The gut microbiota plays a pivotal role in systemic metabolic processes and in particular functions,such as developing and preserving the skeletal muscle ***,the interplay between gut microbiota/metabolites and the regulation of satellite cell(SC)homeostasis,particularly during aging,remains *** propose that gut microbiota and its metabolites modulate SC physiology and homeostasis throughout skeletal muscle development,regeneration,and aging *** investigation reveals that microbial dysbiosis manipulated by either antibiotic treatment or fecal microbiota transplantation from aged to adult mice,leads to the activation of SCs or a significant reduction in the total ***,employing multi-omics(e.g.,RNA-seq,16S r RNA gene sequencing,and metabolomics)and bioinformatic analysis,we demonstrate that the reduced butyrate levels,alongside the gut microbial dysbiosis,could be the primary factor contributing to the reduction in the number of SCs and subsequent impairments during skeletal muscle ***,butyrate supplementation can mitigate the antibiotics-induced SC activation irrespective of gut microbiota,potentially by inhibiting the proliferation and differentiation of SCs/*** butyrate effect is likely facilitated through the monocarboxylate transporter 1(Mct1),a lactate transporter enriched on membranes of SCs and *** a result,butyrate could serve as an alternative strategy to enhance SC homeostasis and function during skeletal muscle *** findings shed light on the potential application of microbial metabolites in maintaining SC homeostasis and preventing skeletal muscle aging.

Precise targeting of specific regions within the central nervous system(CNS)is crucial for both scientific research and gene therapy in the context of brain ***-associated virus 13(AAV13)is known for its restricted diffusion range within the CNS,making it an ideal choice for precise labeling and administration within small brain ***,AAV13 mediates relatively low expression of target ***,we introduced specifically engineered modifications to the AAV13 capsid protein to enhance its transduction *** first constructed AAV13-YF by mutating tyrosine to phenylalanine on the surface of the AAV13 *** then inserted the 7m8 peptide,known to enhance cell transduction,into positions 587/588 and 585/586 of the AAV13 capsid,resulting in two distinct variants named AAV13-587-7m8 and AAV13-585-7m8,*** found that AAV13-YF exhibited superior in vitro infectivity in HEK293T cells compared to AAV13,while AAV13-587-7m8 and AAV13-585-7m8 showed enhanced CNS infection capabilities in C57BL/6 mice,with AAV13-587-7m8 infection retaining a limited spread *** modified AAV13 variants hold promising potential for applications in gene therapy and neuroscience research.

Dissecting the genetic basis of grain color and pre-harvest sprouting resistance in common wheat by association analysis

Pre-harvest sprouting(PHS) adversely affects wheat quality and yield, and grain color(GC) is associated with PHS resistance. However, the genetic relationship between GC and PHS resistance remains unclear. In this study, 168 wheat varieties(lines) with significant differences in GC and PHS resistance were genotyped using an Illumina 90K iSelect SNP array. Genome-wide association study(GWAS) based on a mixed linear model showed that 67 marker-trait associations(MTAs) assigned to 29 loci, including 17 potentially novel loci, were significantly associated with GC, which explained 1.1–17.0% of the phenotypic variation. In addition, 100 MTAs belonging to 54 loci, including 31 novel loci, were significantly associated with PHS resistance, which accounted for 1.1–14.7% of the phenotypic variation. Subsequently, two cleaved amplified polymorphic sequences(CAPS) markers, 2B-448 on chromosome 2B and 5B-301 on chromosome 5B,were developed from the representative SNPs of the major common loci ***-2B.3/***-2B.4controlling GC/PHS resistance and PHS resistance locus ***-5B.4, respectively. Further validation in 171 Chinese mini-core collections confirmed significant correlations of the two CAPS markers with GC and PHS resistance phenotypes under different environments(P<0.05). Furthermore, the wheat public expression database, transcriptomic sequencing, and gene allelic variation analysis identified TraesCS5B02G545100, which encodes glutaredoxin, as a potential candidate gene linked to ***-5B.4. The new CAPS marker CAPS-356 was then developed based on the SNP(T/C) in the coding sequences(CDS) region of TraesCS5B02G545100. The high-density linkage map of the Jing 411/Hongmangchun 21 recombinant inbred lines(RILs) constructed based on specific locus amplified fragment sequencing markers showed that CAPS-356 collocated with a novel QTL for PHS resistance, supporting the role of TraesCS5B02G545100 as the potential candidate gene linked to ***-5B.4. 解冻方式对炝蟹食用品质的影响

为研究不同解冻方式对冻藏炝蟹食用品质的影响,筛选食用前的最佳解冻方式,对低温解冻、室温解冻、流水解冻、超声解冻和脉冲磁场辅助解冻5种方式的解冻曲线、感官评价、电子舌、蟹肉蛋白质及其降解产物和呈味核苷酸含量变化进行分析。结果表明:超声和脉冲磁场辅助解冻最具时效性(10.4、26.2 min),低温解冻最耗时(312.6 min)。脉冲磁场辅助解冻蟹肉的色泽、质地和气味的评分最高,超声解冻的口感评分最好。电子舌结果表明,低温解冻、脉冲磁场辅助解冻和超声解冻蟹肉品质较接近,与流水解冻和室温解冻之间存在较大差异。超声解冻蟹肉的水分质量分数最高(84.37%),脉冲磁场辅助解冻的蛋白降解指数最低(13.83%),流水解冻的蟹肉水分质量分数最低(77.36%),且蛋白降解指数最高(16.64%)。低温解冻蟹肉的总游离氨基酸含量最高(2885.78 mg/100 g),脉冲磁场辅助和超声解冻蟹肉中谷氨基酸、甘氨酸和丙氨酸的含量相近,分别为97.50、665.80 mg/100 g和487.20 mg/100 g及92.30、642.40 mg/100 g和490.50 mg/100 g,显著高于其他3种解冻方式。不同解冻方式对炝蟹蟹肉中呈味核苷酸作用显著,超声和脉冲磁场辅助解冻蟹肉中呈味核苷酸具有较高的含量,以脉冲磁场辅助解冻蟹肉中等效鲜味浓度最高(44.38 g/100 g)。因此,脉冲磁场辅助解冻最适宜作为冻藏炝蟹的解冻方式。

Enhancing the thermoelectric transport properties of conductive polymer materials has been a long-term challenge, in spite of the success seen with molecular doping strategies. However, the strong coupling between the thermopower and the electrical conductivity limits thermoelectric performance. Here, we use polaron interfacial occupied entropy engineering to break through this intercoupling for a PEDOT:PSS(poly（3,4-ethylenedioxythiophene）-poly（4-styrenesulfonate）) thin film by using photochromic diarylethene（DAE） dopants coupled with UV-light modulation. With a 10-fold enhancement of the thermopower from 13.5 μV K-1to 135.4 μV K-1and almost unchanged electrical conductivity, the DAE-doped PEDOT:PSS thin film achieved an extremely high power factor of 521.28 μW m-1K-2from an original value of 6.78 μW m-1K-2. The thermopower was positively correlated with the UV-light intensity but decreased with increasing temperature, indicating resonant coupling between the planar closed DAE molecule and PEDOT. Both the experiments and theoretical calculations consistently confirmed the formation of an interface state due to this resonant coupling. Interfacial entropy engineering of polarons could play a critical role in enhancing the thermoelectric performance of the organic film.

Structural variations(SVs)are a feature of plant genomes that has been largely unexplored despite their significant impact on plant phenotypic traits and local adaptation to abiotic and biotic *** this study,we employed woolly grape(Vitis retordii),a species native to the tropical and subtropical regions of East Asia with both coastal and inland habitats,as a valuable model for examining the impact of SVs on local *** assembled a haplotype-resolved chromosomal reference genome for woolly grape,and conducted population genetic analyses based on whole-genome sequencing(WGS)data from coastal and inland *** demographic analyses revealed recent bottlenecks in all populations and asymmetric gene flow from the inland to the coastal *** total,1,035 genes associated with plant adaptive regulation for salt stress,radiation,and environmental adaptation were detected underlying local selection by SVs and SNPs in the coastal population,of which 37.29% and 65.26% were detected by SVs and SNPs,*** genes such as FSD2,RGA1,and AAP8 associated with salt tolerance were found to be highly differentiated and selected during the process of local adaptation to coastal habitats in SV *** study highlights the importance of SVs in local adaptation;candidate genes related to salt stress and climatic adaptation to tropical and subtropical environments are important genomic resources for future breeding programs of grapevine and its rootstocks.

The light-driven material-microorganism biohybrid system has the potential to transfer solar energy for chemical ***,few studies have reported the construction of biohybrid systems using light-responsive materials with nonmodel strains that have been widely used in practical industrial production for value-added chemicals,especially with regard to the mechanism of action of photogenerated charges in the cytoplasm,probably due to the complexity of their anabolic ***,a biohybrid system as a research mode was constructed by electrostatically self-assembling a highly efficient light-harvesting material of graphite-phase nitrided carbon(g-C_(3)N_(4))nanosheets with nonmodel strains(Phaffia rhodozyma)for synthesis of nutritional chemical *** biohybrid interface enabled efficient separation,transfer,and transport of photogenerated charges from g-C_(3)N_(4) into the interior of ***,which improved the substance metabolism and the energy metabolism of ***,photogenerated charges can significantly promote the accumulation of precursors along the astaxanthin anabolic pathway and enhance the cytoplasmic redox environment and ATP levels in the interior of ***,even under adverse conditions(such as enzyme inhibitors),thus increasing the yield of astaxanthin compared to the traditional culture of *** study not only provides new ideas for converting solar energy into value-added chemicals,but it also provides guidance for regulating microbial synthesis plants.

臭氧(O_(3))浓度升高危害小麦生长,现有研究尚不清楚推广新的小麦品种能否减缓O_(3)对籽粒产量、农田蒸散和水分利用效率的负面效应,且研究O_(3)浓度升高对麦田水分利用效率的影响对农业用水管理具有重要意义。该研究利用完全开放式O_(3)浓度升高平台(O_(3)-FACE),通过连续观测农田小气候特征,结合能量平衡法分析O_(3)浓度升高对麦田水热通量、小麦籽粒产量和田间水分利用效率的影响。结果表明:O_(3)浓度升高显著降低了开花后第32天小麦旗叶的叶绿素含量,降低了乳熟–成熟期的麦田潜热通量平均值和正午峰值,但影响幅度较小。O_(3)浓度升高对麦田平均水热通量及其分配无显著影响,也未影响籽粒产量、产量组成和田间水分利用效率。不同于10年前江淮地区主推的扬辐麦2号、烟农19和嘉兴002等品种,当前推广的农麦88表现出极强的O_(3)抗性。在O_(3)浓度不断升高背景下,种植农麦88有助于减缓O_(3)浓度升高对田间蒸散和水分利用效率的影响。
