The present study aimed to evaluate the clinical outcomes of magnetic-activated cell sorting(MACS)in sperm preparation for male subjects with a sperm DNA fragmentation index(DFI)≥30%.A total of 86 patients who had undergone their first long-term long protocol were *** protocol involved in vitro fertilization(IVF)and intracytoplasmic sperm injection(ICSI)cycles,and the patients were divided into the MACS or control *** MACS group included sperm samples analyzed with MACS that were combined with density gradient centrifugation(DGC)and the swim-up(SU)technique(n=39),and the control group included sperm samples prepared using standard techniques(DGC and SU;n=41).No differences were noted with regard to basic clinical characteristics,number of oocytes retrieved,normal fertilization rate,cleavage rate,or transplantable embryo rate between the two groups in IVF/*** addition,the clinical pregnancy and implantation rates of the first embryo transfer cycles indicated no significant differences between the two ***,there was a tendency to improve the live birth rate(LBR)of the first embryo transfer cycle(63.2%vs 53.9%)and the cumulative LBR(79.5%vs 70.7%)in the MACS group compared with the control ***,the number of transferred embryos(mean±standard deviation[s.d.]:1.7±0.7 vs 2.3±1.6)and the transfer number of each retrieved cycle(mean±s.d.:1.2±0.5 vs 1.6±0.8)were significantly lower in the MACS group than those in the control ***,the selection of nonapoptotic spermatozoa by MACS for higher sperm DFI could improve assisted reproductive clinical outcomes.

Ggps1 deficiency in the uterus results in dystocia by disrupting uterine contraction

Dystocia is a serious problem for pregnant women, and it increases the cesarean section rate. Although uterine dysfunction has an unknown etiology, it is responsible for cesarean delivery and clinical dystocia, resulting in neonatal morbidity and mortality;thus, there is an urgent need for novel therapeutic agents. Previous studies indicated that statins, which inhibit the mevalonate (MVA) pathway of cholesterol synthesis, can reduce the incidence of preterm birth, but the safety of statins for pregnant women has not been thoroughly evaluated. Therefore, to unambiguously examine the function of the MVA pathway in pregnancy and delivery, we employed a genetic approach by using myometrial cell-specific deletion of geranylgeranyl pyrophosphate synthase (Ggps1) mice. We found that Ggps1 deficiency in myometrial cells caused impaired uterine contractions, resulting in disrupted embryonic placing and dystocia. Studies of the underlying mechanism suggested that Ggps1 is required for uterine contractions to ensure successful parturition by regulating RhoA prenylation to activate the RhoA/Rock2/p-MLC pathway. Our work indicates that perturbing the MVA pathway might result in problems during delivery for pregnant females, but modifying protein prenylation with supplementary farnesyl pyrophosphate or geranylgeranyl pyrophosphate might be a strategy to avoid side effects.

In this study, a high-carbon nano-bainitic GCr15Si1Mo bearing steel was investigated. Specifically, the effects of content and size of undissolved carbides on the microstructure and transformation kinetics of nano-bainite were analyzed. The results demonstrated that after prolonged austempering at low temperatures, the mixed microstructure composed of nano-bainite (NB), undissolved carbides (UC), and retained austenite (RA) was obtained in GCr15SiMo steel. When the experimental steel was austenitized at 900 ℃, the undissolved carbides gradually dissolved until reaching a stable state with increasing holding time. Furthermore, at the same austempering temperature, despite different volume fractions of undissolved carbides in the substrate, the volume fractions of nano-bainite in the final microstructures remained essentially the same. Moreover, the higher the content of undissolved carbides in steel, the faster the transformation rate of nano-bainite and the shorter the total transformation time.

Adversarial data splitting for domain generalization

Domain generalization aims to learn a model that is generalizable to an unseen target domain,which is a fundamental and challenging task in machine learning for out-of-distribution generalization. This paper proposes a novel domain generalization approach that enforces the learned model to be able to generalize well over the train/val subset splitting of the training dataset. This idea is modeled herein as an adversarial data splitting framework, formulated as a min-max optimization problem inspired by the metalearning approach. The min-max optimization problem is solved by iteratively splitting the training dataset into the training and val subsets to maximize the domain shift measured by the objective function and updating the model parameters to enable the model to generalize well from the training subset to the val subset by minimizing the objective function. This adversarial training approach does not assume the known domain labels of the training data; instead, it automatically investigates the “hard” splitting of the train/val subsets to learn the generalizable model. Extensive experimental results using three benchmark datasets demonstrate the superiority of this approach. In addition, we derive a generalization error bound for the theoretical understanding of our proposed approach.

Adverse nutritional conditions during the perinatal stage are related to early menopause in adulthood;however,the underlying mechanism is still ***,we revealed that colostrum-activated ketone body elevation during the postnatal stage regulated primordial follicle reservoir size and then affected ovarian *** found that the expression of the ketogenesis rate-limiting enzyme 3-hydroxy-3-methylglutaryl-CoA synthase 2(Hmgcs2)was largely enhanced during primordial follicle pool formation after birth and might be activated in the ovaries by *** oxygen species(ROS)elevation in the ovaries leads to follicle apoptosis to deplete damaged follicles,while Hmgcs2 deficiency enhances follicle apoptosis and thus decreases the size of the primordial follicle pool and leads to premature ovarian ageing(POA),which might be related to the activation of cellular endogenous antioxidant *** these defects could be rescued by ketone body administration,which suppressed ROS-activated follicle *** results suggest that the internal metabolic homeostasis of newborn mice is critical for the primordial reservoir and that any intrauterine and perinatal undernutrition could result in POA.

A projected Newton algorithm based on chemically allowed interval for chemical equilibrium computations

The chemical equilibrium equations utilized in reactive transport modeling are complex and nonlinear,and are typically solved using the Newton-Raphson *** this algorithm is known for its quadratic convergence near the solution,it is less effective far from the solution,especially for ill-conditioned *** such cases,the algorithm may fail to converge or require excessive *** address these limitations,a projected Newton method is introduced to incorporate the concept of *** method constrains the Newton step by utilizing a chemically allowed interval that generates feasible descending ***,we utilize the positive continuous fraction method as a preconditioning technique,providing reliable initial values for solving the *** numerical results are compared with those derived using the regular Newton-Raphson method,the Newton-Raphson method based on chemically allowed interval updating rules,and the bounded variable least squares method in six different test *** numerical results highlight the robustness and efficacy of the proposed algorithm.
