The angiotensin-converting enzyme 2(ACE2)is a primary cell surface viral binding receptor for SARS-CoV-2,so finding new regulatory molecules to modulate ACE2 expression levels is a promising strategy against *** the current study,we utilized islet organoids derived from human embryonic stem cells(hESCs),animal models and COVID-19 patients to discover that fibroblast growth factor 7(FGF7)enhances ACE2 expression within the islets,facilitating SARS-CoV-2 infection and resulting in impaired insulin *** hESC-derived islet organoids,we demonstrated that FGF7 interacts with FGF receptor 2(FGFR2)and FGFR1 to upregulate ACE2 expression predominantly inβ*** upregulation increases both insulin secretion and susceptibility ofβcells to SARS-CoV-2 *** FGFR counteracts the FGF7-induced ACE2 upregulation,subsequently reducing viral infection and replication in the ***,retrospective clinical data revealed that diabetic patients with severe COVID-19 symptoms exhibited elevated serum FGF7 levels compared to those with mild ***,animal experiments indicated that SARS-CoV-2 infection increased pancreatic FGF7 levels,resulting in a reduction of insulin concentrations in *** together,our research offers a potential regulatory strategy for ACE2 by controlling FGF7,thereby protecting islets from SARS-CoV-2 infection and preventing the progression of diabetes in the context of COVID-19.

Background:The impact of corticosteroids on humoral responses in coronavirus disease 2019(COVID-19)sur-vivors during the acute phase and subsequent 6-month period remains *** study aimed to determine how the use of corticosteroids influences the initiation and duration of humoral responses in COVID-19 survivors 6 months after infection ***:We used kinetic antibody data from the lopinavir-ritonavir trial conducted at Jin Yin-Tan Hospital in January 2020,which involved adults hospitalized with severe COVID-19(LOTUS,ChiCTR2000029308).Anti-body samples were collected from 192 patients during hospitalization,and kinetic antibodies were monitored at all available time points after ***,plasma samples were collected from 101 COVID-19 survivors for comprehensive humoral immune measurement at the half-year follow-up *** main focus was comparing the humoral responses between patients treated with systemic corticosteroid therapy and the non-corticosteroid ***:From illness onset to day 30,the median antibody titre areas under the receiver operating characteristic curve(AUCs)of nucleoprotein(N),spike protein(S),and receptor-binding domain(RBD)immunoglobulin G(IgG)were significantly lower in the corticosteroids *** AUCs of N-,S-,and RBD-IgM as well as neutralizing antibodies(NAbs)were numerically lower in the corticosteroids group compared with the non-corticosteroid ***,peak titres of N,S,RBD-IgM and-IgG and NAbs were not influenced by *** 6-month follow-up,we observed a delayed decline for most binding antibodies,except N-IgM(𝛽−0.05,95%CI[−0.10,0.00])in the corticosteroids group,though not reaching statistical *** significant difference was observed for ***,for the half-year seropositive rate,corticosteroids significantly accelerated the decay of IgA and IgM but made no difference to N-,S-,and RBD-IgG or ***,corticosteroids group showed a trend towards d

目的分析子痫前期(PE)患者血清腺苷活化蛋白激酶(AMPK)水平与可溶性血管内皮生长因子受体-1(sFlt-1)及氧化应激的相关性。方法选取2020年6月至2023年4月在成都医学院第一附属医院治疗的67例PE患者为试验组,选择本院同期住院的正常分娩孕妇50名为对照组。比较两组AMPK、sFlt-1和氧化应激反应水平[丙二醛(MDA)、超氧化物歧化物(SOD)];分析试验组不同严重程度的PE患者AMPK、sFlt-1、MDA、SOD水平差异;利用多变量逻辑回归方法对妊娠女性发病风险因子进行分析;分析试验组患者AMPK、sFlt-1与氧化应激的相关性。结果试验组患者AMPK、sFlt-1、MDA水平均高于对照组,SOD水平低于对照组(P<0.05);轻度PE组的AMPK、sFlt-1、MDA水平均低于重度PE组,SOD高于重度PE组(P<0.05);两组妊娠期患糖尿病情况和AMPK、sFlt-1、MDA、SOD水平比较,差异有统计学意义(P<0.05);经多元Logistic回归分析显示,妊娠期合并糖尿病和AMPK、sFlt-1、MDA、SOD水平是影响孕妇发生PE的危险因素(P<0.05);Pearson相关性分析显示,PE患者AMPK、sFlt-1与MDA水平呈正相关,与SOD水平呈负相关(P<0.05)。结论 PE患者AMPK、sFlt-1水平出现异常升高,AMPK、sFlt-1水平与氧化应激指标存在一定相关性。

Enterovirus 71(EV71)poses a serious threat to human health,with scattered outbreaks *** are several vaccines against a few EV71 strains but no efficient drug for the treatment of EV71 ***,it is urgent and of significance to develop anti-EV71 ***,we found that PLX8394,a RAF inhibitor,possesses high antiviral activity against EV71 in vitro,being superior to the traditional clinical drug ***,PLX8394 exhibits broad-spectrum antiviral activity against ***,in a suckling mouse model,PLX8394 provided a 70%protection rate for EV71-infected mice,reduced the viral load in liver and heart tissues,and relieved the inflammatory response.A mechanistic study showed that PLX8394 inhibited EV71 by suppressing the RAF/MEK/ERK signaling ***,PLX8394 lays a foundation for the development of new drugs against EV71.

为了研究摩擦热、介质压力对剖分式机械密封变形的影响,对剖分式机械密封进行热力耦合数值分析。结果表明:在温度场中密封环外径侧温度较低,剖分动环端面最高温度出现在与剖分静环相接触的密封端面处,剖分静环端面最高温度出现在密封端面内径处;在热、力耦合作用下密封端面由外径向内径的变形形成收敛状间隙;随着介质压力增大,密封环的轴向变形增大,密封环分型面的径向变形减小;介质压力的变化导致密封环端面间产生的摩擦热不同,从而引起密封环变形的差异。

Little is known about Subgenomic RNA(sgRNA)dynamics in patients with Coronavirus diseases 2019(COVID-19).We collected 147 throat swabs,74 gut swabs and 46 plasma samples from 117 COVID-19 patients recruited in the LOTUS China trial(ChiCTR2000029308)and compared E and orf7a sgRNA load in patients with different illness duration,outcome,and *** sgRNAs were detected in all the three types of samples,with longest duration of 25,13,and 17 days for E sgRNA,and 32,28,and 17 days for orf7a sgRNA in throat,gut,and plasma,respectively.A total of 95%(57/60)of patients had no E sgRNA detected after 10 days post treatment,though 86%of them were still E RNA *** correlation on titer was observed between sgRNA encoding E and orf7a *** showed similar variation in the standard care and Lopinavir-Ritonavir *** with diabetes and heart diseases showed higher pharyngeal E sgRNA at the first day(P=0.016 and 0.013,respectively)but no difference at five days after treatment,compared with patients without such *** with hypertension and cerebrovascular diseases showed no difference in the pharyngeal sgRNA levels at both one and five days after treatment,compared with patients without these two commodities.E sgRNA levels in the initial infection showed no correlation with the serum antibody against spike,nucleoprotein,and receptor binding domains at ten days *** lasted a long period in COVID-19 patients and might have little effect on humoral response.

A Megatheropod Tooth from the Early Cretaceous of Fusui, Guangxi, Southern China

A nearly complete right maxillary or left dentary tooth（NHMG 10858） from the Lower Cretaceous Xinlong Formation of the Napai Basin, Fusui County, Guangxi, southern China, is described. The tooth is large in size, with a CBL of 37 mm. Given the geological age and its crown morphology, including the size, it is most likely that the tooth belongs to a carcharodontosaurid. The recovered specimen represents one of the largest theropod teeth hitherto reported from the Early Cretaceous of Asia.

Rapid detection and discrimination of single photons are pivotal in various applications,such as deep-space laser communication,high-rate quantum key distribution,and optical quantum ***,conventional single-photon detectors(SPDs),including semiconducting and recently developed superconducting detectors,have limited detection speed and photon number resolution(PNR),which pose significant challenges in practical *** this paper,we present an efficient,fast SPD with good PNR,which has 64 paralleled,sandwiched superconducting nanowires fabricated on a distributed Bragg *** detector is operated in a compact Gifford–McMahon cryocooler that supports 64 electrical channels and has a minimum working temperature of 2.3 *** combined detector system shows a functional nanowire yield of 61/64,a system detection efficiency of 90%at 1550 nm,and a maximum count rate of 5.2 ***,it has a maximum PNR of 61,corresponding to the operating *** SPD signifies a substantial improvement in quantum detector technology,with potential applications in deep-space laser communication,high-speed quantum communication,and fundamental quantum optics experiments.

The outbreak of a novel coronavirus disease(COVID-19,caused by the 2019-nCoV infection)in December 2019 is one of the most severe public health emergencies since the founding of People’s Republic of China in *** personnel(HCP)nationwide are facing heavy workloads and high risk of infection,especially those who care for patients in Hubei ***,as of February 20,2020,over two thousand COVID-19 cases are confirmed among HCP from 476 hospitals nationwide,with nearly 90%of them from Hubei *** on literature search and interviews with some HCP working at Wuhan,capital city of Hubei,we have summarized some of the effective measures taken to reduce infection among HCP,and also made suggestions for improving occupational safety during an infectious disease *** experience and lessons learned should be a valuable asset for international health community to contain the ongoing COVID-19 epidemic around the world.
