Meiosis is the process of producing haploid gametes through a series of complex chromosomal events and the coordinated action of various proteins.The mitochondrial protease complex(ClpXP),which consists of caseinolytic mitochondrial matrix peptidase X(ClpX)and caseinolytic protease P(ClpP)and mediates the degradation of misfolded,damaged,and oxidized proteins,is essential for maintaining mitochondrial homeostasis.ClpXP has been implicated in meiosis regulation,but its precise role is currently unknown.In this study,we engineered an inducible male germ cell-specific knockout caseinolytic mitochondrial matrix peptidase X(Clpx^(cKO))mouse model to investigate the function of ClpX in meiosis.We found that disrupting Clpx in male mice induced germ cell apoptosis and led to an absence of sperm in the epididymis.Specifically,it caused asynapsis of homologous chromosomes and impaired meiotic recombination,resulting in meiotic arrest in the zygotene-to-pachytene transition phase.The loss of ClpX compromised the double-strand break(DSB)repair machinery by markedly reducing the recruitment of DNA repair protein RAD51 homolog 1(RAD51)to DSB sites.This dysfunction may be due to an insufficient supply of energy from the aberrant mitochondria in Clpx^(cKO) spermatocytes,as discerned by electron microscopy.Furthermore,ubiquitination signals on chromosomes and the expression of oxidative phosphorylation subunits were both significantly attenuated in Clpx^(cKO) spermatocytes.Taken together,we propose that ClpX is essential for maintaining mitochondrial protein homeostasis and ensuring homologous chromosome pairing,synapsis,and recombination in spermatocytes during meiotic prophase I.

环境保护的压力和资源短缺问题正驱使我们寻找有效利用废物和减轻资源枯竭的解决方案。在建筑业中,建筑拆除产生的混凝土废料占到总废物的30%~40%。高额的处理费用、垃圾填埋税和有限的处理场地为开发再生混凝土提供了可能性。然而,由于再生混凝土的质量相较于普通混凝土较低,再生混凝土并未得到广泛应用。文章旨在探讨使用再生混凝土替代普通混凝土在建筑应用中的可能性,方法包括使用再生骨料替代不同比例的天然骨料,并使用粉煤灰和矿渣水泥共同替代水泥。研究表明,矿渣和粉煤灰是改善混凝土性能的有效元素。此外,再生骨料对混凝土混合物的抗压强度有积极影响。随着再生骨料的增加,混凝土强度得到提升,这可能与再生骨料的高质量和混合方法有关。

解码芯片在对旋转变压器输出的位置信号做处理的过程中,芯片在采样时容易产生各种非理想偏差,这会造成转子位置估算精度的急剧下降。针对该问题,分析了电压信号中的幅值偏差和正交偏差会对旋转变压器解码的输出角度叠加一个2倍频的波动,直流偏差会叠加一个同频波动的问题,提出了一种基于离散傅里叶变换(DFT)的输入矫正法用于实现旋转变压器输出信号的偏差矫正,同时有效降低一次二次波动。该方法首先离散采样信号,通过DFT公式提取旋转变压器输出信号的直流信号和基频信号;然后基于直流信号和基频信号先后计算直流偏差、幅值偏差和正交偏差,通过控制器实现偏差值矫正。最后,仿真和试验结果表明,在芯片采样产生偏差时,所提出的偏差提取方法有效,试验中相应的正余弦输出信号指定阶次的波动更小。

BACKGROUND Persistent vegetative state(PVS)is a devastating and long-lasting clinical condition with high morbidity and mortality;currently,there are no available effective interventions.CASE SUMMARY We report the case of an 11-year-old boy with PVS caused by severe intracerebral bleeding in the left hemisphere following anticoagulation treatment.The patient’s PVS severity showed no notable improvement after 2-mo neuroprotective treatment and rehabilitation,including nerve growth factor and baclofen,hyperbaric oxygen,and comprehensive bedside rehabilitation therapies.Daily inhalation treatment(4-6 h)of high-concentration hydrogen(H2)gas(66.6%H2+33.3%O2)was provided.Surprisingly,the patient’s orientation,consciousness,ability to speak,facial expressions,and locomotor function were significantly restored,along with improvements in essential general health status,after H2 gas inhalation treatment,which was consistent with stabilized neuropathology in the left hemisphere and increased Hounsfield unit values of computed tomography in the right hemisphere.The patient finally recovered to a near normal conscious state with a Coma Recovery Scale-Revised Score of 22 from his previous score of 3.CONCLUSION Phase 1 clinical trials are needed to explore the safety and efficacy of H2 gas inhalation in patients with PVS.

对于高速永磁同步电机(PMSM)无位置传感器控制研究中,针对传统滑模观测器(SMO)存在滑模抖振,以及在观测到的反电动势信号经过低通滤波器(LPF)消除高频谐波造成的相位滞后的问题和高速下由于电流采样造成的数字延时问题,提出一种基于二阶广义积分器-锁频环(SOGI-FLL)的改进SMO方法。针对上述问题,首先利用双曲正切函数来代替sign函数,来减少滑模的抖振;然后引入SOGI-FLL代替LPF来对观测的反电动势进行滤波,进而消除相位滞后。其次在此基础上补偿由于数字延时造成的角度误差。最后搭建PMSM实验平台验证此方法的可行性,实现PMSM无位置传感器控制在中高速域转子速度和位置的准确估算。

Nanomedicine Disrupts Stromal Barriers to Augment Drug Penetration for Improved Cancer Therapy

Tumor stroma composing diverse extracellular matrixes(ECM)and stromal cells shapes a condensed physical barrier,which severely hampers the efficient accessibility of nanomedicine to tumor cells,especially these deep-seated in the core of tumor.Such barrier significantly compromises the antitumor effects of drug-loaded nanomedicine,revealing the remarkable importance of disrupting stromal barrier for improved tumor therapy with deep penetration ability.To achieve this goal,various nanoparticle-based strategies have been developed,including direct depleting ECM components via delivering anti-fibrotic agents or targeting stromal cells to suppress ECM expression,dynamic regulation of nanoparticles’physicochemical properties(i.e.,size,surface charge,and morphology),mechanical force-driven deep penetration,natural/biomimetic self-driven nanomedicine,and transcytosis-inducing nanomedicine.All these nanostrategies were systemically summarized in this review,and the design principles for obtaining admirable nanomedicine were included.With the rapid development of nanotechnology,elaborate design of multifunctional nanomedicine provides new opportunities for overcoming the critical stromal barriers to maximize the therapeutic index of various therapies,such as chemotherapy,photodynamic therapy,and immunotherapy.

Supermassive black holes triggered by QCD axion bubbles

Supermassive black holes(SMBHs)are ubiquitous in the center of galaxies,although the origin of their massive seeds is still unknown.In this paper,we investigate the formation of SMBHs from the quantum chromodynamics(QCD)axion bubbles.In this case,primordial black holes(PBHs)are considered as the seeds of SMBHs,which are generated from the QCD axion bubbles due to an explicit Peccei–Quinn(PQ)symmetry breaking after inflation.The QCD axion bubbles are formed when the QCD axion starts to oscillate during the QCD phase transition.We consider a general case in which the axion bubbles are formed with the bubble effective angleθeff∈(0,π],leading to the minimum PBH mass∼O(10^(4)−10^(7))M⊙for the axion decay constant f_(a)∼O(10^(16))GeV.The PBHs at this mass region may account for the seeds of SMBHs.

Altered intra- and inter-network brain functional connectivity in upper-limb amputees revealed through independent component analysis

Although cerebral neuroplasticity following amputation has been observed, little is understood about how network-level functional reorganization occurs in the brain following upper-limb amputation. The objective of this study was to analyze alterations in brain network functional connectivity(FC) in upper-limb amputees(ULAs). This observational study included 40 ULAs and 40 healthy control subjects;all participants underwent resting-state functional magnetic resonance imaging. Changes in intra-and inter-network FC in ULAs were quantified using independent component analysis and brain network FC analysis. We also analyzed the correlation between FC and clinical manifestations, such as pain. We identified 11 independent components using independent component analysis from all subjects. In ULAs, intra-network FC was decreased in the left precuneus(precuneus gyrus) within the dorsal attention network and left precentral(precentral gyrus) within the auditory network;but increased in the left Parietal_Inf(inferior parietal, but supramarginal and angular gyri) within the ventral sensorimotor network, right Cerebelum_Crus2(crus Ⅱ of cerebellum) and left Temporal_Mid(middle temporal gyrus) within the ventral attention network, and left Rolandic_Oper(rolandic operculum) within the auditory network. ULAs also showed decreased inter-network FCs between the dorsal sensorimotor network and ventral sensorimotor network, the dorsal sensorimotor network and right frontoparietal network, and the dorsal sensorimotor network and dorsal attention network. Correlation analyses revealed negative correlations between inter-network FC changes and residual limb pain and phantom limb pain scores, but positive correlations between inter-network FC changes and daily activity hours of stump limb. These results show that post-amputation plasticity in ULAs is not restricted to local remapping;rather, it also occurs at a network level across several cortical regions. Derivation of Expanded Isospectral-Nonisospectral Integrable Hierarchies via the Column-vector Loop Algebra

A scheme for generating nonisospectral integrable hierarchies is introduced.Based on the method,we deduce a nonisospectral hierarchy of soliton equations by considering a linear spectral problem.It follows that the corresponding expanded isospectral and nonisospectral integrable hierarchies are deduced based on a 6 dimensional complex linear space ■.By reducing these integrable hierarchies,we obtain the expanded isospectral and nonisospectral derivative nonlinear Schr?dinger equation.By using the trace identity,the biHamiltonian structure of these two hierarchies are also obtained.Moreover,some symmetries and conserved quantities of the resulting hierarchy are discussed.

With continuous population and economic growth in the 21st century,plastic pollution is a major global issue.However,the health concern of microplastics/nanoplastics(MPs/NPs)decomposed from plastic wastes has drawn public attention only in the recent decade.This article summarizes recent works dedicated to understanding the impact of MPs/NPs on the liver-the largest digestive organ,which is one of the primary routes that MPs/NPs enter human bodies.The interrelated mechanisms including oxidative stress,hepatocyte energy re-distribution,cell death and autophagy,as well as immune responses and inflammation,were also featured.In addition,the disturbance of microbiome and gut-liver axis,and the association with clinical diseases such as metabolic dysfunction-associated fatty liver disease,steatohepatitis,liver fibrosis,and cirrhosis were briefly discussed.Finally,we discussed potential directions in regard to this trending topic,highlighted current challenges in research,and proposed possible solutions.
