数字鸿沟、数字普惠金融与就业增长

在数字信息技术发展不均衡的背景下,文章基于2011-2020年中国省级层面宏观经济数据,构建数字鸿沟测度水平指标,实证考察数字鸿沟水平与就业增长之间的关系。研究发现:数字鸿沟的出现抑制了就业增长,特别是私营主体的就业规模受到了更大程度的抑制影响;数字普惠金融的发展能够缓解数字鸿沟所造成的就业增长抑制现象。上述发现不仅理顺了数字鸿沟、数字普惠金融与就业增长之间的逻辑关系,还为政府促进地区就业增长提供新思路。

目的探讨奥希替尼治疗表皮生长因子受体酪氨酸激酶抑制剂(EGFR-TKI)耐药晚期肺腺癌的疗效及对血清CEA、VEGF水平的影响。方法回顾性选取2021年3月-2023年3月本院进行诊治的90例EGFR-TKI耐药型晚期肺腺癌患者,根据治疗方案的不同分为常规组(n=42,采用常规化疗方案)和奥希替尼组(n=48,采用奥希替尼治疗方案),两组患者均经治疗后完成3个月的随访,收集两组患者的临床相关资料,观察两组的临床疗效。记录两组患者治疗前后实验室相关指标:血清肿瘤标志物[糖类抗原125(CA125)、癌胚抗原(CEA)及细胞角蛋白19片段(Cyfra21-1)]和血管生长因子[血管内皮生长因子(VEGF)、血小板衍生生长因子(PDGF)、碱性成纤维细胞生长因子(bFGF)]水平,观察两组患者治疗前后生存质量变化[生命质量测定量表(QLQ-C30)]及患者治疗期间不良反应发生情况。结果奥希替尼组的总临床有效率为89.58%,明显高于常规组的68.05%(χ^(2)=5.904,P=0.015);治疗后,两组患者的CA125、CEA、Cyfra21-1水平均有明显下降(P<0.05),奥希替尼组患者的CA125、CEA、Cyfra21-1水平均低于常规组(P<0.05);两组患者的VEGF、PDGF、b FGF水平均有明显下降(P<0.05),奥希替尼组患者的VEGF、PDGF、b FGF水平均低于常规组(P<0.05);两组患者的生存质量各领域评分均有所上升(P<0.05),奥希替尼组患者的生存质量各领域评分均高于常规组(P<0.05)。两组不良反应发生率相较无差异(18.75%vs 16.67%)(χ^(2)=0.067,P=0.796)。结论奥希替尼治疗EGFR-TKI耐药晚期肺腺癌的临床疗效显著,可有效降低其CEA、VEGF水平,改善其生存质量,且不良反应风险小,可行临床推广。

Shear-thinning hydrogels have emerged for endoscopic submucosal dissection,while wound intervention after surgery has rarely been ***,a catechol-modified chitosa n hydrogel with shear-thinning property was developed for simultaneously facilitating endoscopic submucosal dissection and postoperative wound *** from the shear-thinning and self-healing characteristics,the asprepared hydrogel showed easily endoscopic *** also performed very well as submucosal cushion,which could remain above 70%after injection for 120 min in ex vivo porcine large intestine *** fact,the cushion height of normal saline dramatically decreased to 46%of the initial height at 30 *** nanoparticles encapsulated into the network endowed the hydrogel with almost reached 100%antibacterial effect against *** and *** hemolysis ratio of the hydrogel was calculated to be as low as 0.8%.Combined with good hemocompatibility and cytocompatibility,the as-prepared hydrogel displayed much higher in vivo wound closu re and healing efficacy than normal *** results demonstrated the superiority of the shear-thinning chitosan hydrogel in facilitating clinical endoscopic submucosal dissection surgery.

Cold stress is an important factor that limits apple production. In this study, we examined the tissue-cultured plantlets of apple rootstocks ‘M9T337' and ‘60-160', which are resistant and sensitive to cold stress, respectively. The enriched pathways of differentially expressed genes(DEGs) and physiological changes in ‘M9T337' and ‘60-160' plantlets were clearly different after cold stress(1°C) treatment for 48 h, suggesting that they have differential responses to cold stress. The differential expression of WRKY transcription factors in the two plantlets showed that MdWRKY40is and MdWRKY48 are potential regulators of cold tolerance. When we overexpressed MdWRKY40is and MdWRKY48in apple calli, the overexpression of MdWRKY48 had no significant effect on the callus, while MdWRKY40is overexpression promoted anthocyanin accumulation, increased callus cold tolerance, and promoted the expression of anthocyanin structural gene MdDFR and cold-signaling core gene MdCBF2. Yeast one-hybrid screening and electrophoretic mobility shift assays showed that MdWRKY40is could only bind to the MdDFR promoter. Yeast twohybrid screening and bimolecular fluorescence complementation showed that MdWRKY40is interacts with the CBF2inhibitor MdMYB15L through the leucine zipper(LZ). When the LZ of MdWRMY40is was knocked out, MdWRKY40is overexpression in the callus did not affect MdCBF2 expression or callus cold tolerance, indicating that MdWRKY40is acts in the cold signaling pathway by interacting with MdMYB15L. In summary, MdWRKY40is can directly bind to the MdDFR promoter in order to promote anthocyanin accumulation, and it can also interact with MdMYB15L to interfere with its inhibitory effect on MdCBF2, indirectly promoting MdCBF2 expression, and thereby improving cold *** results provide a new perspective for the cold-resistance mechanism of apple rootstocks and a molecular basis for the screening of cold-resistant rootstocks.

It regulates manyaspects of T-cell biology including thymocyte development, T cell differentiation and effector T cell function.1 Previous work has shown that the loss of TGF-β signaling in the T-cell lineage leads to severe lymphopro- liferafive autoimmunity.2.3 Until now, it has been unclear to what extent the autoimmunity observed in those mice is driven by TGF- unresponsive conventional T cells, as TGF- is also crucial for the maintenance and function of regulatory T cells （Treg）. A recent study pub- lished in Nature Immunology revealed a novel role of TGF-β signaling in non-regulatory naive T cells. It was found that TGF-β controls lym- phopenia-driven T-ceU proliferation in neonatal mice, thereby preventing the development of autoimmtmity.

Dendritic cell(DC)-targeted delivery of mRNA is a prominent method to boost the efficacy of mRNA tumor *** targeting ligands are often modified on nanocarriers by polyethylene glycol(PEG)linker in mRNA delivery *** the PEG linker length influences the targeting delivery efficiency of mRNA nanocarrier in vivo remains ***,we designed and constructed DC-targeted mRNA delivery systems modified by mannose via different PEG linker lengths(100/400/1000/2000)(MPn-LPX).The top candidate MP_(400)-LPX(the linker was PEG400)showed the optimal mRNA expression and antigen presentation owing to the highly efficient uptake by ***,MP_(400)-LPX could better inhibited tumor growth and extended survival in the E.G7-OVA lymphoma and TC-1 cervical tumor mouse ***,these results demonstrated that PEG400 was the optimal linker for the PEGylated DC-targeted mRNA *** findings provided a new platform for the rational design of targeted mRNA nanovaccines with shorter-length PEG.
