Development of viable cell estimation method without sacrificing proliferation and functions of cells cultured on regenerative biomaterials is essential for regenerative *** and depletion of resazurin are critical but often overlooked limitations that hindered applications of resazurin in viable cell *** present work found that cytotoxicity and depletion of resazurin depended on cell concentration,resazurin concentration and resazurin incubation time.A simple strategy which only allowed cells to incubate with resazurin during each measurement was developed to eliminate negative effects of *** strategy was verified by monitoring proliferation of MC3T3-E1 preosteoblasts on poly(D,L-lactic acid)scaffold during a continuous 3D culture process for up to 21 days,comparing the accuracy with MTT assay which is a destructive assay with high sensitivity and accuracy and commonly used in regenerative engineering and comparing viability,proliferation and differentiation functions of MC3T3-E1,which were treated with/without this strategy for nondestructive *** method showed comparable linearity of standard curve and characteristics of growth curve to MTT *** major negative effects of this method on MC3T3-E1 viability and functions were *** work highlighted the importance of the concentration and incubation time of resazurin in designing application-specific nondestructive viability assay and would be helpful in improving the implanted medical devices as well as in regenerative engineering.

Aroma is an important quality parameter for breeding in rice(Oryza sativa).For example,the aromatic rice varieties basmati and jasmine rice,with a popcorn-like scent,are popular worldwide and routinely command a price premium.2-acetyl-1-pyrroline(2AP)is a key flavor compound among over 200 volatiles identified in fragrant rice.A naturally fragrant germplasm exists in multiple plant species besides rice,which all exhibit lower activity of BETAINE ALDEHYDE DEHYDROGENASE 2(BADH2).However,no equivalent aromatic germplasm has been described in maize(Zea mays).Here,we characterized the two maize BADH2 homologs,ZmBADH2a and *** generated zmbadh2a and zmbadh2b single mutants and the zmbadh2a-zmbadh2b double mutant by CRISPR/Cas in four inbred lines.A popcorn-like scent was only noticeable in seeds from the double mutant,but not from either single mutant or in wild *** agreement,we only detected 2AP in fresh kernels and dried mature seeds from the double mutant,which accumulated between 0.028 and 0.723 mg/kg *** results suggest that ZmBADH2a and ZmBADH2b redundantly participate in 2AP biosynthesis in maize,and represent the creation of the world's first aromatic maize by simultaneous genome editing of the two BADH2 genes.

Development of the ignition delay prediction model of n-butane/hydrogen mixtures based on artificial neural network

Based on the experimental ignition delay results of n-butane/hydrogen mixtures in a rapid compression machine,a Genetic Algorithm(GA)optimized Back Propagation(BP)neural network model is originally developed for ignition delay *** the BP model,the activation function,learning rate and the neurons number in the hidden layer are optimized,*** prediction ability of the BP model is validated in wide operating ranges,i.e.,compression pressures from 20 to 25 bar,compression temperatures from 722 to 987 K,equivalence ratios from 0.5 to 1.5 and molar ratios of hydrogen(X_(H2))from 0 to 75%.Compared with the BP model,the GA optimized BP model could increase the average correlation coefficient from 0.9745 to 0.9890,in the opposite,the average Mean Square Error(MSE)decreased from 2.21 to *** the other hand,to assess the BP-GA model prediction ability in the never-seen-before cases,a limited BP-GA model is fostered in the𝑋X_(H2) range from 0 to 50%to predict the ignition delays at the cases of𝑋X_(H2)=75%.It is found that the predicted ignition delays are underestimated due to the training dataset lacking of“acceleration feature”that happened at𝑋X_(H2)=75%.However,three possible options are reported to improve the prediction accuracy in such never-seen-before cases.

In this paper, an asymmetric array structure of space laser communication receiver is proposed. This structure can greatly reduce alignment requirement, and lighten the signal strength jitter caused by atmospheric turbulence. A prototype of the proposed structure is fabricated and a 2.5 Mbit/s on-off keying(OOK) modulated demonstration link over 40 m free space is built. This asymmetric array structure can effectively collect optical signal while rotating in a window angle of ±17°, and the bit error ratio(BER) keeps zero.

Using naturally colored cotton(NCC)can eliminate dyeing,printing and industrial processing,and reduce sewage discharge and energy ***(PAs),the primary coloration components in brown fibers,are polyphenols formed by oligomers or polymers of flavan-3-ol units derived from *** essential structural genes for flavanone and flavonoid hydroxylation encoding flavanone-3-hydroxylase(F3H),flavonoid 3’-hydroxylase(F3’H)and flavonoid 3’5’-hydroxylase(F3’5’H)are initially committed in the flavonoid biosynthesis pathway to produce common *** three genes were all expressed predominantly in developing fibers of NCCs,and their expression patterns varied temporally and spatially among NCC *** GhF3Hi,GhF3’Hi and GhF3’5’Hi silenced lines of NCC varieties XC20 and ZX1,the expression level of the three genes decreased in developing cotton fiber,negatively correlated with anthocyanidin content and fiber color *** color depth and type in RNAi lines changed with endogenous gene silencing efficiency and expression pattern,the three hydroxylase genes functioned in fiber color ***3H showed functional differentiation among NCC varieties and GhF3’H acted in the accumulation of anthocyanin in *** with GhF3’H,GhF3’5’H was expressed more highly in brown fiber with a longer duration of expression and caused lighter color of fibers in GhF3’5’H silenced *** three genes regulating fiber color depth and type could be used to improve these traits by genetic manipulation.

Cell sheet engineering has been proven to be a promising strategy for cardiac remodeling post-myocardial infarction. However, insufficient mechanical strength and low cell retention lead to limited therapeutic efficiency. The thickness and area of artificial cardiac patches also affect their therapeutic efficiency. Cardiac patches prepared by combining cell sheets with electrospun nanofibers, which can be transplanted and sutured to the surface of the infarcted heart, promise to solve this problem. Here, we fabricated a novel cardiac patch by stacking brown adipose-derived stem cells (BADSCs) sheet layer by layer, and then they were combined with multi-walled carbon nanotubes (CNTs)-containing electrospun polycaprolactone/silk fibroin nanofibers (CPSN). The results demonstrated that BADSCs tended to generate myocardium-like structures seeded on CPSN. Compared with BADSCs suspension-containing electrospun nanofibers, the transplantation of the CPSN-BADSCs sheets (CNBS) cardiac patches exhibited accelerated angiogenesis and decreased inflammation in a rat myocardial infarction model. In addition, the CNBS cardiac patches could regulate macrophage polarization and promote gap junction remodeling, thus restoring cardiac functions. Overall, the hybrid cardiac patches made of electrospun nanofibers and cell sheets provide a novel solution to cardiac remodeling after ischemic myocardial infarction.

Oxidation behavior of C/C-SiC gradient matrix composites and C/C composites were compared in stationary air. The results show that oxidation threshold of C-SiC materials increases with the amount of SiC particles in the codeposition matrix. Oxidation rate of C/C-SiC gradient matrix composites is significantly lower than that of C/C material. The micro-oxidation process was observed by SEM.
