Evaluation of extraction protocols for anti-diabetic phytochemical substances from medicinal plants

AIm To examine the efficacy of three extraction techniques: Soxhlet-extraction(SE), cold-maceration(Cm) and microwave-assisted-extraction(mAE) using 80% methanol as solvent. mETHODS The study was performed on each of 50 g of Vernonia amygdalina(VA) and Occimum gratissimum(OG) leaves respectively. The percentage yield, duration of extraction, volume of solvent used, qualitative and quantitative phytoconstituents present was compared. The biological activities(hypoglycemic effect) were investigated using albino wistar rat model of diabetes mellitus(n = 36) with a combined dose(1:1) of the two plants leaf extracts(250 mg/kg b.w.) from the three methods. Theextracts were administered orally, once daily for 21 *** In this report, the percentage VA extract yield from mAE was highest(20.9% ± 1.05%) within 39 min using 250 m L of solvent, when compared to the Cm(14.35% ± 0.28%) within 4320 min using 900 m L of solvent and SE(15.75% ± 0.71%) within 265 min using 500 m L of solvent. The percentage differences in OG extract yield between: mAE vs SE was 41.05%; mAE vs Cm was 46.81% and SE vs Cm was 9.77%. The qualitative chemical analysis of the two plants showed no difference in the various phytoconstituents tested, but differs quantitatively in the amount of the individual phytoconstituents, as mAE had significantly high yield(P > 0.05) on phenolics, saponins and tannins. SE technique gave significantly high yield(P > 0.05) on alkaloid, while Cm gave significant high yield on flavonoids. The extracts from Cm exhibited a significantly(P > 0.05) better hypoglycemic activity within the first 14-d of treatment(43.3% ± 3.62%) when compared to mAE(36.5% ± 0.08%) and SE methods(33.3% ± 1.60%). However, the percentage hypoglycemic activity, 21 d post-treatment with 250 mg/kg b.w. extract from mAE was 72.6% ± 1.03% and it was more comparable to 10 mg/kg b.w. glibenclamide treated group(75.0% ± 0.73%), unlike the SE(69.5% ± 0.71%) and Cm(69.1% ± 1.03%). From pollutant to solution of wastewater pollution: Synthesis of activated carbon from textile sludge for dye adsorption

Synthesis of activated carbon from spent tea leaves for aspirin removal

Adsorption capacity of activated carbon prepared from spent tea leaves （STL-AC） for the removal of aspirin from aqueous solution was investigated in this study. Preliminary studies have shown that treatment with phosphoric acid （H3PO4） increased removal efficiency of STL-AC. Characterizations on STL-AC revealed excellent textural properties （1200 m2.g-1, 51% mesoporosity）, as well as distinctive surface chemistry （1.08 mmol.g-1 and 0.54 mmol.g-1 for acidic and basic oxygenated groups, pHpzc = 2.02）. maximum removal efficiency of aspirin observed was 94.28% after 60 rain when the initial concentration was 100 mg.L-1, 0.5 g of adsorbent used, pH 3 and at a temperature of 30 ℃. The adsorption data were well fitted to the Freundlich isotherm model and obeyed the pseudo-second order kinetics model. The adsorption of aspirin onto STL-AC was exothermic in nature （△H = - 13.808 ***-1） and had a negative entropy change, △S （-41.444 ***-1）. A negative Gibbs free energy, △G was obtained indicating feasibility and spontaneity of the adsorption process. The adsorp- tion capacity of △C-STL （178.57 mg.g-1） is considerably high compared to most adsorbents synthesized from various sources, due to the well-defined textural properties coupled with surface chemistry of STL-AC which fa- vors aspirin adsorption. The results demonstrate the potential of STL-AC as aspirin adsorbent.

Purification and characterization of <i>Aspergillus parasiticus</i>cytosine deaminase for possible deployment in suicide gene therapy

Cytosine Deaminase (CD) from Aspergillus parasiticus was purified and characterized. Time course for maximal CD production (50 μmol/min/mg) was at 72 hrs. The enzyme was purified 387.73 folds with an overall yield of 13%. The CD had pH optimum of 7.2, a temperature optimum of 40℃ - 45℃, activation of energy (Ea) of 8.4 KJ/mol and a t1/2 of 1.10 hr. A. parasiticus CD stoichiometrically deaminated Cyto-sine and 5-fluorocytosine (5-FC) with the Km values of 0.19mm and 0.30 mm respectively. Studies on the effect of pH on Km and Vmax gave pKa1 of 5.8 and pKa2 of 6.3 with enthalpy of ionization of 43.01 KJ/mole suggesting histidine in the active site. The enzyme was strongly inhibited by Ca2+, Co2+, Zn2+ and Hg2+ and completely inhibited by Cu2+ and Fe2+ at 50 mm. Therefore, A. parasiticus CD can be compared with CDs from other sources that are used in suicide gene therapy.

In Vitro Regeneration of Commercial Sugarcane （Saccharum spp.） Cultivars in Nigeria

The effect of different concentrations of 6-benzylaminopurine （BA） with or without 0.2 mg/L NAA on in vitro regeneration of sugarcane （Saccharum spp.） cultivars SP726180, B47419, m1176/77 and m2119/88 were evaluated. Leaf base explants were cultured on murashige and Skoog （mS） basal medium supplemented with 3.0 mg/L 2,4-dichlorophenoxyacetic acid （2,4-D） for 4 weeks. Thereafter, induction of somatic embryogenesis was observed following the transfer of resulting calli to 2,4-D-free medium for another 4 weeks. Regeneration was achieved by transfer of the embryogenic calli to regeneration media fortified with different concentrations of BA ＋ tt-naphthylacetic acid （NAA）. The number, length and vigor of shoots produced in all the genotypes were highest on media supplemented with 1.0 and 1.5 mg/L BA with and without 0.2 mg/L NAA. Among the genotypes tested, B47419 and m1176/77 recorded highest number of shoots, while maximum shoot length and crop vigor was obtained with m1176/77. Induction of callus with 3.0 mg/L 2,4-D and its subsequent incubation on 2,4-D-free media, followed by regeneration on media supplemented with 1.0 or 1.5 mg/L BA with 0.2 mg/L NAA was found to be efficient for in vitro regeneration of the sugarcane genotypes used in this study. This protocol could be applied for micropropagation of other elite genotypes.

media Appraisal for Somatic Embryogenesis of Elite Inbred Lines of maize

Several protocols have been published for somatic embryogenesis in maize. It is essential to compare different protocols on selected germplasm at initial stages of tissue culture to identify a protocol that would yield optimum results. The ultimate goal is to use in vitro system for selecting and generating stress tolerant maize germplasm. Immature embryos of the elite inbred lines of maize （87014 ×Z28-11 and POP43SRS5-57） were cultured on three different semi-solid media （0.5% agar） and the effectiveness of the media for somatic embryogenesis assessed over a period two weeks of sub-culturing. A significant difference was observed among the media in proportion of potentially embryogenic calli and callus cluster size at 1 and 2 weeks after culturing. N6 medium supplemented with casaminoacid （100 mg/L）, 2, 4-dichlorophenoxy acetic acid （1 mg/L）, L-proline （25 mm） and sucrose （2%）, and murashige and Skoog （mS） medium supplemented with 2, 4-D （2 mg/L） and sucrose （3%） gave higher proportion of potentially embryogenic calli and callus size than mS medium supplemented with casaminoacids （100 mg/L）, 2, 4-D （2 mg/L）, abscissic acid （3.3μm）, and silver nitrate （195 μm） and sucrose （3%）. Optimum Protocol for Shoot Formation in karandafi Red Sorghum (<i>Sorghum bicolor</i>(L.) moench) through Somatic Embryogenesis Using mature Embryo

In this study, an optimum protocol for shoot formation through somatic embryogenesis using mature embryo explants was developed. Calli were initiated on murashige and Skoog (mS) media supplemented with varying concentration of 2,4-Dichlorophenoxyl acetic acid (2,4-D) ranging from 1.5 mg/l-4.0 mg/l alone or in combination with 0.5 mg/l Kinetin (KN). Significance difference (p 0.05) was observed among the different concentrations of hormone used for callus induction. The highest percentage callus formation was obtained from the media fortified with 4 mg/l 2, 4-D for mature embryo obtained from imbibed seed while for the preconditioned mature embryo, the media supplemented with 2 m/l 2,4-D + 0.5 mg/l kinetin (KN) recorded more percentage callus formation compared to what was obtained from the media supplemented with 2.5 mg/l2,4-D + 0.5 mg/l. more percentage shoot formation was obtained from the media supplemented with 1 mg/l 6-benzylaminopurine (BA). Average number of shoot per callus was also more in the media fortified with 1 mg/l BA (2.25) but this was not significantly different from what was obtained from the media fortified with 2 mg/l BA+ 0.1 mg/l 2,4-D at 5% level of significance.

以受镉(Cd)污染的棕壤为供试土壤,采用盆栽试验的方法,研究牛粪与秸秆配施对玉米各部位Cd吸收、转运及Cd在各部位亚细胞分布的影响。试验设6个处理,分别为对照处理(CK)、单施秸秆(S)、单施牛粪(m)、牛粪与秸秆含氮量比1∶1(Sm1)、牛粪与秸秆含氮量比3∶1(Sm3)、牛粪与秸秆含氮量比5∶1(Sm5)。结果表明,与对照相比,Sm1处理根部吸收Cd的含量下降了46.2%;Sm1和Sm3处理玉米叶部Cd吸收量与对照相比分别下降了69.8%和61.0%,玉米茎部Cd含量与对照相比分别降低了62.6%和50.4%,玉米籽粒Cd吸收量与对照处理相比分别降低了80.0%和75.0%。与对照相比,Sm1、Sm3和Sm5处理Cd由根部向茎叶部位的转运系数分别降低了0.37、0.21和0.17;Cd由茎叶向籽粒的转运系数分别降低了0.014、0.012和0.010。所有处理玉米各部位亚细胞组分的Cd含量均呈现为细胞壁>细胞器>可溶部分;在Sm1处理下,Cd在根、叶、茎和籽粒四个部位细胞壁中比例与对照相比提高最多,分别提高了16.5%、10.5%、10.7%和19.3%。因此,适宜的牛粪与秸秆配比能有效降低玉米各部位对Cd的吸收和转运,提高Cd在玉米各部位细胞壁中的分布比例,牛粪与秸秆含氮量比1∶1配施有利于Cd污染地区玉米的安全生产。
