Tissue engineering of blood vessels with endothelial cells differentiated from mouse embryonic stem cells

Endothelial cells （TEC3 cells） derived from mouse embryonic stem （ES） cells were used as seed cells to construct blood vessels. Tissue engineered blood vessels were made by seeding 8 × 106 smooth muscle cells （SMCs） obtained from rabbit arteries onto a sheet of nonwoven polyglycolic acid （PGA） fibers, which was used as a biodegradable polymer scaffold. After being cultured in DMEM medium for 7 days in vitro, SMCs grew well on the PGA fibers, and the cell-PGA sheet was then wrapped around a silicon tube, and implanted subcutaneously into nude mice. After 6～8 weeks, the silicon tube was replaced with another silicon tube in smaller diameter, and then the TEC3 cells （endothelial cells differentiated from mouse ES cells） were injected inside the engineered vessel tube as the test group. Human gastric cancer MKN-45 cells were transfectedwith pULB 3238, a plasmid carrying MVMp NS-1 genewith its original P4 promoter replaced by the glucocorticoid inducible promoter MMTV-LTR. After the integration and expression of NS-1 gene, some of the transfectantsdied, while others remained alive, but the growth featuresof survived cells were changed. For further study on theantineoplastic function of parvoviral NS-1 protein in vivo,transgenic mice carrying NS-1 genes were established byconventional method. Among 4 founders, one of them wasfound to be able to transmit the transgene to around 50%of their offsprings. RT-PCR was performed to indicate theexpression of NS-1 gene in transgenic mice and its mRNAappeared in a variety of tissues. The expression of integrated NS-1 gene may correlate with the decreased incidence of tumor induced in vivo by chemical carcinogens.

Bubble Size Distribution for Waves Propagating over A Submerged Breakwater

Experiments are carried out to study the characteristics of active bubbles entrained by breaking waves as these propagate over an abruptly topographical change or a submerged breakwater. Underwater sounds generated by the entrained air bubbles are detected by a hydrophone connected to a charge amplifier and a data acquisition system. The size distribution of the bubbles is then determined inversely from the received sound frequencies. The sound signals are converted from time domain to time-frequency domain by applying Gabor transform. The number of bubbles with different sizes are counted from the signal peaks in the time-frequency domain. The characteristics of the bubbles are in terms of bubble size spectra, which account for the variation in bubble probability density related to the bubble radius r. The experimental data demonstrate that the bubble probability density function shows a - 2.39 power-law sealing with radius for r 〉 0. 8 mm, and a- 1.11 power law for r 〈0.8 mm.

Can overexpression of TGF-β1 gene change the sex ratio in transgenic mice?

Mouse TGF-β1 gene was microinjected into male pronuclei of F2 hybrid fertilized eggs obtained by mating CSJLF1 and C57BL/6J inbred strains to generate transgenic mice with over-expressed TGF-β1 gene. The rate of founder production is 31% and Southern blot analysis of founder mice tail DNAS gave an integration efficiency of 33%. TGF-β1 gene could be stably integrated to the chromosomes of transgenic mice and transmitted to their progeny at a rate of 33% in the second generation. Dot blot analysis of tail RNA of some transgenic mice indicated a moderate expression of the transgene. The most interesting finding of the present work is the striking deviation from the normal male:female sex ratio in transgenic mice,with an average ratio of 6.7:1. The possible nature of the predominance of male sex in transgenic mice overexpressing TGF-β1 is discussed.

BACKGROUNDEwing’s sarcoma is a highly malignant primary bone tumor that commonly affectschildren. For young patients, multidisciplinary treatment and limb salvage arerecommended, and surgical plans considering the growth potential and boneactivity after tumor resection are *** SUMMARYAn 11-year-old Asian boy had a 1-mo history of a right-sided limping *** revealed a proximal tumor with bone destruction and physealinvolvement over the right femoral neck. He was diagnosed with stage IV(T1N0M1aG3) Ewing’s sarcoma with bilateral lung metastases. Neoadjuvantchemotherapy decreased the tumor size and confined it to the metaphysealregion. The patient underwent four stages of surgery: wide tumor excision plusreconstruction with vascular fibular bone graft plus internal fixation;repeat openreduction and internal fixation;femoral lengthening with orthosis after physealmaturity;and orthosis removal and bone elongation (approximately 6 cm).Following surgery, he could walk without discomfort and had almost equal-sizedbilateral femoral heads, indicating physis preservation. The surgery wassuccessful, and normal femoral head growth was achieved after completeremission. The patient was able to resume normal activities with equal length ofthe bilateral lower *** treatment and reconstruction following resection are important in skeletally immature patients with Ewing’s sarcoma to improve quality of life.

AIM:To identify the mucosal patterns of Helicobacter pylori(*** )-related gastritis in the gastric corpus using standard endoscopy and to evaluate their ***:A total of 112 consecutive patients underwent upper gastrointestinal *** endoscopists classified the endoscopic findings into 4 *** the second part of the study,90 images were shown to 3 endoscopists in order to evaluate the inter-observer and intra-observer variability in image ***:The mucosal patterns of the gastric bodywere categorized into 4 *** 1 pattern was defined as cleft-like appearance,type 2 as regular arrangement of red dots,type 3 pattern as the mosaic mucosal pattern and type 4 pattern as the mosaic pattern with a focal area of *** 1 and type 2 mucosal patterns were statistically significant in predicting *** status as compared with other mucosal types(χ 2 = 12.79 and 61.25 respectively,P < 0.01).Type 3 and type 4 mucosal patterns were statistically significant in predicting a ***-positive status as compared with other mucosal types(χ 2 = 21.22 and 11.02 respectively,P < 0.01).Furthermore,the sensitivity,specificity,positive and negative predictive values of type 3 plus type 4 patterns for predicting ***-positive gastric mucosa were 100%,86%,94%,and 100%,*** mean κ values for inter-and intra-observer agreement in assessing the various endoscopic patterns were 0.808(95% CI,0.678-0.938) and 0.826(95% CI,0.727-0.925) ***:Our study suggests that mucosal patterns in ***-infected gastric mucosa without atrophy can be reliably identified using standard endoscopy in the gastric corpus.

Peliosis hepatis(PH)is a vascular lesion of the liver that mimics a hepatic *** is often associated with underlying conditions,such as chronic infection and tumor malignancies,or with the use of anabolic steroids,immunosuppressive drugs,and oral *** patients with PH are asymptomatic,but some present with abdominal distension and *** some cases,PH may induce intraperitoneal hemorrhage and portal *** study analyzed a 46-year-old male who received a transplanted kidney nine years prior and had undergone long-term immunosuppressive therapy following the renal *** patient experienced progressive abdominal distention and pain in the six months prior to this ***,imaging studies revealed multiple liver tumor-like abnormalities,which were determined to be PH by pathological *** the hepatic lesions were progressively enlarged,the patient suffered from complications related to portal hypertension,such as intense ascites and esophageal varices *** the patient was scheduled to undergo liver transplantation,he suffered hepatic failure and died prior to availability of a donor organ.
