptors(taste receptor type 2 family,T2Rs)show taste effects,but extra-oral T2Rs could also be activated by binding with bitter components,regulating physiological activities via modulating hormone secretion,immunity,metabolism,and cell proliferation.This review provided a new perspective on exploring and explaining the nutrition of bitter foods,revealing the relationship between the functions of bitter contributors from food and T2Rs.Future trends may focus on revealing the possibility of T2Rs being targets for the treatment of diseases,exploring the mechanism of T2Rs mediating the bioactivities,and making bitter foods more acceptable without getting rid of bitter contributors.

Triosephosphate isomerase(TPI)is an enzyme that functions in plant energy production,accumulation,and conversion.To understand its function in maize,we characterized a maize TPI mutant,zmtpi4.In comparison to the wild type,zmtpi4 mutants showed altered ear development,reduced kernel weight and starch content,modified starch granule morphology,and altered amylose and amylopectin content.Protein,ATP,and pyruvate contents were reduced,indicating ZmTPI4 was involved in glycolysis.Although subcellular localization confirmed ZmTPI4 as a cytosolic rather than a plastid isoform of TPI,the zmtpi4 mutant showed reduced leaf size and chlorophyll content.Overexpression of ZmTPI4 in Arabidopsis led to enlarged leaves and increased seed weight,suggesting a positive regulatory role of ZmTPI4 in kernel weight and starch content.We conclude that ZmTPI4 functions in maize kernel development,starch synthesis,glycolysis,and photosynthesis.

Thyroid-stimulating hormone(TSH)is important for the thyroid gland,development,growth,and metabolism.Defects in TSH production or the thyrotrope cells within the pituitary gland cause congenital hypothyroidism(CH),resulting in growth retardation and neurocognitive impairment.While human TSH is known to display rhythmicity,the molecular mechanisms underlying the circadian regulation of TSH and the effects of TSH-thyroid hormone(TH)signaling on the circadian clock remain elusive.Here we show that TSH,thyroxine(T4),triiodothyronine(T3),and tshba display rhythmicity in both larval and adult zebrafish and tshba is regulated directly by the circadian clock via both E′-box and D-box.Zebrafish tshba^(−/−)mutants manifest congenital hypothyroidism,with the characteristics of low levels of T_(4)and T_(3)and growth retardation.Loss or overexpression of tshba alters the rhythmicity of locomotor activities and expression of core circadian clock genes and hypothalamic-pituitary-thyroid(HPT)axis-related genes.Furthermore,TSH-TH signaling regulates clock2/npas2 via the thyroid response element(TRE)in its promoter,and transcriptome analysis reveals extensive functions of Tshba in zebrafish.Together,our results demonstrate that zebrafish tshba is a direct target of the circadian clock and in turn plays critical roles in circadian regulation along with other functions.

Methanol to olefins(MTO) as an important reaction in C1 chemistry can effectively transform non-petroleum carbon resources into value-added chemicals. Zeolites have been widely used as MTO catalysts. However, they usually suffer from a rapid deactivation due to bulky coke species production, and thus require continuous regenerations in industrial application. The key to design and develop highly stable zeolite catalysts for MTO process is to unravel the deactivation mechanism and clarify the structure–performance relationship of catalysts. Here, in this mini-review, we investigate the critical intermediate species inducing zeolite deactivation and analyze the formation and evolution pathways of polycyclic aromatic hydrocarbons(PAHs)that are the precursors of carbonaceous deposits. In addition, some methods to alleviate the coking mainly including acid regulation, morphology modification and process optimization, are also summarized.
