Absorbed Bioactive Compounds Replicate GuanxinⅡ-Induced Endothelium-Associated in/ex vivo Vasodilation

Rapid Antidepressant-Like Potential of Chaihu Shugan San Depends on Suppressing Glutamate Neurotransmission and Activating Synaptic Proteins in Hippocampus of Female Mice

Objective To explore the rapid antidepressant potential and the underlying mechanism of Chaihu Shugan San(CSS)in female *** Liquid chromatography mass spectrometry(LC-MS)/MS was used to determine the content of main components in CSS to determine its *** C57BL/6J mice were randomly divided into 4 groups,including control(saline),vehicle(saline),CSS(4 g/kg)and ketamine(30 mg/kg)*** were subjected to irregular stress stimulation for 4 weeks to establish the chronic mild stress(CMS)model,then received a single administration of *** hours later,the behavioral tests were performed,including open field test,tail suspension test(TST),forced swimming test(FST),novelty suppression feeding test(NSF),and sucrose preference test(SPT).Western blot analysis was used to detect the expression levels of N-methyl-D-aspartate receptor(NMDA)subtypes[N-methyl-D-aspartate receptor 1(NR1),NR2A,NR2B],synaptic proteins[synapsin1 and post synaptic density protein 95(PSD95)],and brain-derived neurotrophic factor(BDNF).Moreover,the rapid antidepressant effect of CSS was tested by pharmacological technologies and optogenetic interventions that activated glutamate receptors,*** Compared with the vehicle group,a single administration of CSS(4 g/kg)reversed all behavioral defects in TST,FST,SPT and NSF caused by CMS(Pketamine,CSS increased levels of synaptic proteins and BDNF(Pked by transient activation of NMDA receptors in the hippocampus(all P<0.01).Conclusion Rapid antidepressant effects of CSS by improving behavioral deficits in female CMS mice depended on rapid suppression of NMDA receptors and activation of synaptic proteins.

Microstructural evolution of TiAl base alloy during three-stepped thermo-mechanical treatment

A TiAl base alloy ingot with a height to diameter ratio of 2.2 was broken down by multiple step canned forging. The microstructures after every deformation and subsequent recrystallization were observed by optical microscopy. Results show that at the first step the reduction should be carefully controlled in case of double bulge and crack of the ingot. After the first annealing, recrystallization occurred at the deformed grain boundaries and inside the grain. The recrystallized microstructure is favorable for further deformation. After the second deformation and annealing, coarsening of the lamellae occurred and the microstructure became equiaxed. By the final deformation and subsequent recrystallization, the coarse lamellar colony can be refined to about 20 μm, and homogeneous microstructure was obtained from the ingot with a large initial height to diameter ratio.

Environmental endocrine disruptors,represented by bisphenol A(BPA),have been widely detected in the environment,bringing potential health risks to human ***-containing biocarbon catalyst can activate peroxymonosulfate(PMS)to degrade BPA in water,but its active sites remain ***,in this work,nitrogen-containing biochar,i.e.,C–Nedge,enriched with graphitic-N defects at the edges was prepared by one-pot co-pyrolysis of chitosan and potassium *** results showed that the C–Nedge/PMS system can effectively degrade 98%of BPA(50 mg/L).The electron transfer based non-radical oxidation mechanism was responsible for BPA *** graphitic-N doping endows biochar with strong electron transfer *** catalyst had good recovery and reuse *** catalytic oxidation was also feasible for other refractory pollutants removal and worked well for treating practical *** work may provide valuable information in unraveling the N doping configurationactivity relationship during activating PMS by biochar.

In situ direct reprogramming of astrocytes to neurons via polypyrimidine tract-binding protein 1 knockdown in a mouse model of ischemic stroke

In situ direct reprogramming technology can directly convert endogenous glial cells into functional neurons in vivo for central nervous system repair. Polypyrimidine tract-binding protein 1(PTB) knockdown has been shown to reprogram astrocytes to functional neurons in situ. In this study, we used AAV-PHP.e B-GFAP-sh PTB to knockdown PTB in a mouse model of ischemic stroke induced by endothelin-1, and investigated the effects of GFAP-sh PTB-mediated direct reprogramming to neurons. Our results showed that in the mouse model of ischemic stroke, PTB knockdown effectively reprogrammed GFAP-positive cells to neurons in ischemic foci, restored neural tissue structure, reduced inflammatory response, and improved behavioral function. These findings validate the effectiveness of in situ transdifferentiation of astrocytes, and suggest that the approach may be a promising strategy for stroke treatment.

Effect of the vortical structures on the hydrodynamic performance of a pitching hydrofoil

The objective is to study the vortical structural behaviors of a transient pitching hydrofoil and their effects on the hydrodynamic performance. The pitching motion of the hydrofoil is set to pitch up with an almost constant rate from 5° to 15° and then back to 5°, with the Reynolds number 4.4×10^(5) and the frequency 2 Hz. The results show that the main coherent structures around the pitching hydrofoil include small-scale laminar separation bubble (LSB), large-scale second vortex (SV) and trailing edge vortex (TEV) which are all vortical. The relationship between the vortical structure and the lift is investigated with the finite-domain impulse theory. It indicates that the major part of the lift is contributed by the LSB, whereas the shedding and the formation of the SV and TEV cause the fluctuation of the lift. The proper orthogonal decomposition (POD) method is applied to capture the most energetic modes, revealing that the LSB mode occupies a large amount of energy in the flow field. The dynamic mode decomposition (DMD) method accurately extracts the dominant frequency and modal characteristics, with the first mode corresponding to the mean flow, the second mode corresponding to the LSB structure and the third and fourth modes corresponding to the vortex shedding.
