This study aims to investigate the distribution of HLA-B*27 subtypes in Southeast han population of China.95 HLA-B*27-positive patients diagnosed with Ankylosing Spondylitis(AS) were selected and compared with 73 HLA-B*27-positive healthy controls.All the study objects are from four regions(hangzhou,Lishui,Shaoxing and Zhuji) in Zhejiang Province.HLA-B*27 targeted DNA was amplified by Amplification-Refractory Mutation Systems,and then HLA-B*27 subtypes were characterized by using polymerase chain reaction amplification with a sequence-specific primer.Of 25 subtypes screened,four HLAB*27 subtypes were identified:HLA-B*2704,HLA-B*2705,HLA-B*2706 and HLA-B*2715,among which,HLA-B*2704 was the predominant one,with an average of 86.3%(82/95) in AS patients.The percentages of HLA-B*2704 subtypes varied in four regions:86.4%(19/22) in hangzhou,74.1%(20/27) in Lishui,96.6%(28/29) in Shaoxing and 88.2%(15/17) in Zhuji.HLA-B*2705 was relatively low in patients from four regions.HLA-B*2715,a rare allele,was found in 2 patients(2/27) from Lishui but not patients from the other three regions.Our studies show that the overall frequency of HLA-B*2704 and HLA-B*2705 in Zhejiang province is similar as other regions of China.The frequency of HLA-B*2704 in Shaoxing is the highest and close to the one in Taiwan.This study meets our hypothesis that the Chinese population have a large HLA-B*27 genetic diversity,and we observe such diversity within one province.Additionally,the finding of HLA-B*2715 only in patients from Lishui,a mountainous region with complex terrains different from other three regions,implies a potential association of HLA-B*2715 to specific geographic areas.
