Network-based pharmacology and experimental validation to explore the mechanism of action of the Jiawei Pentongling formula(加味盆痛灵方) for the treatment of endometriosis-related pain

OBJECTIVE: To investigate the effects and mechanisms of the Jiawei Pentongling formula （加味盆痛灵方， JWPTL） in treating endometriosis-related pain using network pharmacology study and experimental ***: Active ingredients and relevant targets of JWPTL， as well as genes for endometriosis-related pain， were collected from public databases. Prediction of core targets and pathways of JWPTL against pain associated with endometriosis by protein-protein interaction （PPI） network work， gene ontology （GO）， and Kyoto Encyclopedia of Genes and Genomes （KEGG） functional enrichment analysis. The Sprague-Dawley rat endometriosis model was constructed using the autologous endosomal transplantation method， and the successfully modeled rats were randomly divided into the model group and the JWPTL group， with 8 rats in each group. Another 8 rats were set up in the sham group. Rats in the JWPTL group used the rectal delivery method with the addition of JWPTL (7.77 g·kg-1·d-1) once a day for 28 d. Rats in the model and sham-operated groups were given equal amounts of saline using the same administration method. The 50% paw withdrawal threshold （PWT） of the rats was measured at different time points. After the intervention， the expression of phosphatidylinositol 3-kinase （PI3K） and protein kinase B （Akt） proteins and mRNA in endometriotic tissues was detected by immunohistochemistry and reverse transcription-polymerase chain reaction. RESULTS: From GeneCards， Online Mendelian Inheritance in Man and other databases， a total of 964 endometriosis （EMs） -related pain targets were screened， 142 active ingredients of JWPTL， 605 targets， and 221 potential targets were obtained by intersection of Venn diagram; 44 core targets were identified by constructing PPI network. Morphological Classification of Infrared Galaxies Based on WISE

Gas therapy is emerging as a highly promising therapeutic strategy for cancer ***,there are limitations,including the lack of targeted subcellular organelle accuracy and spatiotemporal release precision,associated with gas *** this study,we developed a series of photoactivatable nitric oxide(NO)donors NRh-R-NO(R=Me,Et,Bn,iPr,and Ph)based on an N-nitrosated upconversion luminescent rhodamine *** the irradiation of 808 nm light,only NRh-Ph-NO could effectively release NO and NRh-Ph with a significant turn-on frequency upconversion luminescence(FUCL)signal at 740 nm,ascribed to lower N-N bond dissociation *** also investigated the involved multistage near-infrared-controlled cascade release of gas therapy,including the NO released from NRh-Ph-NO along with one NRh-Ph molecule generation,the superoxide anion O_(2)^(⋅−)produced by the photodynamic therapy(PDT)effect of NRh-Ph,and highly toxic peroxynitrite anion(ONOO‒)generated from the co-existence of NO and O_(2)^(⋅−).After mild nano-modification,the nanogenerator(NRh-Ph-NO NPs)empowered with superior biocompatibility could target *** an 808 nm laser irradiation,NRh-Ph-NO NPs could induce NO/ROS to generate RNS,causing a decrease in the mitochondrial membrane potential and initiating apoptosis by caspase-3 activation,which further induced tumor immunogenic cell death(ICD).In vivo therapeutic results of NRh-Ph-NO NPs showed augmented RNS-potentiated gas therapy,demonstrating excellent biocompatibility and effective tumor inhibition guided by real-time FUCL ***,this versatile strategy defines the targeted RNS-mediated cancer therapy.

The environmental magnetic proxies of stalagmites hold significant potential for reconstructing regional hydroclimate changes by revealing the content and grain size of magnetic particles within stalagmites. In this study， we present the contents and grain sizes of magnetic particles within a stalagmite SZ-1， from Shizhu Cave in southwestern China from 70.4 to 22.3 thousand years ago (ka) during the last glacial period. Specifically， the parameters IRMsoft， soil-derived magnetic minerals， and ARM/SIRM (anhysteretic remanent magnetization/saturation isothermal remanent magnetization)， the ratio of fine magnetic particles to total ferrimagnetic particles preserved in stalagmite SZ-1， indicate the fluctuation of regional precipitation. Obvious half-precessional cycles are evident in these two proxies， indicating that hydroclimatic variations in southwestern China may predominantly arise from the heat and moisture transported from tropical oceans. These variations are likely influenced by shifts in the Intertropical Convergence Zone and fluctuations in the Asian summer monsoon.
