HMGB2 knockdown ameliorates retinal ganglion cell injury by inhibiting NLRP3 inflammasome activation after retinal ischemia

AIM:To explore the neuroprotective effects of high mobility group box 2(HMGB2)knockdown on retinal ganglion cells(RGCs)in the retinal ischemia-reperfusion injury(RIRI).METHODS:Oxygen-glucose deprivation(OGD)-injured RGCs from postnatal three-day C57BL/6 mice pups and high intraocular pressure(IOP)-induced RIRI mice were used as cellular and animal models of *** expression of HMGB2 in the retina of RIRI mice and OGD-injured RGCs was detected through reverse transcription-polymerase chain reaction(RT-qPCR)and Western *** effects of HMGB2 silencing on the morphological changes,RGCs survival,and cell apoptosis in mouse retinal tissues were observed through H&E staining,immunofluorescence staining with RNA-binding protein with multiple splicing(RBPMS)antibody,and TUNEL staining,*** viability and apoptosis were examined by CCK-8 and flow cytometry *** levels of proteins associated with NOD-like receptor thermal protein domain associated protein 3(NLRP3)-mediated pyroptosis[NLRP3,Caspase-1,GSDMD-N,interleukin(IL)-1β,IL-18]in vivo and in vitro were measured by Western ***:HMGB2 protein and NLRP3 were upregulated in the retina of RIRI mice and OGD-injured RGCs(P<0.001).The retina was edematous,accompanied by disorganized cell arrangement and decreased thickness of all layers,and obvious vacuoles in ganglion cell ***2 silencing alleviated the reduction in total retinal thickness and the severity of retinal tissue damage as well as suppressed RGC loss and retinal cell apoptosis in RIRI ***-induced RGC apoptosis was ameliorated after downregulation of HMGB2 in *** injection of the AAV-sh-HMGB2 and si-HMGB2 resulted in significantly decrease of NLRP3,Caspase-1,GSDMD-N,IL-1β,and IL-18 protein levels in the retinal tissues of RIRI mice and OGD-injured RGCs,respectively(all P<0.001).CONCLUSION:HMGB2 knockdown protects against RGC apoptosis and pyroptosis after RIRI through suppressing NLRP3 inflammasome activ

Effect of order–disorder transition on thermodynamic and electronic properties of σ and χ phases in W–Re alloy: the first-principles calculation

The Tungsten-Rhenium(W-Re) alloys,celebrated for their high melting point,strength at elevated temperatures,electrical resistivity,and radiation resistance,have been widely utilized in high-temperature components,aerospace,electronics,and nuclear *** constituents of the topologically close-packed(TCP) phases,the sigma phase(σ) and chi phase(χ) formed within W-Re alloys wield considerable influence on the mechanical properties and the stability of the *** calculations were utilized in the present work to explore the structural,thermodynamic,and electronic properties of both ordered and disordered configurations within the σ and χ phases,culminating in a systematic elucidation of the higher phase stability exhibited by the ordered *** is found that the bulk modulus of these two phases is directly proportional to the concentration of Re in the alloy,while the equilibrium volume is inversely *** thermodynamic parameters of the σ and χ phases are calculated via the mean-field potential *** similar trends observed in the isobaric heat capacity,enthalpy increment,and entropy change curves for these two phases suggest they possess comparable thermodynamic *** is noteworthy that the contribution of ionic vibrations predominantly affects the isobaric heat capacity,while the contribution of thermal electronic excitations increases linearly with *** the structure and thermodynamic properties of TCP phases in W-Re alloys at low temperatures has profound significance for optimizing material performance,microstructures features,establishing theoretical foundations,and predicting material behavior.

BACKGROUND Patients with depression following coronary heart disease often exhibit insufficient psychological resilience and self-care abilities;therefore,emphasis must be placed on nursing *** To analyze the application value of problem-oriented education combined with nursing interventions based on the Snyder hope theory model in depressed patients after percutaneous coronary intervention(PCI).METHODS This study included 150 patients diagnosed with PCI postoperative depression because of coronary heart disease between February 2022 and February *** were divided into two groups:A control group(n=75)receiving problem-oriented education and an observation group(n=75)receiving combined nursing interventions based on the Snyder hope theory *** status,psychological resilience,self-care ability,and quality of life were compared between the two *** Before nursing interventions,there were no significant differences between the two groups(P>0.05).After the interventions,depression scores decreased while psychological resilience,self-care ability,and quality of life scores increased significantly in the observation group compared to that in the control group,with statistically significant differences noted(P<0.05).This combined approach can enhance psychological resilience,improve self-care abilities,and elevate the overall quality of life,warranting further promotion in clinical *** Combination of problem-oriented education and nursing interventions based on the Snyder hope theory model effectively alleviates depression in patients following PCI for coronary heart disease.

Finite temperature physics of ID to pological Kondo insulator: Stable Haldane phase, emergent energy scale and beyond

In recent years, mteracting topological insulators have emerged as new frontiers in condensed matter physics, and the hotly studied topological Kondo insulator (TKI) is one of such prototypes. Although its zero-temperature ground-state has been widely investigated, the finite temperature physics on TKI is largely unknown. Here, we explore the finite temperature properties in a simplified model for TKI. namely the one-dimensional p-wave periodic Anderson model, with numerically exact determinant quantum Monte Carlo simulation. It is found that the topological Haldane phase established for groimdstate is still stable against small thermal fluctuation and its characteristic edge magnetization develops at low temperature. Such facts emphasize the robustness of (symmetry-protected) topological order against temperature effect, which always exists at real physical world. Moreover, we use the saturated low-T spin structure factor and the 1/T-law of susceptibility to detect the free edge spin moment, interestingly the low-temperature upturn behaxior of the latter one is similar to experimental finding in SniBg at T < 50 K. It implies that similar physical mechanism may work both for idealized models and realistic correlated electron *** have also identified an emergent energy scale Tcr. which signals a crossover into interesting low-T regime and seems to be the expected Ruderman-Kittel- Kasuya Yosida coupling. Finally, the collective Kondo screening effect has been examined and it is heavily reduced at boundary which may give a fruitful playground for novel physics beyond the wellestablished Haldane phase and topological band insulators.

Based on ab initio calculations,we utilize the mean-field potential approach with the quantum modification in conjunction with stress–strain relation to investigate the elastic anisotropies and sound velocities of hcp and bcc Be under high-temperature(0–6000 K)and high-pressure(0–500 GPa)*** propose a general definition of anisotropy for elastic moduli and sound *** suggest that the elastic anisotropy of Be is more significantly influenced by pressure than by *** pressure-induced increase of c/a ratio makes the anisotropy of hcp Be significantly ***,the hcp Be still exhibits smaller anisotropy than bcc Be in terms of elastic moduli and sound *** suggest that measuring the anisotropy in shear sound velocity may be an approach to distinguishing the hcp–bcc phase transition under extreme conditions.

Development and Validation of a Simoa Assay for Determination of Recombinant Batroxobin in Human Serum

Recombinant batroxobin(S3101)is a thrombin-like serine protease that binds to fibrinogen or is taken up by the reticuloendothelial system.A literature survey showed no adequate method that could determine sufficient concentrations to evaluate pharmacokinetic parameters for phase I clinical ***,a sensitive method is urgently needed to support the clinical pharmacokinetic evaluation of *** this study,a sensitive bioanalytical method was developed and validated,using a Quanterix single molecular array(Simoa)***,to thoroughly assess the platform,enzyme-linked immunosorbent assay and electrochemiluminescence assay were also developed,and their performance was compared with that of this novel technology *** assay was validated in compliance with the current *** with the Simoa assay were precise and accurate,presenting a valid assay range from 6.55 to 4000 pg/*** intra-and inter-run accuracy and precision were within-19.3%to 15.3%and 5.5%to 17.0%,respectively.S3101 was stable in human serum for 280 days at-20℃and-70℃,for 2 h prior to pre-treatment and 24 h post pre-treatment at room temperature(22℃-28℃),respectively,and after five and two freeze-thaw cycles at-70℃and-20oC,*** Simoa assay also demonstrated sufficient dilution linearity,assay sensitivity,and parallelism for quantifying S3101 in human *** Simoa assay is a sensitive and adequate method for evaluating the pharmacokinetic parameters of S3101 in human serum.

The Institute of Urology, Peking University prostatectomy score： a simple preoperative classification of prostate cancer for predicting surgical difficulty and risk

Traditional laparoscopic radical prostatectomy is a treatment choice in many developing countries and regions for most patients with localized prostate cancer; however, no system for predicting surgical difficulty and risk has been established. This study aimed to propose a simple and standard preoperative classification system of prostate cancer using preoperative data to predict surgical difficulty and risk and to evaluate the relationship between the data and postoperative complications. We collected data from 236 patients and divided them into three groups to evaluate and validate the relationships among preoperative, operative, and postoperative data. This new scoring system is based on the body mass index, ultrasonic prostate volume, preoperative prostate-specific antigen level, middle lobe protrusion, and clinical stage. In the scoring group, we classified 89 patients into two groups： the low-risk group （score of 〈4） and high-risk group （score of ~4）, and then compared the postoperative data between the two groups. The positive surgical margin rate was higher in the high-risk group than low-risk group. The results in validation Groups A and B were similar to those in the scoring group. The focus of our scoring system is to allow for preliminary assessment of surgical difficulty by collecting the patients＇ basic information. Urologists can easily use the scoring system to evaluate the surgical difficulty and predict the risks of a positive surgical margin and urinary incontinence in patients undergoing laparoscopic radical prostatectomy.

Elaborating strengthen mechanism of Pt-Ir solid solution superalloy at finite temperature

Pt-Ir alloy is potential superalloys used above 1300℃because of their high strength and creep ***,the ductility of Pt-Ir alloy has rapidly deteriorated with the increase of Ir,resulting in poor *** work quantitatively evaluated the solid solution strengthening(SSS)and grain refinement strengthening(GRS)of Pt-Ir alloy using first-principles calculations combined with experimental ***,the stretching force constants in the second nearest neighbor region(SFC^(2nd))of pure Ir(193.7 eV·nm^(-2))are 3.40 times that of pure Pt(57.0 eV·nm^(-2)),i.e.,the interatomic interaction is greatly enhanced with the increase of Ir content,which leads to the decrease of ductility,and modulus misfit plays a dominant role in ***,the physical mechanisms responsible for the hardness(H_(V))of Pt-Ir alloy,using the power-law-scaled function of electron work function coupled SSS and GRS,are attributed to the electron redistribution caused by different Ir ***,a thorough assessment of the thermodynamic characteristics of Pt-Ir binary alloy was conducted,culminating in development of a mapping model that effectively relates enmposition,temperature and *** results revealed that the compressive strength incrcases with the Ir content,and the highest strength was observed in Pt_(0.25)Ir_(0.75).This study provides valuable insights into the Pt-Ir alloy system.

BACKGROUND Jackstone is a rare entity of calculi in urinary tracts and has the characteristic appearance resembling toy *** are nearly always reported to occur in the urinary bladder,we first report a rare case of jackstone located in the obstructed renal *** SUMMARY We report a 46-year-old man presenting with intermittent,painless gross hematuria and left flank *** computed tomography revealed staghorn stones and secondary hydronephrosis.A jackstone with radiating branches was found in one of the dilated renal *** nephrolithotomy was performed and endoscopic images were recorded during the *** stone composition analysis revealed it as calcium oxalate monohydrate *** Jackstones can occur in the renal collecting system besides the *** unique appearance and imaging manifestations are the most important factors in the diagnosis of jackstones,and further exploration of the formation mechanism is required.
