From an evolutionary point of view, reproduction timing is an important adaptation which enables the transfer of genetic properties, thus enabling species continuation. Rodents inhabiting arid environments need reliable cues for triggering their reproduction. Results of previous studies showed that increased dietary salinity plays an important role as an ultimate regulator for desert adapted rodents＇ reproductive system. The authors aimed discovering pathways by which high salinity can affect the reproductive system and metabolic status of desert adapted common spiny mice, Acomys cahirinus. Mice were challenged with osmotic stress, water source salinity increased gradually from 0.9% - 5% NaCI under short days （SD） and long days （LD）. The authors assessed leptin and free fatty acid （FFA） levels using ELISA while, SYBR green technology was used for relative receptor expression （RQ） of target genes. Results revealed that serum levels of the hormone leptin were significantly （P 〈 0.05） reduced in salinity treated （ST） mice. Levels of FFA were significantly （P 〈 0.05） increased in LD- and SD-ST-males. In ST-SD females a significant increase （P 〈 0.05） in expression levels of leptin （Ob-Rt） mRNA receptor gene, in ovaries was noted. Aldosteron （Nr3c2） and vasopressin （AVP） mRNA receptor expression genes levels were significantly （P 〈 0.05） increased in both LD- and SD- ST- males.

Human skeletal remains were found in a sandy area in the western part of Israel. Owing to their scattered nature, multiple analyses were required to confirm that they belonged to the same individual. The analyses were completed by estimating the age and sex of each bone, if possible, as well as its probable articulation and relative site location. The probable articulation was found to be largely insufficient here owing to low and moderate confidence in the state of the bones that were found and the significant signs of postmortem scavenging. Sex estimation analysis of different aspects of the bones revealed various mixed or “mosaic” results. This case exemplifies a usage of numerous analytical methods in order to relate unduplicated scattered remains, found at a long distance, to a single individual.

Semi Operational Prediction of the Dead Sea Evaporation—A Synoptic Systems Approach

The predictability of pan evaporation and air temperature in the southern part of the Dead-Sea region (Sdom) was investigated according to two approaches, prediction by mesoscale models and with the aid of synoptic classification. First, the predicted temperature, wind speed and relative humidity that directly affect the evaporation are obtained from the WRF mesoscale model predictions. Predictions according to multilinear regression equations and a Penman-Monteith approach were also validated against observations in Sdom. The WRF model predicts the temperature reasonably well. However, the wind speed and relative humidity predictions were found to be very poor. The unique approach in this paper is employing a semi-objective synoptic systems classification according to the global GFS model. Relationships were defined between the 19 Eastern Mediterranean’s (EM) synoptic systems and the Sdom evaporation, temperature, wind speed and relative humidity. A monthly evaluation was performed for each of the systems and the semi-objective prediction was verified by the semi-objective classification. Since some synoptic systems affect the evaporation and temperature similarly, the 19 synoptic systems were grouped into seven clusters, each containing systems with similar evaporation and temperature records. This method has yielded a significant improvement in the daily prediction of evaporation and temperature. Semi-objective definitions for the synoptic systems were performed for the ranges of 12 - 132 hours. The synoptic system approach succeeded in the prediction of the evaporation and temperature changes in Sdom for a few days in advance. The predictability skill for the 12 hour forecast achieved about 80% of success, dropping to 70% at 36 hours. An immunochemical sol-gel-based immunoaffinity purification (IAP) method for purification and detection of the progestin drug medroxyprogesterone acetate (MPA) was developed. A polyclonal antibody (Ab) for MPA was generated, and two competitive (indirect and direct) sensitive enzyme-linked immunosorbent assays (ELISAs) for its detection were developed and implemented to determine the recovery and efficiency of the sol-gel based IAP method. The detection limits of the assays were 1.4 ± 0.2 ng·mL-1 (n = 4) and 4.0 ± 0.4 ng·mL-1 (n = 25) for the indirect and direct ELISAs, respectively. The Abs did not exhibit cross-reactivity with any other progestin or steroid hormone, with the exception of megestrol acetate, with which the Ab exhibited 76% cross-reactivity. The sol-gel IAP method successfully eliminated serum interference to a degree that enabled ELISA analysis of spiked serum samples. This method was also found fully compatible with subsequent chemical analytical methods, such as liquid chromatography followed by mass spectrometry (LC-MS/MS). The approaches developed in this study form a basis for analysis of MPA in biological samples and may be further used to study population exposure to MPA and to monitor MPA contamination in water samples.

Liquid biopsy is a highly promising method for non-invasive detection of tumor-associated nucleic acid fragments in body fluids but is challenged by the low abundance of nucleic acids of clinical interest and their sequence homology with the vast background of nucleic acids from healthy ***,programmable endonucleases such as clustered regularly interspaced short palindromic repeats(CRISPR)associated protein(Cas)and prokaryotic Argonautes have been successfully used to remove background nucleic acids and enrich mutant allele fractions,enabling their detection with deep next generation sequencing(NGS).However,the enrichment level achievable with these assays is limited by futile binding events and off-target *** overcome these shortcomings,we conceived a new assay(Programmable Enzyme-Assisted Selective Exponential Amplification,PASEA)that combines the cleavage of wild type alleles with concurrent polymerase *** PASEA increases the numbers of both wild type and mutant alleles,the numbers of mutant alleles increase at much greater rates,allowing PASEA to achieve an unprecedented level of selective enrichment of targeted *** combining CRISPR-Cas9 based cleavage with recombinase polymerase amplification,we converted samples with0.01%somatic mutant allele fractions(MAFs)to products with 70%MAFs in a single step within 20 min,enabling inexpensive,rapid genotyping with such as Sanger ***,PASEA's extraordinary efficiency facilitates sensitive real-time detection of somatic mutant alleles at the point of care with custom designed Exo-RPA ***-time PASEA'performance was proved equivalent to clinical amplification refractory mutation system(ARMS)-PCR and NGS when testing over hundred cancer patients'*** strategy has the potential to reduce the cost and time of cancer screening and genotyping,and to enable targeted therapies in resource-limited settings.
