WSB1 regulates c-Myc expression through β-catenin signaling and forms a feedforward circuit

The dysregulation of transcription factors is widely associated with tumorigenesis.As the most well-defined transcription factor in multiple types of cancer,c-Myc can transform cells by transactivating various downstream genes.Given that there is no effective way to directly inhibit c-Myc,c-Myc targeting strategies hold great potential for cancer therapy.In this study,we found that WSB1,which has a highly positive correlation with c-Myc in 10 cancer cell lines and clinical samples,is a direct target gene of c-Myc,and can positively regulate c-Myc expression,which forms a feedforward circuit promoting cancer development.RNA sequencing results from Bel-7402 cells confirmed that WSB1 promoted cMyc expression through theβ-catenin pathway.Mechanistically,WSB1 affectedβ-catenin destruction complex-PPP2CA assembly and E3 ubiquitin ligase adaptorβ-TRCP recruitment,which inhibited the ubiquitination ofβ-catenin and transactivated c-Myc.Of interest,the effect of WSB1 on c-Myc was independent of its E3 ligase activity.Moreover,overexpressing WSB1 in the Bel-7402 xenograft model could further strengthen the tumor-driven effect of c-Myc overexpression.Thus,our findings revealed a novel mechanism involved in tumorigenesis in which the WSB1/c-Myc feedforward circuit played an essential role,highlighting a potential c-Myc intervention strategy in cancer treatment.

Landforms are an important element of natural geographical environment,and textures are the research basis for the spatial differentiation,evolution features,and analysis rules of the landform.Using the regional difference of texture to describe the spatial distribution pattern of macro landform features is helpful to the landform classification and identification.Digital elevation model(DEM)image texture,which gives full expression to texture difference,is key data source to reflect the surface features and landform classification.Following the texture analysis,landform features analysis is assistant to different landforms classification,even in landform boundary.With the increasing accuracy requirement of landform information acquisition in geomorphic thematic mapping,hierarchical landform classification has become the focus and difficulty in research.Recently,the pattern recognition represented by Convolutional Neural Network has made great achievements in landform research,whose multichannel feature fusion structure satisfies the network structure of different landform classification.In this paper,DEM image texture was taken as the data source,and gray level co-occurrence matrix was applied to extract texture measures.Owing to the similarity of similar landform and the difference of different landform in a certain scale,a comprehensive texture factor reflecting landform features was proposed,and the spatial distribution pattern of landform features was systematically analyzed.On this basis,the coupling relationship between texture and landform type was explored.Thus,the deep learning method of Convolutional Neural Network is used to train the texture features,and the second-class landform classification is carried out through softmax.The classification results in small relief and mid-relief low mountains,overall accuracy are 84.35%and 69.95%respectively,while kappa coefficient are 0.72 and 0.40 respectively,were compared to that of traditional unsupervised landform classification results,and the superiority o

In 2006,Takahashi and Yamanaka first created induced pluripotent stem cells from mouse fibroblasts via the retroviral introduction of genes encoding the transcription factors Oct3/4,Sox2,Klf44,and c-Myc.Since then,the future clinical application of somatic cell reprogramming technology has become an attractive research topic in the field of regenerative medicine.Of note,considerable interest has been placed in circumventing ethical issues linked to embryonic stem cell research.However,tumorigenicity,immunogenicity,and heterogeneity may hamper attempts to deploy this technology therapeutically.This review highlights the progress aimed at reducing induced pluripotent stem cells tumorigenicity risk and howto assess the safety of induced pluripotent stem cells cell therapy products.

Scale is the basic attribute for expressing anddescribing spatial entity and phenomena. It offerstheoretical significance in the study of gully structureinformation, variable characteristics of watershed mor-phology, and development evolution at different scales.This research selected five different areas in China＇s LoessPlateau as the experimental region and used DEM data atdifferent scales as the experimental data. First, the changerule of the characteristic parameters of the data at differentscales was analyzed. The watershed structure informationdid not change along with a change in the data scale. Thiscondition was proven by selecting indices of gullybifurcation ratio and fractal dimension as characteristicparameters of watershed structure information. Then, thechange rule of the characteristic parameters of gullystructure with different analysis scales was analyzed bysetting the scale sequence of analysis at the extractiongully. The gully structure of the watershed changed withvariations in the analysis scale, and the change rule wasobvious when the gully level changed. Finally, the changerule of the characteristic parameters of the gully structure atdifferent areas was analyzed. The gully fractal dimensionshowed a significant numerical difference in differentareas, whereas the variation of the gully branch ratio wassmall. The change rule indicated that the developmentdegree of the gully obviously varied in different regions,but the morphological structure was basically similar.

Incidence,prevalence,and causes of spinal injuries in China,1990-2019:Findings from the Global Burden of Disease Study 2019

Background:Spinal injuries are an urgent public health priority;nevertheless,no China-wide studies of these injuries exist.This study measured the incidence,prevalence,causes,regional distribution,and annual trends of spinal injuries in China from 1990 to 2019.Methods:We used data from the Global Burden of Diseases,Injuries,and Risk Factors Study 2019 to estimate the incidence and prevalence of spinal injuries in China.The data of 33 provincial-level administrative regions(excluding Taiwan,China)provided by the National Center for Chronic and Noncommunicable Disease Control and Prevention,Chinese Center for Disease Control and Prevention(CDC)were use to systematically analyze the provincial etiology,geographical distribution,and annual trends of spinal injuries.The Bayesian meta-regression tool DisMod-MR 2.1 was used to ensure the consistency among incidence,prevalence,and mortality rates in each case.Results:From 1990 to 2019,the number of living patients with spinal injuries in China increased by 138.32%,from 2.14 million to 5.10 million,while the corresponding age-standardized prevalence increased from 0.20%(95%uncertainty interval[UI]:0.18-0.21%)to 0.27%(95%UI:0.26-0.29%).The incidence of spinal injuries in China increased by 89.91%(95%UI:72.39-107.66%),and the prevalence increased by 98.20%(95%UI:89.56-106.82%),both the most significant increases among the G20 countries;71.00%of the increase could be explained by age-specific prevalence.In 2019,the incidence was 16.47(95%UI:12.08-22.00,per 100,000 population),and the prevalence was 358.30(95%UI:333.96-386.62,per 100,000 population).Based on the data of 33 provincial-level administrative regions provided by CDC,age-standardized incidence and prevalence were both highest in developed provinces in Eastern China.The primary causes were falls and road injuries;however,the prevalence and specific causes differed across provinces.Conclusions:In China,the overall disease burden of spinal injuries increased significantly during the past three decades but varied consi

Frequent periods of drought conditions are known to limit plant performance,primary production,and ecosystem stability in arid and semi-arid desert steppe environments.Plants often avoid competition by shifting their water use seasonally,which affects the water-use patterns of dominant species as well as the composition and structure of plant communities.However,the water-use strategies of dominant herbaceous species,which grow under natural field conditions in the desert steppe region of Ningxia Hui Autonomous Region,China,are poorly known.Here,we explored the possible sources of water uptake and water-use efficiency(WUE)of three dominant herbaceous plant species(Stipa breviflora,Agropyron mongolicum,and Glycyrrhiza uralensis)in a native desert steppe in the semi-arid area of Ningxia through an analysis of multiple parameters,including(1)the stable isotopic oxygen and hydrogen(δ18O andδ2H)compositions of precipitation,soil water,and stem water,(2)the carbon isotope(13C)composition of leaves,and(3)the soil water contents,based on field sampling across varying water conditions from June to September,2017.Frequent small precipitation events replenished shallow soil water,whereas large events only percolated down to the deep soil layers.Changes in soil water availability affected the water-use patterns of plants.Generally,during light precipitation periods,the deep root system of G.uralensis accessed deeper(>80 cm)soil water,whereas S.breviflora and A.mongolicum,which only have shallow roots,primarily absorbed water from the shallow and middle soil layers.As precipitation increased,all three plant species primarily obtained water from the shallow soil layers.Variation in soil water uptake between the dry and wet seasons enabled plants to make better use of existing satoil water.In addition,theδ13C values of G.uralensis and S.breviflora were higher than those of A.mongolicum.Theδ13C values of the three plant species were significantly negatively correlated with soil water content.Therefore,G.uralensis and S.breviflor

The discovery and application of antibiotics in animal feeds have boomed the development of intensive animal husbandry in the last century,until the emergence of antibiotics-resistant bacteria.To alleviate the risks aroused by antibiotics-resistant bacteria,effective antibiotic substitutes are urgently needed to replace antibiotics.Essential oils(EOs)derived from plants are illustrated as the promising antibiotic substitutes used in animal feeds,as same as their current views for poultry and livestock industries in the future.It has been widely demonstrated that the phytochemicals in EOs show multiple biofunctionability and are less likely to induce resistance in bacteria.The beneficial effects of EOs feed supplementation on the intestinal inflammation,intestinal flora,immunity,digestion,and growth performances have been already extensively examined.However,the cost-effectiveness,odor,volatility,instability and bioavailability are the challenges in effectively utilizing EOs in animal intestines.Based on previous researches,and these challenges can partially be resolved by microencapsulation and nanotechnology are promising techniques to deal with these challenges.This article presents the feasibility and foundation of EOs application as antibiotic substitutes in animal feeds,and illustrates the mechanisms,functional performances and superiority of EOs compared with antibiotics.

The drag on non-spherical particles is an important basic parameter for multi-phase flows such as in biomass combustion, chemical blending, and mineral processing. Though there is much experimental research on such particles, there are few results for cuboids. This paper presents data for cuboids with a square base in static glycerin-water solutions of various volume concentrations. Complex motions were observed and characterized. A dimensionless expression is given for terminal velocity ut as a function of Archimedes number Ar which is used to develop an accurate correlation for friction factor CD. The accuracy of the correlation is 7.9% compared to experimental data in the literature. For both square plates and square rods, the terminal velocity per unit mass, ut/mp, was used to characterize the influence of narticle geometry on velocity, which was shown to be linear.

Texture and its analysis methods are crucial for image feature extraction and classification. Digital elevation model (DEM) is the most important data source of digital terrain analysis and landform classification, and considerable research values are gained from texture feature extraction and analysis from DEM data. In this research, on the basis of optimal texture feature extraction, the hilly area in Shandong, China, was selected as the study area, and DEM data with a resolution of 500 m were used as the experimental data for landform classification. First, second-order texture measures and texture image were extracted from DEM data by using a gray level cooccurrence matrix (GLCM). Second, the variation characteristics of each texture measure were analyzed, and the optimal feature parameters, such as direction, gray level, and texture window, were determined. Meanwhile, the texture feature value, combined with maximum information, was calculated, and the multiband texture image was obtained by resolving three optimal texture measure images. Finally, a support vector machine (SVM) method was adopted to classify landforms on the basis of the multiband texture image. Results indicated that the texture features of DEM data can be sufficiently represented and measured via the quantitative GLCM method. However, the feature parameters during the texture feature value calculation required further optimization. Based on the image texture from DEM data, efficient classification accuracy and ideal classification effect were achieved.
