MAGI3 Suppresses Glioma Cell Proliferation via Upregulation of PTEN Expression

Objective To investigate the role and molecular mechanism of membrane-associated guanylate k.nase inverted 3 （MAGI3） in glioma cell proliferation. Methods The expression levels of MAGI3 and PTEN were assessed in glioma samples by Western blotting. MAGI3 was stably transfected into C6 glioma cells to obtain C6-MAGI3 cells. Then, the proliferation, the expression levels of MAGI3 and PTEN, and Ak. phosphorylation were evaluated in C6 and C6-MAGI3 cells. Xenograft tumor models were established by subcutaneous injection of C6 and C6-MAGI3 cells into nude mice, and the growth rates of xenografts in the mice were compared. The potential role of MAGI3 expression in PI3k.Ak. signaling activation was further investigated by examining the correlation between MAGI3 expression and the expression of PI3k.Ak. signaling downstream target genes in a glioma dataset using gene set enrichment analysis （GSEA）. Results Expression levels of MAGI3 and PTEN were significantly downregulated in gliomas. Overexpression of MAGI3 in the glioma C6 cell line upregulated PTEN protein expression, inhibited the phosphorylation of Ak., and suppressed cell proliferation. MAGI3 overexpression also inhibited the growth of C6 glioma tumor xenografts in nude mice. Analysis based on the GEO database confirmed the negative correlation between activation of PI3k.Ak. pathway and MAGI3 mRNA levels in human glioma samples. Conclusion The loss of MAGI3 expression in downregulation of PTEN expression, leading potential glioma suppressor. glioma may enhance the proliferation of glioma cells via to the activation of the PI3k.Ak. pathway. Study on the flow structure in curved open channels with suspended vegetation using multi relaxation time lattice Boltzmann method

Suspended vegetation in rivers,lak.s,reservoirs and canals can change flow structure,which will in turn affect the sediment transport and cause the variation of water ecological *** order to study the characteristics of bend flow through suspended vegetation,three-dimensional numerical simulations are carried out by using the multi-relaxation-time lattice Boltzmann method(MRT-LBM).The drag force induced by vegetation is added in the velocity correction in the equilibrium distribution and a hybrid format combined bounce and specular reflection scheme is applied in the solid-fluid *** the validation of this model,six cases are designed to conduct the numerical simulations according to the root depth and the arrangement of *** simulated results show that the suspended vegetation can redistribute the flow structure in curved open *** the arrangement of suspended vegetation,the main flow moves to the side without vegetation,and the distribution of velocity tends to be balanced when vegetation is arranged on the entire cross section,the range of circulating current is reduced from the whole cross section to the local position without vegetation,however,the circulating current can still exist in the curve where the suspended vegetation enters less than half of the water *** addition,it can also be concluded that the suspended vegetation can affect the lateral gradient of flow velocity,and the bed shear stress in the curved channel.

Big Bang nucleosynthesis(BBN)theory predicts the primordial abundances of the light elements^(2) H(referred to as deuterium,or D for short),^(3)he,^(4)he,and^(7) Li produced in the early *** these,deuterium,the first nuclide produced by BBN,is a k.y primordial material for subsequent *** date,the uncertainty in predicted deuterium abundance(D/H)remains larger than the observational *** this study,the Monte Carlo simulation code PRIMAT was used to investigate the sensitivity of 11 important BBN reactions to deuterium *** found that the reaction rate uncertainties of the four reactions d(d,n)^(3)he,d(d,p)t,d(p,γ)^(3)he,and p(n,γ)d had the largest influence on the calculated D/H ***,the calculated D/H uncertainty cannot reach observational precision even with the recent LUNA precise d(p,γ)^(3) he *** the nuclear physics aspect,there is still room to largely reduce the reaction-rate uncertainties;hence,further measurements of the important reactions involved in BBN are still necessary.A photodisintegration experiment will be conducted at the Shanghai Laser Electron Gamma Source Facility to precisely study the deuterium production reaction of p(n,γ)d.

Populus trinervis is native to China and plays an irreplaceable role in maintaining the ecological balance of boreal and temperate *** mainly grows in high-altitude *** present,there are limited studies on the response of *** to different light qualities,so it is necessary to investigate the photosynthetic physiological changes of *** in different light *** our study,*** was grown for 8 months under light filtered by three different colored *** three treatments were blue film,green film,and white plastic *** effects of blue(B),green(G),and white(W)light on photosynthetic pigment content,absolute growth,photosynthetic parameters,soluble sugar content,and chlorophyll fluorescence parameters were studied,*** to the Wtreatment,the chlorophyll a and b,carotenoids,total chlorophyll content(a+b),absolute growth of seedling height,net photosynthetic rate(PN),water use efficiency(WUE),total soluble sugars,sucrose,and nonphotochemical quenching(NPQ)of *** were significantly increased under B ***,chlorophyll a and b,carotenoids,total chlorophyll(a+b),transpiration rate(Tr),intercellular CO_(2) concentration(Ci),stomatal conductance(gs),absolute growth of seedling height and leaf length,reducing sugar,total soluble sugar content,and NPQ were significantly increased under G *** results showed that the absolute growth and chlorophyll content of *** were increased under B light,while the sugar and photosynthetic parameters were increased under G *** studies may look.into how B light impacts absolute growth and promotional mechanisms,as well as how G light affects the accumulation of sugar levels.

Construction of algal-bacterial consortia using green microalgae Chlorella vulgaris and As(Ⅲ)-oxidizing bacteria:As tolerance and metabolomic profiling

Selective N-Methylation of N-Methylaniline with CO_(2) and H_(2) over Cu/In_(2)O_(3) Catalyst

N-Methylation of amines with CO_(2) and H2 is a potential approach for CO_(2) utilization because N-methylated amines can be used as solvents and organic intermediates. In_(2)O_(3)-supported Cu (Cu/In_(2)O_(3)) acts as an effective heterogeneous catalyst for N-methylation reaction of N-methylaniline (MA) with CO_(2) and H_(2),showing higher N,N-dimethylaniline (DMA) selectivity than other supported Cu catalysts. On one hand,the dispersion of Cu can be improved by the defective In_(2)O_(3) support. On the other hand,In_(2)O_(3) support is active in the dissociative adsorption of CO_(2) through C—O bond break.ng. In addition,the H_(2) dissociation ability of In_(2)O_(3) can also be enhanced by Cu. The combination of Cu and In_(2)O_(3) is effective in the activation of CO_(2),the adsorption of intermediate N-methylformanilide (MFA),the hydrogenation of MFA to DMA and the prohibition of C—N bond cleavage side reactions,thereby enhancing the reaction rate of MA conversion and the selectivity to DMA.

Joint optimization of production,maintenance,and quality control considering the product quality variance of a degraded system

The joint optimization of production,maintenance,and quality control has shown effectiveness in reducing long-term operational costs in production ***,existing studies often assume that changes in the mean value of product quality characteristics in a deteriorating system follow a specific distribution while k.eping variance *** address this limitation,we propose an innovative method based on the continuous rank.ng probability score(CRPS).This method enables the simultaneous detection of changes in mean and variance in nonconformities,thus removing the assumption of a specific distribution for quality *** approach focuses on developing optimal strategies for production,maintenance,and quality control to minimize cost per unit of ***,we employ a stochastic model to optimize the production time allocated to the inventory buffer,resulting in significant cost *** effectiveness of our proposed joint optimization method is demonstrated through comprehensive numerical experiments,sensitivity analysis,and a comparative *** results show that our method can achieve cost reductions compared to several other related methods,highlighting its practical applicability for manufacturing companies aiming to reduce costs.

Ballistic performances of the hourglass lattice sandwich structures under high-velocity fragments

In this paper,the numerical simulation method is used to study the ballistic performances of hourglass lattice sandwich structures with the same mass under the vertical incidence of *** is paid to elucidating the influences of rod cross-section dimensions,structure height,structure layer,and rod inclination angle on the deformation mode,ballistic performances,and ability to change the ballistic direction of *** results show that the ballistic performances of hourglass lattice sandwich structures are mainly affected by their structural *** this respect,structural parameters optimization of the hourglass lattice sandwich structures enable one to effectively improve their ballistic limit velocity and,consequently,ballistic performances.
