Small extracellular vesicles derived from human induced pluripotent stem cell-differentiated neural progenitor cells mitigate retinal ganglion cell degeneration in a mouse model of optic nerve injury

Several studies have found that transplantation of neural progenitor cells(NPCs)promotes the survival of injured ***,a poor integration rate and high risk of tumorigenicity after cell transplantation limits their clinical *** extracellular vesicles(sEVs)contain bioactive molecules for neuronal protection and *** studies have shown that stem/progenitor cell-derived sEVs can promote neuronal survival and recovery of neurological function in neurodegenerative eye diseases and other eye *** this study,we intravitreally transplanted sEVs derived from human induced pluripotent stem cells(hiPSCs)and hiPSCs-differentiated NPCs(hiPSC-NPC)in a mouse model of optic nerve *** results show that these intravitreally injected sEVs were ingested by retinal cells,especially those localized in the ganglion cell *** with hiPSC-NPC-derived sEVs mitigated optic nerve crush-induced retinal ganglion cell degeneration,and regulated the retinal microenvironment by inhibiting excessive activation of *** analysis further revealed that hiPSC-NPC derived sEVs transported neuroprotective and anti-inflammatory miRNA cargos to target cells,which had protective effects on RGCs after optic nerve *** findings suggest that sEVs derived from hiPSC-NPC are a promising cell-free therapeutic strategy for optic neuropathy.

Transcriptome analysis of adherens junction pathway-related genes after peripheral nerve injury

The neural regeneration process is driven by a wide range of molecules and pathways. Adherens junctions are critical cellular junctions for the integrity of peripheral nerves. However, few studies have systematically characterized the transcript changes in the adherens junction pathway following injury. In this study, a rat model of sciatic nerve crush injury was established by forceps. Deep sequencing data were analyzed using comprehensive transcriptome analysis at 0, 1, 4, 7, and 14 days after injury. Results showed that most individual molecules in the adherens junctions were either upregulated or downregulated after nerve injury. The m RNA expression of ARPC1 B, ARPC3, TUBA8, TUBA1 C, CTNNA2, ACTN3, MET, HGF, NME1 and ARF6, which are involved in the adherens junction pathway and in remodeling of adherens junctions, was analyzed using quantitative real-time polymerase chain reaction. Most of these genes were upregulated in the sciatic nerve stump following peripheral nerve injury, except for CTNNA2, which was downregulated. Our findings reveal the dynamic changes of key molecules in adherens junctions and in remodeling of adherens junctions. These key genes provide a reference for the selection of clinical therapeutic targets for peripheral nerve injury.

Density change is ubiquitous in phase transformation, and it can induce melt convection which strongly influences the crystal growth. Here, an anisotropic lattice Boltzmann-phase-field method was extended to predict the dendritic growth under the shrinkage or expansion melt convection by density change induced. A novel LB equation with an anisotropic coefficient was constructed to model the advancement of ordering parameter, coupling with the passive scalar LB equation for convective and diffusive heat transfer during phase transition. We studied dendritic growth and shape selection with melt convection induced by density change in crystal growth. Results show that the melt convection induced by density change affects strongly the dendritic growth. The shrinkage flow results in a higher tip velocity while the expansion flow leads to a slower one. Predicted Péclet number with respect to the relative density change was compared with an analytical solution. Moreover, the modified selection parameter has been verified by numerical simulations.

Soil Acidification of Alfisols as Influenced by Tea Cultivation in Eastern China

Soil acidification is an important process in land degradation around the world as well as in *** of Alfisols was investigated in the tea gardens with various years of tea cultivation in the eastern *** of tea plants caused soil acidification and soil acidity increased with the increase of tea cultivation *** pH of composite samples from cultivated layers decreased by 1.37,1.62 and 1.85,respectively,after 13,34 and 54 years of tea plantation,as compared to the surface soil obtained from the unused *** acidification rates at early stages of tea cultivation were found to be higher than those at the later *** acidification rate for the period of 0-13 years was as high as 4.40 kmol H + ha ?1 year ?1 for the cultivated layer *** acidification induced the decrease of soil exchangeable base cations and base cation saturation and thus increased the soil exchangeable *** acidification also caused the decrease of soil cation exchange capacity,especially for the 54-year-old tea *** acidification induced by tea plantation also led to the increase of soil exchangeable Al and soluble Al,which was responsible for the Al toxicity to plants.

In this paper,the spherical particles growth during lithium electrodeposition was investigated by directly solving the governing equations based on the Landau transformation *** basic growth kinetic characteristics of a spherical particle during electrodeposition was *** results show that the dynamic oscillation of the growth velocity occurs during the spherical particle *** was found from numerical simulations that applied electrical potential difference,electrolyte concentration,and diffusion coefficient are 3 main factors influencing the spherical growth and the existence of the dynamic oscillation state of the growth velocity during *** increase in both the applied electrical potential difference and the electrolyte concentration can lead to the increase of the growth velocity of the spherical particle,while the growth velocity is independent of the diffusion ***,it was found that the wavelength and the amplitude of the dynamic oscillation of the growth velocity can be influenced by the applied electrical potential difference,the electrolyte concentration,and the diffusion *** determined that the dynamic competition between electrochemical reactions and ion transport in the electrodeposition is the reason for the existence of the oscillation of the growth velocity.

Morphological evolution of the solid-liquid interface near grain boundaries during directional solidification

Morphological evolution of the solid-liquid interface near grain boundaries has been studied during directional solidification of succinonitrile-based transparent alloys (SCN-0.9wt%DCB). Experimental results show that the grain boundary provides the starting point of morphological instability of the solid-liquid interface. The initial perturbation near the grain boundary is significantly larger than other perturbations on the interface. The initial shape of the interface and the competition between the thermal direction and preferred crystalline orientations determine the subsequent growth pattern selections. The temporal variations of the curvature radius of cell/ridge tips near the grain boundary have also been studied when the instability occurs. This process is divided into three parts. As the pulling velocity increases, dendrites at the grain boundary grow in two different directions to form a bicrystal microstructure. Side branches on either side of the dendrite exhibit different growth patterns.

Mixed surfactant washing of petroleum contaminants from the soils and sediments in the unsaturated zone

A laboratory study was conducted to evaluate the suitability of a mixed surfactant for washing oil from soils in the unsaturated zone. The commercial surfactant used for making mixed surfactant were alcohol polyethoxylate (AEO\|9, nonionic) and sodium alcohol polyethoxylated ethers sulfate (AES, anionic). Based on the properties (critical micelle concentration, hydrophilic/ lipophilic balance and surface tension) of individual surfactants and the mixed surfactant, the preferred composition of an aqueous mixed surfactant solution was determined which consisted of 0 5% AEO\|9 and 0 1% AES. The batch washing experiments have shown that the removal efficiency of oil from soils with mixed surfactant was higher than that with individual surfactants. Soil column washing results have shown that the sequence of oil removal efficiency was mixed surfactant (0.5% AEO\|9 and 0.1% AES)>AEO\|9 (0.1%)>AES (0.2%)>water, and the ratio was 1∶0 84∶0 66∶0 06. Biodegradation experiments by growth cells and resting cells were carried out to evaluate the environmental acceptability of the test surfactants. The results have shown that the surfactants were able to serve as sole carbon source for strains Pseudomonos sp. 52 and Weeksella sp. 6 which were isolated from petroleum contaminated soil. More than 90% of the parent mixed surfactants disappeared in two weeks under growth cell conditions.

Effects of simulated acid rain on fertility of litchi

The regulatory role of calcium in fertility of pollen and pistil under simulated acid rain was investigated. The germination percentage of pollen treated with acid rain of pH 4.5 was 9.42% lower than that of control, and that of pH 3.5, pH 2.5 and pH 1.5 were 22.47%, 45.49% and 71.62%, respectively. Simultaneously, the injury character of pollen was obviously observed when flowers were treated with acid rain of pH 3.5. The difference in fruit setting rate between the female flower treated with acid rain of pH 4.0 and the control was significant at p 〈 0.05. Ca（NO3 ）5 of 0.2-0.4 mmol/L could promote pollen germination under the stress of acid rain. The beneficial function was reduced when calcium concentration surpassed 0.8 mmol/L. Spraying 2 mmol/L Ca（NO3 ）5 reduced the injury of acid rain to pistil and increased fruit-setting rate significantly. The physiological importance of calcium during pollen germination and pistil development was also discussed.
