本文基于转录组数据研究了新型冠状病毒感染(Coronavirus disease 2019,COVID-19)和2型糖尿病(Type 2diabetes,T2D)的相互作用机制,寻找疾病的潜在功能模块,为其治疗提供新思路。本文收集了来自GEO(Gene expression omnibus)数据库的10个COVID-19和T2D基因表达谱数据集,并按照4:6的比例分为训练集和验证集,并进行差异表达基因(Differential expression genes,DEGs)的富集分析和构建蛋白质相互作用(Protein-protein interaction,PPI)网络进行模块化分析,找到了与COVID-19和T2D最紧密相关的关键基因和蛋白质特征模块。基于各组织的DEGs交集结果,本文发现43个共有DEGs,并确定了5个关键基因(HSP90AA1、SRC、EGFR、MAPK3、CDK1),其中CDK1表现出最高的网络连接性。通过XGBoost等6种机器学习算法进行性能检验显示了关键基因对诊断COVID-19和T2D的潜在价值,并通过qPCR实验证实了关键基因在COVID-19患者和正常对照组PBMC中的表达差异。此外,本文基于模块划分算法得到了PPI网络的6个潜在功能模块,这些模块主要与糖类、脂类代谢和细胞复制相关联。最后,本文建立了miRNA-TF-mRNA调控网络,并筛选出TP53和NFIC作为调节共有基因转录的枢纽转录因子(Transcription factor,TF)。总之,本文分析了COVID-19和T2D之间共有的表达特征、生物学通路和调控因子,并探索了基因在转录和翻译中的复杂相互作用,为未来COVID-19和T2D的治疗提供了新思路。

The configurations of molecular clusters have significant impacts on their growth into fine particles in *** this paper,we explore the topology space of the structure of H2SO4·NH3 dimer with a novel sampling technique of meta-dynamics(MTD)method and ab initio molecular dynamics *** simulations are carried out at the temperatures of both 50 K and 242 K,which represent the typical high and low latitudes of *** results show that,compared with only traditional MD simulations,the structure samplings are significantly accelerated with MTD ***,more isomers of the dimer are discovered within the same simulation time *** addition,the results show that MTD is more efficient for circumstances with high temperature.

In the space Γ_n=span｛sint,cost,1,t,t2,…,t n-2 ｝, a C-Bézier basis is constructed by an integral approach. However, the C-Bézier basis is not orthogonal. For some applications, we construct an orthogonal basis based on the C-Bézier basis, which has remarkable properties similar to that of the Legendre basis. Then we derive the transformation matrices that convert the C-Bézier basis and the orthogonal basis into each other. As an example of application, we apply this orthogonal basis to the degree reduction approximation of the C-Bézier curves.

Efficacy of Short-term Dual Antiplatelet Therapy after Implantation of Second-generation Drug-eluting Stents:A Meta-analysis and Systematic Review

Objective The benefit of short-term dual antiplatelet therapy(DAPT) following second-generation drug-eluting stents implantation has not been systematically evaluated. To bridge the knowledge gap,we did a meta-analysis to assess the efficacy of ≤6 months versus ≥12 months DAPT among patients with second-generation drug-eluting stents. Methods We searched online databases and identified randomized controlled trials that assess the clinical impact of short-term DAPT(≤6 months) published before March 3,2016. The efficacy endpoints included the incidence of all-cause death,myocardial infarction,cerebrovascular accidents,and definite or probable stent thrombosis. Safety endpoint defined as major bleeding was also evaluated and discussed. Results We included 5 trials that randomized 9473 participants(49.8%,short-term DAPT duration vs. 50.2%,standard duration). A total of 9445(99.7%) patients reported the efficacy endpoints,and the safety endpoint was available from 4 studies(n=8457). There was no significant difference in efficacy endpoints between short-term and standard DAPT duration(≥12 months) [risk ratio(RR) 0.96; 95% confidence intervals(CI),0.80-1.15]. Short-term DAPT duration did not significantly increase the individual risk of all-cause death,myocardial infarction,cerebrovascular accidents,or definite or probable stent thrombosis. Although short-term DAPT obviously reduced risk of major bleeding compared with standard DAPT(RR 0.53; 95% CI,0.29-0.96),significant publication bias was found when accessing the safety endpoint of the 4 studies(Egger's test,P=0.009). Conclusions The efficacy of short-term DAPT was comparable with that of standard duration *** less than 6 months may be appropriate for patients receiving second-generation drug-eluting stents implantation.

Molecular dynamics method is applied to study the machining mechanisms of polishing based on coupling vibrations of liquid. The physical phenomena of abrasive particles bombarding on silicon monocrystal surface are simulated using Tersoff potentials. The effects of vibration parameters, particle size, incident angle and particle material are analyzed and discussed. Material removal mechanisms are studied. Deformation and embedment phenomena are found in the simulations, Bombardment will destroy the crystal structures near the impact point, and adhesion effect is responsible for final removal of material.
