Experimental studies of the effects of ZnPcS2P2-based-photodynamic therapy on bone marrow purging

Background An effective purging technique plays an important role in autologous hematopoietic stem cells transplantation Photodynamic therapy (PDT) provides a novel approach for this purpose This study dealt with the purging effects of disulfodiphthalimidomethyl phthalolcyanine zinc (ZnPcS2P2)based photodynamic therapy (ZnPcPDT) Methods Fluorescence intensity of cell extracts was measured using a fluoresence spectrophotometry The proliferative potency of K562 cells and HL60 cells was detected using MTT colorimetric assay, Typan blue dye exclusion method, colony formation test The proliferative potency of normal hematopoietic cells was evaluated using mixture colonyforming unit (CFUMix), granulocytemacrophage colonyforming unit (CFUGM), and erythrocyte colonyforming unit (CFUE) assays K562 cells were mixed with normal mononuclear cells (MNCs) at ratios of 1∶100 and 1∶1000 for creating the model of simulated remission bone marrow Colony formation test and nestedRTPCR were carried out to detect the residual K562 cells in cell mixture Results After a 5hour incubation with ZnPcS2P2, the content of ZnPcS2P2 in normal MNCs was the lowest value At the same time, the content in K562 cells and HL60 cells was very high Therefore, the time point was selected as the optimal one for irradiating the cell suspensions ZnPcPDT could significantly kill proliferative K562 cells and HL60 cells in a dosedependent manner At the concentration of 10 μg/ml, the inhibitory rate of ZnPcPDT on the colony formation was 100% for K562 cells, 897% for HL60 cells 025 μg/ml ZnPcPDT could completely photoinactivate residual K562 cells in the simulated remission bone marrow Under an identical condition, the inhibitory rates of CFUMix, CFUGM, CFUE were 180%, 186%, and 178% respectively Conclusion ZnPcPDT appears to be a promising approach for bone marrow purging

Recombinant fibronectin polypeptide antagonizes hepatic failure induced by endotoxin in mice

AIM: To study the preventive effect of recombinant human fibronectin (rhFN) polypeptide on hepatic failure in- duced by endotoxin in mice. METHODS: A cDNA fragment coding for Ile1363-Tyr1725 of human FN was inserted into the PinPoint Xa-3 plasmid, and the constructed plasmid was transformed into E coli BL21 (DE3) cells, and then the expression of rhFN polypeptide in DE3 cells was identified through SDS-PAGE. The target protein from the supernatant of bacteria lysate was purified through biotin-affinity chromatography. The bioactivity of the purified rhFN polypeptide was determined with cell adhesiveactivity. The survival rate was observed in endotoxemia mice injected with rhFN polypeptide. The tissue damage in hepatocyte was detected using histology, ultrastructure, and DNA fragmentation assay. RESULTS: The expression of rhFN polypeptide reached approximately 20 % of the total cellular protein. The adhesion ability of rhFN polypeptide was concentration-dependent. The value of EC50 was 0.8 nmol/L. The survival rate of endotoxemia mice sensitized by D-galactosamine (D-GalN) was 60 % in rhFN polypeptide treated group, while that of endotoxemia mice sensitized by D-GalN was 20 % in the control group (P<0.01). Histopathology showed that less necrosis occurred on the hepatocyte of endotoxemia mice in- jected with rhFN polypeptide compared with saline control. Ultrastructure and DNA fragmentation assay showed that no apoptotic hepatocyte was observed in the liver of rhFN-treated endotoxemia mice. CONCLUSION: Re- combinant fibronectin polypeptide antagonizes hepatic failure induced by endotoxin in mice.

Mitochondria-dependent apoptosis induced by a novel amphipathic photochemotherapeutic agent ZnPcS2P2 in HL60 cells

Aim:To investigate the mechanism underlying the killing effects of a novelamphipathic photosensitizer,disulfonated diphthalimidomethyl phthalocyaninezinc(ZnPcS2P2),mediated photodynamic therapy(ZnPc-PDT)in human myelog-enous leukemia HL60 ***:After incubation for 5 h with 0.5 μmol/LZnPcS2P2,the HL60 cells were exposed to a light source of 670 nm ***,the cells were detected at different time intervals after *** char-acteristics of apoptosis were detected by observation of ultrastructure assay,DNA fragmentation assay and terminal deoxynucleotidyl transferase deoxyuridinenick-end labeling method(TUNEL).Mitochondria-dependent apoptosis was de-termined by the detection of mitochondrial membrane potential(Δψm),activitiesof caspase family protease and of caspase-3,cytosol cytochrome *** Bcl-2and Bax were detected by immunoblot ***:Evident characteristicsof apoptosis were observed post-ZnPc-PDT with ultrastructure assay,DNA frag-mentation assay and TUNEL *** assay showed that apoptotic ratesin the cells collected from 6 h,12 h and 24 h after PDT were 9.6%,24.4%,and 33.0%,***60 cells underwent mitochondria-dependent apoptosis as a re-sult of cytochrome c release from mitochondria into cytosol accompanied by areduction of Δψ*** activities of caspase family protease and of caspase-3were ***,ZnPc-PDT could remarkably down-regulate the Bcl-2pro-apoptotic protein and up-regulate the anti-apoptotic Bax ***:ZnPc-PDT could induce mitochondria-dependent apoptosis in HL60 cells.

There is considerable potential for integrating transarterial chemoembolization(TACE),programmed death-(ligand)1(PD-[L]1)inhibitors,and molecular targeted treatments(MTT)in hepatocellular carcinoma(HCC).It is necessary to investigate the therapeutic efficacy and safety of TACE combined with PD-(L)1 inhibitors and MTT in real-world *** this nationwide,retrospective,cohort study,826 HCC patients receiving either TACE plus PD-(L)1 blockades and MTT(combination group,n=376)or TACE monotherapy(monotherapy group,n=450)were included from January 2018 to May *** primary endpoint was progression-free survival(PFS)according to modified *** secondary outcomes included overall survival(OS),objective response rate(ORR),and *** performed propensity score matching approaches to reduce bias between two *** matching,228 pairs were included with a predominantly advanced disease *** PFS in combination group was 9.5 months(95%confidence interval[CI],8.4-11.0)versus 8.0 months(95%CI,6.6-9.5)(adjusted hazard ratio[HR],0.70,P=0.002).OS and ORR were also significantly higher in combination group(median OS,19.2[16.1-27.3]vs.15.7 months[13.0-20.2];adjusted HR,0.63,P=0.001;ORR,60.1%vs.32.0%;P<0.001).Grade 3/4 adverse events were observed at a rate of 15.8%and 7.5%in combination and monotherapy groups,*** results suggest that TACE plus PD-(L)1 blockades and MTT could significantly improve PFS,OS,and ORR versus TACE monotherapy for Chinese patients with predominantly advanced HCC in real-world practice,with an acceptable safety profile.

Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains *** A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in china. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of *** Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two *** Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. Women with comorbidities were more likely to have severe disease, while men with comorbidities were more likely to have unfavorable treatment outcomes.

Intrahepatic cholangiocarcinoma(iCCA)can originate from the large bile duct group(segment bile ducts and area bile ducts),small bile duct group(septal bile ducts and interlobular bile ducts),and terminal bile duct group(bile ductules and canals of Hering)of the intrahepatic biliary tree,which can be histopathological corresponding to large duct type iCCA,small duct type iCCA and iCCA with ductal plate malformation pattern,and cholangiolocarcinoma,*** challenge in pathological diagnosis of above subtypes of iCCA falls in the distinction of cellular morphologies,tissue structures,growth patterns,invasive behaviors,immunophenotypes,molecular mutations,and surgical *** these reasons,this expert consensus provides nine recommendations as a reference for standardizing and refining the diagnosis of pathological subtypes of iCCA,mainly based on the 5th edition of the World Health Organization Classification of Tumours of the Digestive System.
