Microcurvature landscapes induce neural stem cell polarity and enhance neural differentiation

Tissue curvature has long been recognized as an important anatomical parameter that affects intracellular behaviors,and there is emerging interest in applying cell-scale curvature as a designer property to drive cell fates for tissue engineering *** neural cells are known to undergo dramatic and terminal morphological changes during development and curvature-limiting behaviors have been demonstrated in neurite outgrowth studies,there are still crucial gaps in understanding neural cell behaviors,particularly in the context of a three-dimensional(3D)curvature landscape similar to an actual tissue engineering *** this study,we fabricated two substrates of microcurvature(curvature-substrates)that present a smooth and repeating landscape with focuses of either a concave or a convex *** these curvature-substrates,we studied the properties of morphological differentiation in N2a neuroblastoma *** contrast to other studies where two-dimensional(2D)curvature was demonstrated to limit neurite outgrowth,we found that both the concave and convex substrates acted as continuous and uniform mechanical protrusions that significantly enhanced neural polarity and differentiation with few morphological changes in the main cell *** enhanced differentiation was manifested in various properties,including increased neurite length,increased nuclear displacement,and upregulation of various neural *** demonstrating how the micron-scale curvature landscape induces neuronal polarity,we provide further insights into the design of biomaterials utilizing the influence of surface curvature in neural tissue engineering.

Value of gastrografin in adhesive small bowel obstruction after unsuccessful conservative treatment: A prospective evaluation

AIM: Gastrografin is a hyperosmolar water-soluble contrast medium. Besides its predictive value for the need for operative treatment, a potential therapeutic role of this agent in adhesive small bowel obstruction has been suggested. This study aimed at evaluating the effectiveness of gastrografin in adhesive small bowel obstruction when conservative treatment failed. METhoDS: Patients with adhesive small bowel obstruction were given trial conservative treatment unless there was fear of bowel strangulation. Those responded in the initial 48 h had conservative treatment continued. Patients who showed no improvement in the initial 48 h were given 100 mL of gastrografin through nasogastric tube followed by serial abdominal radiographs. Patients with the contrast appeared in large bowel within 24 h were regarded as having partial obstruction and conservative treatment was continued. Patients in which the contrast failed to reach large bowel within 24 h were considered to have complete obstruction and laparotomy was performed. RESULTS: Two hundred and twelve patients with 245 episodes of adhesive obstruction were included. Fifteen patients were operated on soon after admission due to fear of strangulation. One hundred and eighty-six episodes of obstruction showed improvement in the initial 48 h and conservative treatment was continued. Two patients had subsequent operations because of persistent obstruction. Forty-four episodes of obstruction showed no improvement within 48 h and gastrografin was administered. Seven patients underwent complete obstruction surgery. Partial obstruction was demonstrated in 37 other cases, obstruction resolved subsequently in all of them except one patient who required laparotomy because of persistent obstruction. The overall operative rate in this study was 10%. There was no complication that could be attributed to the use of gastrografin. Purpose:This article aims to illustrate from the author’s insider perspective the lived experiences of engaging in private tutoring in hong Kong as a tutee,a tutor,and a researcher and draw implications on several issues arising from the prevalence of shadow ***/Approach/Methods:This article adopted an autobiographical narrative *** were collected through the author’s memoir of events,stimulated by the tutorial materials he used when he was a tutee and a tutor,his own video-recorded lessons of tutoring,and reflective journals from his research ***:Various issues are discussed based on the narrative of the author playing different roles in the tutoring industry,including(1)the positive and negative washback on mainstream education,(2)the lack of strict regulation of the quality of tutors and advertisements,and(3)how shadow education may exacerbate education inequality and how some tutorial companies and nonprofit organizations are addressing the ***/Value:This article,to the best of the author’s knowledge,is the only one that discusses the issues of shadow education from an author’s own personal experiences as a tutee,a tutor,and a *** illustrates how practices and policies of the private tutoring industry are evolving in hong Kong from an insider perspective.
