Analysis of SSRs Information in Capsicum spp. from EST Database

SSR markers are useful in pepper linkage mapping and gene location. 446 SSR markers have been reported, but they are insufficient. It is costly to develop SSR markers from DNA library, whereas it seems much easy to find in EST sequences in the GenBank of pepper through internet. In this study, attempts have been made to develop SSR markers in the EST sequences by using bioinformatics. EST sequences were trimmed by ‘***’ software, while 116 915 EST sequences were obtained without poly ‘A’ or poly ‘T’, ranged between 100 and 700 bp. Using ‘e-PCR’ and ‘***’ softwares, SSR sequences were identified. 2 508 microsatellite loci （larger than 20 repeats） were established and 755 SSR primers were designed using SSR finder software and Primer 3 software. There were 498 （0.43%） mono-, 1 026 （0.89%） di-, 518 （0.45%） tri-, 245 （0.21%） tetra-, 114 （0.10%） penta-, and 107 （0.09%） hexa-nucleotide SSRs. The estimated frequency of SSRs was approximately 1/25.12 kb. According to the distribution of SSRs in pepper, the mean length of pepper SSRs was 22.68 bp and the adenine rich repeats such as A/T, AG, AT, AAG, AAAT, and AAAC were predominant in each type of SSRs （mono-, di-, tri-, tetra-, penta-, and hexa-）, whereas the C/G, CG, CCG repeats were less abundant. 210 primers were tested in 8 pepper cultivars and the PCR result revealed the existence of polymorphism among 127 （60.48%） SSR primers within 8 pepper cultivars. It is confirmed that pepper EST database could be efficiently exploited for availability of SSR markers.

Greatly enhanced mechanical properties of thermoelectric SnSe through microstructure engineering

The outstanding thermoelectric material, SnSe, is also known for its inferior mechanical properties, which bring great inconvenience for its application in thermoelectric devices. In this work, SnSe bulks were prepared via a sequential procedure of high-pressure synthesis (HPS), ball milling, and spark plasma sintering (SPS). The produced polycrystalline samples with a unique microstructure of tightly-bound quasi-equiaxed grains exhibited excellent mechanical properties. The Vickers hardness (HV), compressive strength (σ_(c)), and bending strength (σ_(b)) reached 1.1 GPa, 300 MPa, and 90 MPa, respectively, all of which are far superior to those of ordinary polycrystalline SnSe. Furthermore, the microstructures did not deteriorate thermoelectric performance. This work demonstrated an effective procedure to prepare polycrystalline microstructure-engineered SnSe materials, which not only show advantages in device applications but also shed light on property enhancement for other layer-structured thermoelectric materials.

Heavy-atom engineered hypoxia-responsive probes for precisive photoacoustic imaging and cancer therapy

Photoacoustic agents combining photodynamic therapy(PDT) and photothermal therapy(PTT) functions have emerged as potent theranostic agents for combating cancer. The molecular approaches for enhancing the near-infrared(NIR)-absorption and maximizing non-radiative energy transfer are essential for effective photoacoustic imaging(PAI) and therapy applications. In addition, such molecules with high specificity and affinity to cancer cells are urgently needed, which would further decrease the side effect during treatments. In this study, we applied a heavy-atom engineering strategy and introduced p-aminophenol,-thio, and-seleno moieties into NIR heptamethine cyanine(Cy7) skeleton(Cy7-X-NH_(2), X = O, S, Se) to significantly increase photothermal conversion efficiency for PTT and promote intersystem crossing for ***, we designed a series of nitroreductase(NTR)-activated photoacoustic probes(Cy7-X-NO_(2),X = O, S, Se), and target hypoxic tumors with NTR overexpression. Our prostate cancer targeting probe,Cy7-Se-NO_(2)-KUE, exhibited specific tumor photoacoustic signals and effective tumor killing through outstanding synergistic PTT/PDT in vivo. These findings highlighted a versatile strategy for cancer photoacoustic diagnosis and enhanced phototherapy.

Predictors for malignant potential and deep submucosal invasion in colorectal laterally spreading tumors

Separation, determination of six impurities in methotrexate drug substance using ultra-performance liquid chromatography

Methotrexate （MTX） is an antineoplastic therapeutic medicine as antimetabolite of folic acid. In this paper, a sensitive and rapid ultra-performance liquid chromatographic （UPLC） method was developed and validated for the separation and determination of impurities in MTX drug substances. The UPLC method was accomplished on an Agilent Zorbax Extend C-18 （50 mm × 4.6 mm, 1.8 μm） with a gradient elution system composed of sodium dihydrogen phosphate in water （20 mmol/L, pH 3.0） and acetonitrile. The flow rate was 2.2 mL/min. The method was validated. The calibration curves displayed good linearity （r 〉 0.999） within the tested concentration ranges. The limit of detection （LOD） and limit of quantification （LOQ） of the six analytes were all less than 0.774 μg/mL and 1.03μg/mL. The relative standard deviation （RSD） for intra- and inter-day precision of the six analytes was less than 9.8%, including at the LOQ. The average recovery ranged from 95.2% to 103 except at the LOQ where recovery ranged from 82.7% to 117%. The validated method was successfully used to determine the relative abundance of six impurities in the MTX drug substances.

Quantum key distribution over a mimicked dynamic-scattering channel

Free-space quantum key distribution(QKD) plays an important role in the global quantum network. However, free space channels suffer from the atmospheric turbulence and scattering effects of haze,fog, and dust, which significantly weaken the performance of QKD or even block the secure quantum ***, we prove the performance of QKD over a dynamic scattering channel can be enhanced significantly using a fast wavefront shaping technique. The system achieves on average 10-dB enhancement of quantum transmission efficiency and establishes a secure quantum link for QKD. Our work demonstrates the feasibility of QKD through free-space dynamic scattering channels and enhances the deployment capability of complex-channel QKD system.
