黄秋葵(Abelmoschus esculentus L.)是锦葵科秋葵属的一年生或二年生草本植物,营养价值高,已成为深受欢迎一种蔬菜,在亚热带和热带国家和地区均有栽培。其未成熟的豆荚是作为蔬菜食用部位,也用做工业原料制为纤维和种子粉末。印度的秋葵产量为550万吨,是全球秋葵生产的领先国家。它是人体膳食均衡的重要来源,含有维生素,蛋白质,矿物质,并提供预防癌症,糖尿病,溃疡和肝炎的重要化合物。最近有报道其多汁豆荚和粉状种子中存在有益健康的植物化学物质和酚类化合物。然而,秋葵的生长及其产量受到各种环境胁迫的影响(包括生物和非生物胁迫)而造成巨大的经济损失。基因工程和植物遗传转化可以将来自细菌和植物的抗性基因转移到有价值的作物中,以使作物在胁迫条件下有效生长和产量。黄秋葵是最难进行遗传转化的作物之一,主要原因是其体外再生体系难以建立。因此,本研究的目的是开发一种有效的体外再生系统,以黄秋葵绿色栽培品种“祥福”为材料进行了两项不同的体外再生研究,通过降低改良的MS培养基的氮素水平研究胚性愈伤组织的形成,间接诱导子叶和下胚轴外植体芽的形成。通过研究硝酸银(AgNo3)与细胞分裂素组合用于直接诱导子叶节外植体形成芽。1.降低培养基中的氮水平,诱导黄秋葵高频再生胚性愈伤组织的形成。本研究旨在探讨降低MS培养基中氮素水平对黄秋葵胚性愈伤组织形成及高效再生体系的影响。以下胚轴和子叶为外植体,研究了黄秋葵绿色品种“祥富”组织培养体系。通过含有不同水平的无机氮(KN03和NH4N03比例为1:2,含量为30,40,50和60 mM)的MS培养基以及生长素和细胞分裂素的组合,在两种外植体诱导愈伤组织的同时,添加1.0 mgL-1 BAP + 1.5 mgL-1 2,4-D的MS培养基在下胚轴上诱导愈伤组织的最高频率为71%;而在添加1.5mgL-1NAA + 0.5mgL-1 BAP的培养基的子叶外植体诱导愈伤组织的最高频率为58%;以含有40 mM总氮水平的改良MS培养基的效果最佳。在添加有1.5 mgL-1 KIN + 0.5 mgL-1 IBA和40 mM总氮水平的培养基中,下胚轴外植体上每个愈伤组织诱导3.5个芽形成,芽诱导率为66.5%;在添加有30 mM总氮水平和1.5 mgL-1 BAP + 0.5 mgL-1 NAA的培养基中,再生芽上每愈伤组织诱导芽数为2.95,芽诱导率为49.50%。高氮水平60 mM由于形成的愈伤组织白色致密,仅能诱导产生0.92个芽,芽诱导率仅9.5%。因此,下胚轴为外植体的诱导效果较佳。与标准MS培养基相比,氮素水平低的MS培养基有利于改善愈伤组织结构,形成具有芽诱导高的胚性愈伤组织。添加有2.0 mgL-1 IBA和100 mgL-1活性炭的1/2 MS培养基上生根率最高,为79%。将幼苗转移到含有蛭石和泥炭土(1:1)的无菌塑料袋中,在湿度充分的环境生长,植株存活率为85%。本研究将为黄秋葵的高效再生和转化体系的建立奠定基础。2.硝酸银(AgN03)有效诱导了黄秋葵子叶节外植体的芽再生。黄秋葵的再生频率在很大程度上取决于其遗传结构及其顽固性质。直接再生有可能在内部和外部因素的影响下改善植物遗传基因,用于遗传转化。在本研究开发了通过子叶节外植体进行增殖的黄秋葵高效再生体系。此外,还进行了硝酸银(AgN03)与细胞分裂素如6-苄基氨基嘌呤(BAP)和激动素(KIN)组合对增殖的影响研究。在添加***-1 BAP的MS培养基上获得芽再生频率最高,为75%。在添加1.5 mgL-1 BAP + 1.5 mgL-KIN + 3.0 mgL-1 AgN03的MS培养基上最有利于芽的增值,芽增殖频率为85%,每个外植体有6.75芽,平均长度为3.75 cm;而不含AgN03,1.5 mgL-1 BAP+1.5mg-1 KIN组合的每个外植体则产生具有2.3个芽,芽的长度为3.75 cm,芽增殖频率为53%。在添加有1.5 mgL-1 BAP + 1.5 mgL-1 GA3(赤霉酸)的MS培养基上芽增长效果最佳,芽长为5.60 cm。在1/2 MS培养基+1.0 mgL-1 IBA(吲哚3-丁酸)+ 200 mgL-1 AC(活性炭)上生根效果最佳,生根率为83%,根数5.20个,根长4.8 cm。在25±2 ℃和70%相对湿度生长室生长良好,植株存活率为87%。在含有蛭石和泥炭土(1:1)的塑料袋中幼苗生长良好。用芽茎段扩繁到再生成植株的整个过程仅需3.5至4.0个月。这种快速和可重复黄秋葵的高频再生将是进一步遗传转化的有利工具。综上所述,本研究为秋葵的体外再生和高频增殖提供了新的有价值的组培体系。培养基的营养和外植体类型是影响体外再生频率的主要因素。MS培养基中的氮水平为40 mM有利于下胚轴外植体高频再生

The cases and deaths due to Crimean-Congo haemorrhagic fever(CCHF)virus commonly known as Congo virus(fatality rate 15%)have been reported throughout Pakistan from the last few years especially during religious occasion,*** annual increase in death rates due to CCHF demonstrate the importance of awareness of Congo fever at academia as well as public *** symptoms of Congo fever which appear one to nine days after tick bite,include sudden high fever,muscle aches,abdominal pain,headache,dizziness,sore eyes,jaundice,mood swings,confusion,aggression,and sensitivity to *** other signs include sore throat,joint pain,vomiting,diarrhea,hemorrhages,and bleeding from skin and large *** Infection has been reported in many species of wild as well as domestic animals including hares,cattle,sheep,goats,dogs,mice and *** least 31 species of Hyalomma,Boophilus,Rhipicephalus,Dermacentor(Ixodidae:hard ticks)act as vector of CCHF in which transovarial,transstadial and venereal transmission *** virus attacks the immune system of the host and influences the immune *** Congo fever virus can be isolated from blood,plasma and many body tissues(kidneys,liver,spleen,lungs,brain and bone marrow).Mice inoculation,enzyme-linked immunosorbent assay(ELISA),reverse transcription polymerase chain reaction(RT-PCR)can be used for detection of the ***,IgM and IgG antibodies against CCHFV can also be detected and *** of general public,tick control with acaricides,use of anti-CCHFV immunoglobulin,usage of approved repellents to prevent tick bites,wearing neutral-coloured garments,application of a permethrin spray to the clothing,avoiding tall grasses and shrubs,applying sunscreen,avoiding direct contact with the blood or tissues of animals are the factors for successful prevention of the infection.

摘要基于百香果转录组数据筛选ABC转运蛋白基因，分析其序列特征、编码蛋白理化性质以及不同成员在果实发育阶段的表达模式。以本地blast和二级结构数据ABC＿tran（PF00005）筛选ABC转运蛋白，对鉴定到的ABC转运蛋白基因进行蛋白基本理化性质、亚细胞定位、保守结构域以及基序分析，并利用实时定量PCR技术研究17个基因在不同果实发育阶段的表达模式。结果显示：总计获得80个百香果ABC转运蛋白基因，它们分为8个亚族，G、B、C、F、I、D、A和E亚家族，分别有23、18、18、7、7、3、2和2个成员。亚细胞定位预测结果显示: ABC转运蛋白主要定位于质膜（62.5%）、液泡膜（11.3%）和细胞质（10%）等部位。保守结构域及基序分析显示同一亚家族成员结构域以及基序种类及数量相似。ABC转运蛋白基因在果实发育阶段的表达各异，PeABCA1、PeABCB14和PeABCD2等基因在幼果期、绿果期以及成熟期，紫果的相对表达显著高于黄果，而在转色期的表达，黄果显著高于紫果；基因PeABCG22只有在转色期的相对表达紫果显著高于黄果。本研究表明ABC转运蛋白基因在果实的生长发育中有一定作用，可能在果实生长发育中生长素运输、蜡质转运及木质素合成等生物过程中发挥作用。为进一步研究ABC转运蛋白基因在百香果发育中的作用奠定了一定基础。（图4 表2 参32）

Enhanced catalytic features of sol-gel immobilized MnP isolated from solid state culture of Pleurotus ostreatus IBL-02

An indigenous and industrially important manganese peroxidase （MnP） isolated from solid state culture of Pleurotus ostreatus IBL-02 was 2.1-fold purified and immobilized using sol-gel matrix of trimethoxysilane and proplytetramethoxysilane. Lower KM, higher Vmax, hyper-activation, enhanced acidic and thermal stability up to 70 ℃, were the improved]enhanced catalytic features of the presently sol-gel immobilized MnP. A maximum of 100% decolorization was achieved within a short time period for the different real dyes containing textile industry effluents i.e. Nishat textile （NIT）, K＆N textile （KIT）, Sitara textile （SIT）, and Crescent textile （CRT）.

A probabilistic model of quantum states for classical data security

The phenomenal progress of quantum information theory over the last decade has substantially broadened the potential to simulate the superposition of states for exponential speedup of quantum algorithms over their classical ***,the conventional and modern cryptographic standards(encryption and authentication)are susceptible to Shor’s and Grover’s algorithms on quantum *** significant improvement in technology permits consummate levels of data protection by encoding classical data into small quantum states that can only be utilized once by leveraging the capabilities of quantum-assisted classical *** the frequent data breaches and increasingly stringent privacy legislation,we introduce a hybrid quantum-classical model to transform classical data into unclonable states,and we experimentally demonstrate perfect state transfer to exemplify the classical *** alleviate implementation complexity,we propose an arbitrary quantum signature scheme that does not require the establishment of entangled states to authenticate users in order to transmit and receive arbitrated states to retrieve classical *** consequences of the probabilistic model indicate that the quantum-assisted classical framework substantially enhances the performance and security of digital data,and paves the way toward real-world applications.

Hierarchical medical diagnosis approach for COVID-19 based on picture fuzzy fairly aggregation operators

In Pakistan, a hierarchical healthcare system is an efficient way of addressing the issueof limited and insufficient healthcare services. Identifying the various degrees of diseasebased on the doctor's diagnosis is an important step in developing the hierarchical healthcaretreatment structure. This research presents a framework for dealing with the issueof diagnosis values presented as "picture fuzzy numbers (PFNs)". Specifically, the goal ofthis study is to establish some innovative operational laws and "aggregation operators"(AOs) in a picture fuzzy environment. In this regard, we proposed some new neutral orfair operational laws that incorporate the concept of proportional distribution in orderto achieve a neutral or fair remedy to the positive, neutral and negative aspects ofPFNs. Based on the developed operational laws, we proposed the "picture fuzzy fairlyweighted average operator" and the "picture fuzzy fairly ordered weighted averagingoperator". Compared to previous techniques, the proposed AOs provide more generalizedand reliable. Furthermore, using proposed AOs with multiple decision-makers andpartial weight information under PFNs, a "multi-criteria decision-making" algorithmis developed. Finally, we provide an example to show how the novel approach can aidhierarchical treatment systems. This is essential for merging the healthcare capabilitiesof the general public and optimizing the medical care system's service performance.

Design optimization of composite egg-shaped submersible pressure hull for minimum buoyancy factor

This paper describes a design optimization study of the composite egg-shaped submersible pressure hull employing optimization and finite element analysis(FEA)tools as a first attempt to provide an optimized design of the composite egg-shaped pressure hull for manufacturing or further investigations.A total of 15 optimal designs for the composite egg-shaped pressure hull under hydrostatic pressure are obtained in terms of fibers’angles and the number of layers for 5 lay-up arrangements and 3 unidirectional(UD)composite *** optimization process is performed utilizing a genetic algorithm and FEA in *** minimization of the buoyancy factor eB:FT is selected as the objective for the optimization under constraints on both material failure and buckling *** buckling analysis is conducted for one optimal design considering both geometric nonlinearity and imperfections.A sensitivity study is also conducted to further investigate the influence of the design variables on the optimal design of the egg-shaped pressure hull.
