Ultrasensitive and point-of-care detection of plasma phosphorylated tau in Alzheimer’s disease using colorimetric and surface-enhanced Raman scattering dual-readout lateral flow assay

Phosphorylation of tau at Ser(396,404)(p-tau^(396,404))is one of the earliest phosphorylation events,plasma p-tau^(396,404) level appears to be a potentially promising biomarker of Alzheimer’s disease(AD).The low abundance and easy degradation of p-tau in the plasma make the lateral flow assay(LFA)a suitable choice for point-of-care detection of plasma p-tau^(396,404) levels.Herein,based on our screening of a pair of p-tau^(396,404)-specific antibodies,we developed a colorimetric and surface-enhanced Raman scattering(SERS)dual-readout LFA for the rapid,highly sensitive,robust detection of plasma p-tau^(396,404) levels.This LFA realized a detection limit of 60 pg/mL by the naked eye or 3.8 pg/mL by SERS without cross-reacting with other tau species.More importantly,LFA rapidly and accurately differentiated AD patients from healthy controls,suggesting that it has the potential for clinical point-of-care application in AD diagnosis.This dual-readout LFA has the advantages of simple operation,rapid,ultra-sensitive detection,providing a new way for early AD diagnosis and intervention,especially in primary and community AD screening.

Interdecadal Variations of ENSO Impacts over the Indo-Northwest Pacific Region and the Related Mechanisms

Owing to limited observations,it remains unknown whether the impact of El Niño-Southern Oscillation(ENSO)on the Indian Ocean-Northwest Pacific(IO-NWP)climate showed decadal changes in the early 20th century.Using multi-source reanalysis and hindcast datasets from the ECMWF and NOAA extending back to 1901,this study investigates interdecadal variations of the impact of ENSO on the IO-NWP climate from 1901 to 2009.It is found that the influence of ENSO on the IO-NWP climate shows“strong-weak-strong”interdecadal change during 1901-2009.This is characterized by much weaker Indian Ocean sea surface temperature(SST)warming and a weaker NWP subtropical anticyclone(NWPSA)in the following summer of El Ni?o during 1946-1967,compared with those in the other two periods(1901-1945 and 1968-2009).Analyses of the datasets indicate that the interdecadal variation is mainly associated with the change in ENSO amplitude.In contrast to the period of 1946-1967,a greater SST variance occurred in the central-eastern equatorial Pacific during 1901-1945 and 1968-2009.A stronger El Ni?o tends to generate more significant anticyclonic anomalies over the southeast Indian Ocean through teleconnection.The northwesterly anomalies to the south of the anticyclone weaken the southeast trade winds and warm the south Indian Ocean SST via wind-evaporation-SST feedback,and the positive south Indian Ocean SST anomalies trigger westward-propagating oceanic Rossby waves to induce stronger warming of the southwest Indian Ocean,leading to a significant asymmetric wind pattern across the equator in spring.The profound northeastward winds on the north side weaken the southwest monsoon,leading to a“second warming”over the north Indian Ocean in summer,which anchors the eastward-propagating warm Kelvin waves and results in a stronger NWPSA by inducing surface divergence and suppressing deep convection.

The frontier of microplastics and nanoplastics: Soil health and carbon neutrality

Microplastics and nanoplastics(MNPs)in soil have drawn increasing concerns about their potential threats to soil ecosystems due to their ubiquitous occurrence and persistence.The interactions of MNPs with soil components,microbial community,plants,and fauna determine their impacts on soil biogeochemical processes and food security.However,they are not largely explored.Further research is also needed to develop reliable and standardized techniques and methods to characterize the relevant MNPs interacting with different types of soil ecosystems and to deeply examine their impacts on soil health,food security,and climate changes.In addition,mitigation protocols and regulation guidelines need to be developed.New findings will provide scientific and technological support for the understanding and management of soil health and carbon neutrality as influenced by MNPs.

Ligand accommodation causes altered reactivity of silver clusters with iodomethane:superatomic stability of Ag_(9)I_(2)^(+)in mimicking XeF_(2)

Exploring metal cluster reactivity with alkyl halides enables to understand the related chemical mechanism of metal surfaces in terms of active sites.Here we report a study of Ag_(n)^(+)(n=1-27)clusters reacting with iodomethane by a flow tube apparatus in tandem with a customized triple quadrupole mass spectrometer.Strong even/odd alternation of the Ag_(n)^(+)is observed in their reactions with CH_(3)I,where silver clusters with even-number,Ag_(2n)^(+),find favorable products of Ag_(2n)I_(1,3)^(+)series,while the Ag_(2n−1)^(+)clusters form Ag_(2n−1)I_(2,4)^(+)products.Interestingly,Ag_(9)^(+)shows up with prominent mass abundance but allows for the formation of Ag_(9)I_(2)^(+),which finds an echo with the formation of Ag_(10)I_(3)^(+).We illustrate the enhanced stability of Ag_(9)I_(2)^(+)and Ag_(10)I_(3)^(+)by showing their significantly enlarged highest occupied molecular orbital(HOMO)-lowest unoccupied molecular orbital(LUMO)gaps and balanced charge distribution compared with the bare metal clusters,respectively.Also elucidated,is the superatomic nature of these bare and iodinated silver clusters,especially Ag_(9)I_(2)^(+)which mimics the rare-gas compound XeF_(2).This study expands a vivid example of special and general superatoms,and enriches the general knowledge on how a ligand stabilizes a metal cluster.

Cerebrospinal fluid and blood biomarkers in the diagnostic assays of Alzheimer's disease

The anti-amyloid-j(anti-Aβ)fibrils and soluble oligomers antibody aducanumab were approved to effectively slow down the progression of A lzheimer's disease(AD)at higher doses in 2019,reaffirming the therapeutic effects of targeting the core pathology of AD.A timely and accurate diagnosis in the prodromal or pre-dementia stage of AD is essential for patient recruitment,stratifcation,and monitoring of treat ment effects.A D core biomarkers amyloid-B(Aβ1-42),total tau(t-tau),and phosphorylated tau(p-tau)have been clinically validated to reflect AD-type pathological changes through cerebrospinal fuid(CSF)measurement or positron-emission to-mography(PET)and found to have high diagnostic performance for AD identification in the stage of mild cognitive impairment.The development of ultrasensitive immunoassay technology enables AD pathological proteins such as tau and neurofilament light(NFL)to be measured in blood samples.However,combined biomarker detection or targeting multiple biomarkers in immunoassays will increase detection sensitivity and specifcity and improve diagnostic accuracy..This review summarizes and analyzes the performance of current detection methods for early diagnosis of AD,and provides a concept of detection method based on multiple biomarkers instead of a single target,which may become a potential tool for early diagnosis of AD in the future.

Multiple-Sized Amphiphilic Janus Gold Nanoparticles by Ligand Exchange at Toluene/Water Interface

Amphiphilic Janus gold nanoparticles （GNPs） are prospected to encapsulate drug molecules in cancer therapy and to serve as heterogeneous catalysts at oil/water interfaces, where Janus GNPs with differ- ent sizes are required. In this work, multiple-sized precursor GNPs were synthesized by seeded growth method protected with tris（hydroxymethyl）phosphine oxide （THPO） ligand molecule, and a ligand ex- change reaction with triphenylphosphine （PPh3） at the toluene/water interface was employed to prepare amphiphilic Janus GNPs. UV-vis and transmission electron microscopy （TEM） analyses indicate that the as-prepared GNPs are nanocrystals with average diameters of 2.3 nm, 9.5 nm, 16.1 nm and 18.8 nm, re- spectively. Contact angle, Raman and X-ray photonic spectroscopy （XPS） analyses reveal that the self- assembled GNP films exhibit hydrophilic on one side and hydrophobic on another, owing to the adsorption of hydrophilic ligands （THPO and THP） and a similar amount of hydrophobic ligands （PPh3 and PPh30）. Angle-resolved XPS analysis further demonstrates that the individual GNPs actually possess hydro- philic and hydmphobic compartments on the surface, which regularly packed by supramolecular interactions at toluene/water interface to form the self-assembled GNP films.

How ligand coordination and superatomic-states accommodate the structure and property of a metal cluster:Cu_(4)(dppy)_(4)Cl_(2)vs.Cu_(21)(dppy)_(10)with altered photoluminescence

We have synthesized two copper nanoclusters(NCs)with a protection of the same ligand diphenylphosphino-2-pyridine(C_(17)H_(14)NP,dppy for short),formulated as Cu_(4)(dppy)_(4)Cl_(2)and Cu21(dppy)10,respectively.The former one bears a distorted tetrahedron Cu4 core with its six edges fully protected by chlorine and dppy ligands,while the latter presents a symmetric Cu_(21)core on which ten dppy molecules function as monolayer protection via well-organized monodentate or bidentate coordination.Interestingly,the Cu_(4)(dppy)_(4)Cl_(2)cluster exhibits a strong yellow emission at∼577 nm,while Cu_(21)(dppy)_(10)displays dual emissions in purple(∼368 nm)and green(∼516 nm)regions respectively.In combination with TD-DFT calculations,we demonstrate the origin of altered emissions and unique stability of the two copper nanoclusters pertaining to the ligand coordination and metallic superatomic states.

Chlorine-passivated superatom Al37clusters for nonlinear optics

Utilizing a facile top-down synthetic procedure, here we report the finding of a chlorine-passivated Al_(37) superatom cluster. It is demonstrated that the presence of electrophilic groups, severing as protecting ligands, alters the valence electron count of the metallic core and stabilize the as-prepared aluminum clusters especially when even-numbered chlorine atoms are located at equilibrium positions. Following the discussion regarding their reasonable stabilities, we illustrate the feasible reaction pathways in forming such chlorine-passivated Al_(37) superatom clusters which bear delocalized superatomic orbitals with five valence 3P^5 electrons shifting to the chlorine ligands indicative of a closed electron shell 2F^(14) of the metal core. The successful synthesis of such chlorine-protected aluminum clusters evidences the compatibility of general theory of cluster chemistry in both gas phase and wet chemistry. Such simple-ligand-protected aluminum clusters exhibit reverse-saturated-absorption(RSA) nonlinear optical property pertaining to electronic transitions within the discrete energy states of cluster materials.

Coronavirus disease 2019(COVID-19)is a newly emerged infectious disease caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2).The number of COVID-19 cases is continuously increasing and no effective drugs or vaccines are currently available.Accurate and efficient diagnostic testing methods are desperately needed for the detection of SARS-CoV-2 and antiviral antibodies in infected individuals.Various assay techniques,including nucleic acid tests[eg,polymerase chain reaction(PCR),reverse transcription-PCR,real-time loop-mediated isothermal amplification,and CRISPR-Cas-based detection],serological tests[eg,immunoglobulin(Ig)A,IgM/G],imaging tests(eg,computed tomography and positron-emission tomography),and nanoparticle-based detections have been reported for COVID-19 diagnosis.This review aims to present the current diagnostic tools for SARS-CoV-2 and their performance characteristics to inform the appropriate selection of diagnostic and surveillance technologies at optimal testing times.We also describe the advantages of detection using combined nucleic acid and imaging tests,or serological testing and point-of-care diagnostics.Developing reliable protein biomarkers targeting the conserved proteins of SARS-CoV-2 rather than IgA,IgM,or IgG would be useful to manage SARS-CoV-2.

Small gold clusters catalyzing oxidant-free dehydrogenation of glycerol initiated by methene hydrogen atom transfer

Developing oxygen-free methodology for the conversion of alcohols to carbonyls is essentially important because it suppresses the over-oxidation of alcohols to carboxylic acids and enables the production of energetic hydrogen.Here we report a finding of feasible oxidant-free dehydrogenation of glycerol over chemically-pure Au clusters synthesized by a green chemistry method named as laser ablation in liquid(LAL).As results,glycerol is dehydrogenated to form glyceraldehyde which undergoes subsequent dehydrogenation to hydroxymethyl glyoxal.For this,reaction dynamics calculations find interesting dehydrogenation reaction pathways with a low-energy barrier of transition state initiated by hydrogen atom transfer from methene,which differs from the general reaction mechanism based on hydroxyl.Furthermore,it is interesting that the presence of additionalààOH group molecules especially H2O can effectively lower the energy barrier in the activation of the OààH and CààH bonds in glycerol.This principle is also applicable to the oxidant-free dehydrogenation of methanol and ethanol,helping to fully understand the catalytic mechanism of alcohols conversion chemistry.
