杨树响应腐皮镰刀菌侵染的转录组学分析

[目的]基于RNA-Seq测序技术,初步探究了杨树-腐皮镰刀菌互作过程中相关基因的表达、主要信号通路和代谢途径,筛选了杨树防御腐皮镰刀菌侵染的关键基因,为进一步揭示此类病害的分子机制奠定基础。[方法]以生长2个月的银腺杨84K为材料,用1×10^(7)个·mL^(-1)的腐皮镰刀菌孢子液侵染根系,于侵染0h(对照组)、48 h和72 h(接菌组)后取根组织进行转录组测序,挖掘杨树响应腐皮镰刀菌侵染的相关基因。[结果](1)与对照组0 h相比,侵染48 h和72 h分别检测到8 939个和8 246个DEGs(Differentially Expressed Genes)。(2)GO分析发现,差异表达基因主要富集在单一生物体过程、对刺激反应、碳水化合物代谢、生物调节和对激素响应等过程。(3)KEGG分析表明,糖酵解/糖异生、碳代谢、植物激素信号转导和苯丙烷生物合成等途径在病原菌侵染后发生显著变化。(4)杨树糖转运蛋白SWEETs家族中21个成员的表达受腐皮镰刀菌侵染诱导。乙烯受体ETR、乙烯信号通路主要基因EIN3、ERF1、ERF2上调表达,木质素合成关键酶基因CCR、4CL、C3’H、COMT表达量显著升高。[结论]杨树可能通过调节体内糖代谢和转运,激活乙烯信号通路,促进木质素积累、细胞壁增厚等策略,响应腐皮镰刀菌的侵染。

We experimentally fabricate a non-spherical Ag and Co surface-enhanced Raman scattering（SERS） substrate, which not only retains the metallic plasmon resonant effect, but also possesses the magnetic field controllable *** detections are carried out with the test crystal violet（CV） and rhodamine 6G（R6G） molecules with the initiation of different magnitudes of external magnetic field. Experimental results indicate that our prepared substrate shows a higher SERS activity and magnetic controllability, where non-spherical Ag nanoparticles are driven to aggregate effectively by the magnetized Co and plenty of hot-spots are built around the metallic Ag nanoparticles, thereby leading to the enhancement of local electromagnetic field. Moreover, when the external magnetic field is increased, our prepared substrate demonstrates excellent SERS enhancement. With the 2500 Gs and 3500 Gs（1 Gs = 10^（-4）T） magnetic fields, SERS signal can also be obtained with the detection limit lowering down to 10^（-9）M. These results indicate that our proposed magnetic field controlled substrate enables us to freely achieve the enhanced and controllable SERS effect, which can be widely used in the optical sensing, single molecule detection and bio-medical applications.

Background Over-expression of P-glycoprotein （P-gp）, encoded by the MDR1 gene, confers multidrug resistance （MDR） in renal cell carcinoma （RCC） and is a major reason for unsuccessful chemotherapy. This study aimed to determine the effct of RNA interference （RNAi） on the reversal of MDR in human RCC. Methods We designed and selected one short hairpin RNA （shRNA） targeting MDR1 gene, which is stably expressed from integrated plasmid and transfected by lentivirus fluid in human RCC A498 cell. Results The MDRl-targeted RNAi resulted in decreased MDR1 gene mRNA level （P 〈0.001）, almost abolished P-gp expression and reversed MDR to different chemotherapy drugs in the RCC A498 cell line. Conclusion MDR could be reversed by RNAi in human RCC A498 cell line, which may be used for clinical application

Spontaneous imbibition(SI)is an important mechanism for enhancing oil recovery in low-permeability *** to the strong heterogeneity,and the non-Darcy flow,the construction of SI model for lowpermeability reservoirs is extremely ***,traditional SI models based on single or averaged capillary tortuosity ignore the influence of heterogeneity of pore seepage channels and the threshold pressure(TP)on ***,in this work,based on capillary model and fractal theory,a mathematical model of characterizing SI considering heterogeneity of pore seepage channels is *** this basis,the threshold pressure was introduced to determine the pore radius at which the wetted phase can displace *** proposed new SI model was verified by imbibition experimental *** study shows that for weakly heterogeneous cores with permeability of 0-1 m D,the traditional SI model can characterize the imbibition process relatively accurately,and the new imbibition model can increase the coefficient of determination by 1.05 ***,traditional model has serious deviations in predicting the imbibition recovery for cores with permeability of 10-50 m *** new SI model coupling with heterogeneity of pore seepage channels and threshold pressure effectively solves this problem,and the determination coefficient is increased from 0.344 to 0.922,which is increased by2.68 *** low-permeability reservoirs,the production of the oil in transitional pores(0.01-0.1μm)and mesopores(0.1-1μm)significantly affects the imbibition recovery,as the research shows that when the heterogeneity of pore seepage channels is ignored,the oil recovery in transitional pores and mesopores decreases by 7.54%and 4.26%,*** analysis shows that increasing interfacial tension,decreasing contact angle,oil-water viscosity ratio and threshold pressure will increase imbibition *** addition,there are critical values for the influence of these factors o

Distinction of weak and strong AC grids for emerging hierarchical-infeed LCC-UHVDC systems is important for planning and operation departments. However, accuracy of earlier distinction methods is limited as they were developed by empirical reasoning without rigorous theoretical analysis. Hence in this letter, hierarchical-infeed interactive effective short-circuit ratio (HIESCR) index is first used for strength evaluation of HIDC systems with complex inter-inverter interactions considered. Boundary HIESCR (BHIESCR) is also introduced in the proposed distinction method of weak and strong AC grids. That is, weak (or strong) AC grids are, respectively, identified when HIESCR is less (or greater) than BHIESCR. Second, it is shown BHIESCR remains almost unchanged as 3.0 versus various system parameters and rated operation variables based on rigorous theoretical analysis. This salient feature makes the proposed method more accurate than earlier methods. Finally, the proposed method is validated by simulations based on the PSCAD/EMTDC program.

Simultaneous enhanced ammonia and nitrate removal from secondary effluent in constructed wetlands using a new manganese-containing substrate

Constructed wetlands(CWs)are widely used to treat secondary ***,simultaneously removing ammonia(NH_(4)^(+)-N)and nitrate(NO_(3)^(–)–N)is challenging because of insufficient oxygen and carbon *** this study,a novel composite material(MPCM)comprising MnO_(2) and polycaprolactone was developed as a substrate for CWs to enhance the synchronous removal of NH_(4)^(+)–N and NO_(3)^(–)–*** CWs with a higher MPCM content(H-CW),lower MPCM content(LCW),and controlled CW(C-CW)exhibited average NH_(4)^(+)–N removal efficiencies of 75.69%,70.49%,and 52.40%,*** 15N isotope tracking technique showed that NH_(4)^(+)–N removal was attributed to anaerobic ammonia oxidation mediated by MnO2 reduction(Mnammox),which accounted for 17.16%–27.24%of the NH_(4)^(+)–N removal in the composite material layers(0–20 cm)of the H-CW and *** richness of ammonia oxidizers in the upper layers(40–50 cm)of the HCW and L-CW further facilitated NH_(4)^(+)–N ***,the average total nitrogen(TN)removal efficiencies of the H-CW and L-CW were 1.99 and 1.59 times that of C-CW,respectively,owing to enhanced denitrification by ***,N_(2)O emissions were reduced by 81.31%and 70.83%in the H-CW and L-CW,*** study provides an effective approach for improving nitrogen removal and reducing N2O emissions during the treatment of secondary effluent by CWs.

We study superradiant phase transitions in a hybrid system of a two-dimensional Bose–Einstein condensate of atoms and two cavities arranged with a tilt *** adjusting the loss rate of cavities,we map out the phase diagram of steady states within a mean field *** is found that when the loss rates of the two cavities are different,superradiant transitions may not occur at the same time in the two cavities.A first-order phase transition is observed between the states with only one cavity in superradiance and both in *** the case that both cavities are superradiant,a net photon current is observed flowing from the cavity with small decay rate to the one with large decay *** photon current shows a non-monotonic dependence on the loss rate difference,owing to the competition of photon number difference and cavity field phase *** findings can be realized and detected in experiments.

Deterministic inversion based on deep learning has been widely utilized in model parameters *** by logging data,seismic data,wavelet and modeling operator,deterministic inversion based on deep learning can establish nonlinear relationships between seismic data and model ***,seismic data lacks low-frequency and contains noise,which increases the non-uniqueness of the *** conventional inversion method based on deep learning can only establish the deterministic relationship between seismic data and parameters,and cannot quantify the uncertainty of *** order to quickly quantify the uncertainty,a physics-guided deep mixture density network(PG-DMDN)is established by combining the mixture density network(MDN)with the deep neural network(DNN).Compared with Bayesian neural network(BNN)and network dropout,PG-DMDN has lower computing cost and shorter training time.A low-frequency model is introduced in the training process of the network to help the network learn the nonlinear relationship between narrowband seismic data and low-frequency *** addition,the block constraints are added to the PG-DMDN framework to improve the horizontal continuity of the inversion *** illustrate the benefits of proposed method,the PG-DMDN is compared with existing semi-supervised inversion *** synthetic data examples of Marmousi II model are utilized to quantify the influence of forward modeling part,low-frequency model,noise and the pseudo-wells number on inversion results,and prove the feasibility and stability of the proposed *** addition,the robustness and generality of the proposed method are verified by the field seismic data.
