Glioma-derived ANXA1 suppresses the immune response to TLR3 ligands by promoting an anti-inflammatory tumor microenvironment

A highly immunosuppressive tumor microenvironment(TME)and the presence of the blood‒brain barrier are the two major obstacles to eliciting an effective immune response in patients with high-grade glioma(HGG).Here,we tried to enhance the local innate immune response in relapsed HGG by intracranially injecting poly(I:C)to establish a robust antitumor immune response in this registered clinical trial(NCT03392545).During the follow-up,12/27(44.4%)patients who achieved tumor control concomitant with survival benefit were regarded as responders in our *** found that the T-cell receptor(TCR)repertoire in the TME was reshaped after poly(I:C)*** on the RNA-seq analysis of tumor samples,the expression of annexin A1(ANXA1)was significantly upregulated in the tumor cells of nonresponders,which was further validated at the protein *** vitro and in vivo experiments showed that ANXA1 could induce the production of M2-like macrophages and microglia via its surface receptor formyl peptide receptor 1(FPR1)to establish a Treg cell-driven immunosuppressive TME and suppress the antitumor immune response facilitated by poly(I:C).The ANXA1/FPR1 signaling axis can inhibit the innate immune response of glioma patients by promoting an anti-inflammatory and Treg-driven ***,ANXA1 could serve as a reliable predictor of response to poly(I:C),with a notable predictive accuracy rate of 92.3%.In light of these notable findings,this study unveils a new perspective of immunotherapy for gliomas.

Incompatible numerical manifold method for fracture problems

The incompatible numerical manifold method （INMM） is based on the finite cover approximation theory, which provides a unified framework for problems dealing with continuum and discontinuities. The incompatible numerical manifold method employs two cover systems as follows. The mathematical cover system provides the nodes for forming finite covers of the solution domain and the weighted functions, and the physical cover system describes geometry of the domain and the discontinuous surfaces therein. In INMM, the mathematical finite cover approximation theory is used to model cracks that lead to interior discontinuities in the process of displacement. Therefore, the discontinuity is treated mathematically instead of empirically by the existing methods. However, one cover of a node is divided into two irregular sub-covers when the INMM is used to model the discontinuity. As a result, the method sometimes causes numerical errors at the tip of a crack. To improve the precision of the INMM, the analytical solution is used at the tip of a crack, and thus the cover displacement functions are extended with higher precision and computational efficiency. Some numerical examples are given.

Electrocatalytically converting nitrate waste to value-added ammonia(NO3−RR)is beneficial to environmental restoration and an attractive candidate for green ammonia production[1].However,developing NO_(3)^(-)RR in an energy-saving way remains a significant challenge due to the slow kinetics inherent in multi-step electron and proton transfer,making previous investigations for NO_(3)^(-)RR generally operate at a large overpotential(<-0.2 V versus reversible hydrogen electrode(RHE))to achieve optimal efficiency and industrial current for NH3 synthesis[2].

Understanding the influencing mechanism of turbulent fluctuation on the ignition characteristics of millimeter coal particles is *** this work,to study the effect of turbulent fluctuation on ignition time,millimeter coal particles are subjected to a specific flow field,generated in a furnace with symmetric fans.A one-dimensional model with the new proposed correlation and the Ranz-Marshall(R-M)correlation for Nu(Nusselt number)is established to simulate the coal ignition *** addition,the effects of fan speed,temperature,particle diameter,particle distance and coal type on the ignition time are *** is found that an increase in fan speed from 0 to 3000 rpm leads to a particle Reynolds number Re_(p)increase from 0 to 22.5,and a turbulent particle Reynolds number Re_(t)*increase from 0 to *** a consideration of the fluctuation effect,the new correlation of Nu gives a better prediction of ignition time compared to the R-M ***,the ignition time is revealed to decrease with an increasing fan speed and an elevating *** the ignition time shows merely an initial boost with enlarging particle distance,it exhibits a linearity with the term of particle diameter dp1.3-1.7 and Reynolds numbers(Nu*/Nu)-0.6(Nu*is turbulent Nusselt number).Based on this relationship,the difference of predicted ignition time is calculated at different Re_(p)and Re_(t)*.It is shown that at low Re_(p)or high Re_(t)*values,the new correlation should substitute for the R-M correlation.

Investigation on the key factors in the hydrothermal synthesis of BN: The way of introducing sodium azide

The way of introducing sodium azide (NaN_3) into the reaction solution played an important role in the preparation of cBN by hydrothermal synthesis method. The results showed that both cBN content and crystalline perfection of the samples improved with increasing R_N value, and pure cBN could be obtained at 300℃ and 10 MPa when R_N increased to 3:1. Here R_N is defined as R_N =NaN_3(I)/NaN_3(II), wherNaN_3(I) denotes the amount of NaN_3 (in molar) that is added into the autoclave at the beginning of threaction process, and NaN_3(II) is the amount of NaN_3 (also in molar) introduced into the autoclave ahigh temperature and high pressure (i.e. 300℃ and 10 MPa). In order to explain the experimental results, a preliminary model was proposed in this paper.

Prognostic implications of epidermal growth factor receptor variant Ⅲ expression and nuclear translocation in Chinese human gliomas

Objective: To determine the prognostic implications and clinical significance of epidermal growth factor receptor variant Ⅲ(EGFRvⅢ) expression and EGFRvⅢ nuclear translocation in Chinese human ***: We retrospectively examined EGFRvⅢ expression and EGFRvⅢ nuclear translocation using immunohistochemistry in specimens of 240 Chinese patients with glioma, including 84 World Health Organization(WHO) II gliomas, 84 WHO Ⅲ gliomas and 72 glioblastomas(WHO IV). Factors that correlated with EGFRvⅢ and EGFRvⅢ nuclear translocation expression were analyzed by the Chi-square test. Kaplan-Meier methodology and Cox regression were used for the survival ***: Log-rank tests showed that patient age, Karnofsky performance scale(KPS) score, tumor grade,EGFRvⅢ expression, EGFRvⅢ nuclear translocation, 1 p/19 q codeletion, isocitrate dehydrogenase(IDH)mutation, Ki-67 labeling index and O6-methylguanine-DNA methyltransferase(MGMT) status(P<0.05) were significantly correlated with overall survival(OS) time. Multivariate Cox regression analysis revealed that patient age, tumor grade, EGFRvⅢ nuclear translocation, 1 p/19 q codeletion, and IDH mutation(P<0.05) were significantly correlated with OS. Patients with a high level of EGFRvⅢ nuclear translocation(≥7%) had both significantly shorter OS [hazard ratio(HR): 1.920, 95% confidence interval(95% CI): 1.228-3.003, P=0.004] and progression-free survival(PFS) times(HR: 1.661, 95% CI: 1.116-2.471, P=0.012) than those with a low level of EGFRvⅢ nuclear translocation(<7%).Conclusions: A high level of EGFRvⅢ nuclear translocation in glioma is an independent factor indicating a poor prognosis, but EGFRvⅢ expression is not an independent clinical prognostic factor. The level of EGFRvⅢ nuclear translocation maybe a novel and crucial prognostic biomarker in glioma.

随着我国风电领域的发展,行业内产生的相关数据在来源和数量上都达到了一个新的高度。这些数据被分散在各个业务系统中,若能对其进行统一管理并应用,则对行业具有积极作用。提出了一种用知识图谱管理风电数据的方法。首先,从多源风电数据中抽取出实体、关系、属性等知识图谱元素。在此过程中,分别给出了针对结构化与非结构化数据的抽取方法。其次,将得到的元素在图形数据库Neo4j中构建出了风电数据全景知识图谱,实现了不同类型、业务数据间的贯穿统一。最后,通过故障推理、业务查询等典型案例与传统的基于关系型数据库的数据管理模式作对比,结果显示提出的方法具有较好的运用效果。
