Repressing iron overload ameliorates central poststroke pain via the Hdac2-Kv1.2 axis in a rat model of hemorrhagic stroke

Thalamic hemorrhage can lead to the development of central post-stroke pain.Changes in histone acetylation levels,which are regulated by histone deacetylases,affect the excitability of neurons surrounding the hemorrhagic area.However,the regulato ry mechanism of histone deacetylases in central post-stroke pain remains unclea r.here,we show that iron overload leads to an increase in histone deacetylase 2expression in damaged ventral posterolateral nucleus neurons.Inhibiting this increase restored histone H3 acetylation in the Kcna2 promoter region of the voltage-dependent potassium(Kv)channel subunit gene in a rat model of central post-stroke pain,thereby increasing Kcna2expression and relieving central pain.However,in the absence of nerve injury,increasing histone deacetylase 2 expression decreased Kcna2expression,decreased Kv current,increased the excitability of neurons in the ventral posterolateral nucleus area,and led to neuropathic pain symptoms.Moreover,treatment with the iron chelator deferiprone effectively reduced iron overload in the ventral posterolateral nucleus after intracerebral hemorrhage,reversed histone deacetylase 2 upregulation and Kv1.2 downregulation,and alleviated mechanical hypersensitivity in central post-stroke pain rats.These results suggest that histone deacetylase 2 upregulation and Kv1.2 downregulation,mediated by iron overload,are important factors in central post-stroke pain pathogenesis and co uld se rve as new to rgets for central poststroke pain treatment.

Ca_(2)MnO_(4)-layered perovskite modified by NaNO_(3)for chemical-looping oxidative dehydrogenation of ethane to ethylene

Chemical-looping oxidative dehydrogenation(CL-ODH)is a process designed for the conversion of alkanes into olefins through cyclic redox reactions,eliminating the need for gaseous O_(2).In this work,we investigated the use of Ca_(2)MnO_(4)-layered perovskites modified with NaNO_(3) dopants,serving as redox catalysts(also known as oxygen carriers),for the CL-ODH of ethane within a temperature range of 700-780℃.Our findings revealed that the incorporation of NaNO_(3) as a modifier significantly-nhanced the selectivity for-thylene generation from Ca_(2)MnO_(4).At 750℃and a gas hourly space velocity of 1300 h^(-1),we achieved an-thane conversion up to 68.17%,accompanied by a corresponding-thylene yield of 57.39%.X-ray photoelectron spectroscopy analysis unveiled that the doping NaNO_(3) onto Ca_(2)MnO_(4) not only played a role in reducing the oxidation state of Mn ions but also increased the lattice oxygen content of the redox catalyst.Furthermore,formation of NaNO_(3) shell on the surface of Ca_(2)MnO_(4) led to a reduction in the concentration of manganese sites and modulated the oxygen-releasing behavior in a step-wise manner.This modulation contributed significantly to the enhanced selectivity for ethylene of the NaNO_(3)-doped Ca_(2)MnO_(4) catalyst.These findings provide compelling evidence for the potential of Ca_(2)MnO_(4)-layered perovskites as promising redox catalysts in the context of CL-ODH reactions.

Study on the flow structure in curved open channels with suspended vegetation using multi relaxation time lattice Boltzmann method

Suspended vegetation in rivers,lakes,reservoirs and canals can change flow structure,which will in turn affect the sediment transport and cause the variation of water ecological environment.In order to study the characteristics of bend flow through suspended vegetation,three-dimensional numerical simulations are carried out by using the multi-relaxation-time lattice Boltzmann method(MRT-LBM).The drag force induced by vegetation is added in the velocity correction in the equilibrium distribution and a hybrid format combined bounce and specular reflection scheme is applied in the solid-fluid boundaries.After the validation of this model,six cases are designed to conduct the numerical simulations according to the root depth and the arrangement of vegetation.The simulated results show that the suspended vegetation can redistribute the flow structure in curved open channels.After the arrangement of suspended vegetation,the main flow moves to the side without vegetation,and the distribution of velocity tends to be balanced when vegetation is arranged on the entire cross section,the range of circulating current is reduced from the whole cross section to the local position without vegetation,however,the circulating current can still exist in the curve where the suspended vegetation enters less than half of the water depth.In addition,it can also be concluded that the suspended vegetation can affect the lateral gradient of flow velocity,and the bed shear stress in the curved channel.

From common biomass materials to high-performance tissue engineering scaffold: Biomimetic preparation, properties characterization, in vitro and in vivo evaluations

Converting common biomass materials to high-performance biomedical products could not only reduce the environmental pressure associated with the large-scale use of synthetic materials,but also increase the economic value.Chitosan as a very promising candidate has drawn considerable attention owing to its abundant sources and remarkable bioactivities.However,pure chitosan materials usually exhibit insufficient mechanical properties and excessive swelling ratio,which seriously affected their in vivo stability and integrity when applied as tissue engineering scaf-folds.Thus,simultaneously improving the mechanical strength and biological compatibility of pure chitosan(CS)scaffolds becomes very important.here,inspired by the fiber-reinforced con-struction of natural extracellular matrix and the porous structure of cancellous bone,we built silk microfibers/chitosan composite scaffolds via ice-templating technique.This biomimetic strategy achieved 500%of mechanical improvement to pure chitosan,and meanwhile still maintaining high porosity(>87%).In addition,the increased roughness of chitosan pore walls by embedded silk microfibers significantly promoted cell adhesion and proliferation.More importantly,after subcutaneous implantation in mice for four weeks,the composite scaffold showed greater struc-tural integrity,as well as better collagenation,angiogenesis,and osteogenesis abilities,suggesting its great potential in biomedicine.

Safflower Yellow Inhibits Progression of hepatocellular Carcinoma by Modulating Immunological Tolerance via FAK Pathway

Objective To explore the anti-tumor effect of safflower yellow(SY)against hepatocellular carcinoma(HCC)and the underlying potential mechanism.Methods An in vitro model was established by mixing Luc-hepa1-6 cells and CD3^(+)CD8^(+)T cells,followed by adding programmed cell death protein 1(PD-1)antibody(Anti-mPD-1)with or without SY.The apoptosis was detected by flow cytometry and the level of inflammatory cytokines was determined by enzyme-linked immunosorbent assay.The protein levels of programmed cell death 1 ligand 1(PD-L1),chemokine ligand(CCL5),C-X-C motif chemokine ligand 10(CXCL10)were measured by Western blot.An in situ animal model was established in mice followed by treatment with anti-mPD-1 with or without SY.Bioluminescence imaging was monitored with an AniView 100 imaging system.To establish the FAK-overexpressed Luc-hepa1-6 cells,cells were transfected with adenovirus containing pcDNA3.1-FAK for 48 h.Results The fluorescence intensity,apoptotic rate,release of inflammatory cytokines,and CCL5/CXCL10 secretion were dramatically facilitated by anti-mPD-1(Pher enhanced by SY(Phermore,the enhanced effects of SY on inhibiting tumor cell growth,facilitating apoptosis and inflammatory cytokine releasing,suppressing the PD-1/PD-L1 axis,and inducing the CCL5/CXCL10 secretion in Anti-mPD-1 treated mixture of Luc-hepa1-6 cells and CD3+CD8+T cells were abolished by FAK overexpression(Phe progression of HCC by mediating immunological tolerance through inhibiting FAK.

The evolution and functional divergence of 10 Apolipoprotein D-like genes in Nilaparvata lugens

Apolipoprotein D(ApoD),a member of the lipocalin superfamily of proteins,is involved in lipid transport and stress resistance.Whereas only a single copy of the ApoD gene is found in humans and some other vertebrates,there are typically several ApoD-like genes in insects.To date,there have been relatively few studies that have examined the evolution and functional differentiation of ApoD-like genes in insects,particularly hemi-metabolous insects.In this study,we identified 10 ApoD-like genes(NlApoD1−10)with distinct spatiotemporal expression patterns in Nilaparvata lugens(BPH),which is an important pest of rice.NlApoD1−10 were found to be distributed on 3 chromosomes in a tandem array of NlApoD1/2,NlApoD3−5,and NlApoD7/8,and show sequence and gene structural divergence in the coding regions,indicating that multiple gene duplication events occurred during evolution.Phylogenetic analysis revealed that NlApoD1−10 can be clustered into 5 clades,with NlApoD3−5 and NlApoD7/8 potentially evolving exclusively in the Delphacidae family.Functional screening using an RNA interference approach revealed that only NlApoD2 was essential for BPH development and survival,whereas NlApoD4/5 are highly expressed in testes,and might play roles in reproduction.Moreover,stress response analysis revealed that NlApoD3−5/9,NlApoD3−5,and NlApoD9 were up-regulated after treatment with lipopolysaccharide,H2O2,and ultraviolet-C,respectively,indicating their potential roles in stress resistance.

Numerical Investigations on the Transient Behavior of Sand Waves in Beibu Gulf Under Normal and Extreme Sea Conditions

In this study, a morphodynamic numerical model is established with the Regional Ocean Modeling System(ROMS)to investigate the transient behavior of sand waves under realistic sea conditions. The simulation of sand wave evolution comprises two steps: 1) a regional-scale model is configured first to simulate the ocean hydrodynamics, i.e., tides and tidal currents, and 2) the transient behavior of sand waves is simulated in a small computational domain under the time-variant currents extracted from the large model. The evolution of sand waves on the continental shelf in the Beibu Gulf is specifically investigated. The numerical results of the two-year evolution of sand waves under normal sea conditions compare well with the field survey data. The transient behavior of sand waves in individual months shows that the sand waves are more stable in April and October than that in other months, which can be selected as the windows for seabed operations. The effects of sediment properties, including settling velocity, critical shear stress and surface erosion rate, on sand wave evolution are also analyzed. Then, the typhoon-induced currents are further superimposed on the tidal currents as the extreme weather conditions. Sand waves with the average wavelength generally have more active behavior than smaller or larger sand waves. The characteristics of the evolution of sand waves in an individual typhoon process are quite different for different hydrodynamic combinations. For the storm conditions, i.e., the real combination and maximum combination cases, the sand waves experience a significant migration together with a damping in height due to the dominant suspended sediment transport. For the mild conditions, i.e., the pure tidal current and minimum combination cases, the sand waves migrate less, but the heights continue growing due to the dominant bedload transport.

Gaofen-3(GF-3)is the first Chinese spaceborne multi-polarization synthetic aperture radar(SAR)instrument at C-band(5.43 GHz).In this paper,we use data collected from GF-3 to observe Super Typhoon Lekima(2019)in the East China Sea.Using a VH-polarized wide ScanSAR(WSC)image,ocean surface wind speeds at 100m horizontal resolution are obtained at 21:56:59 UTC on 8 August 2019,with the maximum wind speed,38.9 m·s^(-1).Validating the SAR-retrieved winds with buoymeasured wind speeds,we find that the root mean square error(RMSE)is 1.86 m·s^(-1),and correlation coefficient,0.92.This suggests that wind speeds retrieved from GF-3 SAR are reliable.Both the European Centre for MediumRange Weather Forecasts(ECMWF)fine grid operational forecast products with spatial resolution,and China Global/Regional Assimilation and Prediction Enhance System(GRAPES)have good performances on surface wind prediction under weak wind speed condition(he maximum wind speed when the storm is intensified as a severe tropical storm(>24m·s^(-1)).With respect to SAR-retrieved wind speeds,the RMSEs are 5.24 m·s^(–1) for ECMWF and 5.17 m·s^(–1) for GRAPES,with biases of 4.16 m·s^(–1) for ECMWF and 3.84 m·s^(-1)for GRAPES during Super Typhoon Lekima(2019).

Deep hybrid: Multi-graph neural network collaboration for hyperspectral image classification

With limited number of labeled samples,hyperspectral image(HSI)classification is a difficult Problem in current research.The graph neural network(GNN)has emerged as an approach to semi-supervised classification,and the application of GNN to hyperspectral images has attracted much attention.However,in the existing GNN-based methods a single graph neural network or graph filter is mainly used to extract HSI features,which does not take full advantage of various graph neural networks(graph filters).Moreover,the traditional GNNs have the problem of oversmoothing.To alleviate these shortcomings,we introduce a deep hybrid multi-graph neural network(DHMG),where two different graph filters,i.e.,the spectral filter and the autoregressive moving average(ARMA)filter,are utilized in two branches.The former can well extract the spectral features of the nodes,and the latter has a good suppression effect on graph noise.The network realizes information interaction between the two branches and takes good advantage of different graph filters.In addition,to address the problem of oversmoothing,a dense network is proposed,where the local graph features are preserved.The dense structure satisfies the needs of different classification targets presenting different features.Finally,we introduce a GraphSAGEbased network to refine the graph features produced by the deep hybrid network.Extensive experiments on three public HSI datasets strongly demonstrate that the DHMG dramatically outperforms the state-ofthe-art models.

Potentially fatal electrolyte imbalance caused by severe hydrofluoric acid burns combined with inhalation injury: A case report

BACKGROUND Hydrofluoric acid(HF)is one of the most common causes of chemical burns.HF burns can cause wounds that deepen and progress aggressively.As a result,HF burns are often severe even if they involve a small area of the skin.Published cases of HF burns have mostly reported small HF burn areas.Few cases of HF inhalation injury have been reported to date.CASE SUMMARY A 24-year-old man suffered from extensive hydrofluoric acid burns covering 60%of the total body surface area(TBSA),including deep second degree burns on 47%and third degree burns on 13%of the TBSA,after he fell into a pickling pool containing 15%HF.Comprehensive treatments were carried out after the patient was admitted.Ventricular fibrillation occurred 9 times within the first 2 h,and the lowest serum Ca2+concentration was 0.192 mmol/L.A dose of calcium gluconate(37 g)was intravenously supplied during the first 24 h,and the total amount of calcium gluconate supplementation was 343 g.Extracorporeal membrane oxygenation(ECMO)was applied for 8 d to handle the acute respiratory distress syndrome(ARDS)induced by the HF inhalation injury.The patient was discharged after 99 d of comprehensive treatment,including skin grafting.CONCLUSION Extensive HF burns combined with an inhalation injury led to a potentially fatal electrolyte imbalance and ARDS.Adequate and timely calcium supplementation and ECMO application were the keys to successful treatment of the patient.
