Extracellular sulfatase-2(Sulf-2)influences receptor-ligand binding and subsequent signaling by chemokines and growth factors,yet Sulf-2 remains unexplored in inflammatory cytokine signaling in the context of rheumatoid arthritis(RA).In the present study,we characterized Sulf-2 expression in RA and investigated its potential role in TNF-α-induced synovial inflammation using primary human RA synovial fibroblasts(RASFs).Sulf-2 expression was significantly higher in serum and synovial tissues from patients with RA and in synovium and serum from hTNFtg *** sequencing analysis of TNF-α-stimulated RASFs showed that Sulf-2 siRNA modulated~2500 genes compared to scrambled *** Pathway Analysis of RNA sequencing data identified Sulf-2 as a primary target in fibroblasts and macrophages in *** blot,ELISA,and qRT‒PCR analyses confirmed that Sulf-2 knockdown reduced the TNF-α-induced expression of ICAM1,VCAM1,CAD11,PDPN,CCL5,CX3CL1,CXCL10,and *** studies identified the protein kinase C-delta(PKCδ)and c-Jun N-terminal kinase(JNK)pathways as key in the TNF-α-mediated induction of proteins related to cellular adhesion and *** of Sulf-2 abrogated TNF-α-induced RASF ***-2 knockdown with siRNA and inhibition by OKN-007 suppressed the TNF-α-induced phosphorylation of PKCδand JNK,thereby suppressing the nuclear translocation and DNA binding activity of the transcription factors AP-1 and NF-κBp65 in human ***,Sulf-2 expression positively correlated with the expression of TNF receptor 1,and coimmunoprecipitation assays demonstrated the binding of these two proteins,suggesting they exhibit crosstalk in TNF-α*** study identified a novel role of Sulf-2 in TNF-αsignaling and the activation of RA synoviocytes,providing the rationale for evaluating the therapeutic targeting of Sulf-2 in preclinical models of RA.

Natural and anthropogenic sources of lead, zinc, and nickel in sediments of Lake Izabal, Guatemala

Sediments in Lake Izabal,Guatemala,contain substantial lead (Pb),zinc (Zn),and nickel (Ni).The lack of historical data for heavy metal concentrations in the sediments makes it difficult to determine the sources or evaluate whether inputs of metals to the lake have changed through *** measured the relative abundances and concentrations of Pb,Zn,and Ni by X-Ray Fluorescence core scanning and by Inductively Coupled Plasma Optical Emission Spectrometry in three sediment cores to explore stratigraphic distributions of metals in the lake *** amounts of Pb and Zn in the core taken near the Polochic Delta suggest that galena and sphalerite mining increased Pb and Zn delivery to Lake Izabal between ~1945 and 1965 *** up-core Ni increase in the core taken near a different mine on the north shore of Lake Izabal suggests that recent nickel mining operations led to an increase in Ni concentrations in the local sediments,but amounts in the other cores indicate that Ni is not widely distributed throughout the *** cores from Lake Izabal are reliable recorders of heavy metal input to the lake,and were measured to establish background metal levels,which would otherwise be *** of Pb,Zn,and Ni in older,pre-20th-century Lake Izabal sediments reflect input from natural erosion of *** results provide previously unavailable estimates of background metal concentrations in Lake Izabal before the onset of *** results are necessary for future monitoring related to mining contamination of the lake ecosystem.

Evaluation of seismic potential in a longwall mine with massive sandstone roof under deep overburden

A recent seismic event was recorded by a deep longwall mine in Virginia at 3.7 ML on the local magnitude scale and 3.4 MMS by the United States Geological Survey(USGS) in *** investigations by the National Institute for Occupational Safety and health(NIOSh) and Coronado Coal researchers have shown that this event was associated with geological features that have also been associated with other, similar seismic events in *** mapping and geological exploration in the mining area has made it possible to forecast possible locations for future seismic *** order to use the geology as a forecaster of mining-induced seismic events and their energy potential, two primary components are *** first component is a long history of recorded seismic events with accurately plotted *** second component is a high density of geologic data within the mining *** this case, 181 events of 1.0 MLor greater were recorded by the mine's seismic network between January, 2009, and October, *** the mining area, 897 geophysical logs, 224 core holes, and 1031 fiberscope holes were examined by mine *** this information, it was found that overburden thickness, sandstone thickness, and sandstone quality contributed greatly to seismic *** the data was analyzed, a pattern became apparent indicating that the majority of seismic events occurred under specific *** forecast maps were created based on geology of previous seismic *** forecast maps have shown an accuracy of within 74%–89% when compared to the recorded 181 events that were1.0 MLor greater when considering three major geological criteria of overburden thickness of 579.12 m or greater, 6.096–12.192 m of sandstone within 15.24 m of the Pocahontas number 3 seam, and a longwall caving height of 4.572 m or less.

Reproductive behavior in horseshoe crabs:Does density matter?

While the four species of horseshoe crabs share many common reproductive traits with respect to their reproductive systems, they do differ with respect to their mating behavior （monogamy vs. polygynandry）. Past research has attributed these differences to a number of factors including： spawning densities, operational sex ratios （OSR＇s）, male condition （or age）, environmental and/or genetic factors, or a combination thereof. Mating behaviors in the three Asian horseshoe crab species （Tachy-pleus gigas, T. tridentatus, and Carcinoscorpius rotundicauda） with low spawning densities and 1：1 operational sex ratios are typically monogamous. In Limuluspolyphemus, mating behavior is more variable ranging fi＇om monogamy to polygynandry. here we provide evidence, through a long term behavioral study, that variation in mating behavior is influenced by population density in L. polyphemus. Our study population on two beaches in Connecticut （Long Island Sound） have a spawning density 400 times less than that found in Delaware Bay （0.002 females/m2 vs. 0.8 females/m2） but similar operational sex ratios. Between 90%-95% of all spawning females in CT were paired with only one male, thus exhibiting monogamous behavior. In contrast, between 30 and 60% of spawning females in Delaware Bay have more than one mate and produce clutches of eggs with multiple paternities. Male condition played no role in mating behavior in CT populations. We also observed that on average 18% of the females on the spawning beaches are single. These results suggest that population density is an important condition that determines mating behavior. Also, low population density may lead to decreased mate finding ability and lost opportunities for spawning

Proper cell-cell and cell-matrix contacts mediated by integrin adhesion receptors are important for development, immune response, hemostasis and wound healing. Integrins pass trans-membrane signals bidirectionally through their regulated affinities for extracellular ligands and intracellular signaling molecules. Such bidirectional signaling by integrins is enabled by the conformational changes that are often linked among extracellular, transmembrane and cytoplasmic domains. here, we review how talin-integrin and kindlin-integrin interactions, in cooperation with talin-lipid and kindlin-lipid interactions, regulate integrin affinities and how the progress in these areas helps us understand integrin-related diseases.

Effect of longwall-induced subsurface deformations on shale gas well casing stability under deep covers

This paper presents the results of a 2017 study conducted by the National Institute for Occupational Safety and health(NIOSh), Pittsburgh Mining Research Division(PMRD), to evaluate the effects of longwall-induced subsurface deformations within a longwall abutment pillar under deep cover. The 2017 study was conducted in a southwestern Pennsylvania coal mine, which extracts 457 m-wide longwall panels under 361 m of cover. One 198 m-deep, in-place inclinometer monitoring well was drilled and installed over a 45 m by 84 m center abutment pillar. In addition to the monitoring well, surface subsidence measurements and underground coal pillar pressure measurements were conducted as the 457 m-wide longwall panel on the south side of the abutment pillar was being mined. Prior to the first longwall excavation, a number of simulations using FLAC3D^(TM) were conducted to estimate surface subsidence, increases in underground coal pillar pressure, and subsurface horizontal displacements in the monitoring well. Comparisons of the pre-mining FLAC3D simulation results and the surface, subsurface,and underground instrumentation results show that the measured in-place inclinometer casing deformations are in reasonable agreement with those predicted by the 3D finite difference models. The measured surface subsidence and pillar pressure are in excellent agreement with those predicted by the 3D *** from this 2017 research clearly indicate that, under deep cover, the measured horizontal displacements within the abutment pillar are approximately one order of magnitude smaller than those measured in a 2014 study under medium cover.

Isola in a linear one-degree-of-freedom feedback system with actuator rate saturation

This short communication uses numerical continuation to highlight the existence of an isola in a simple one-degree-of-freedom harmonically forced feedback system with actuator rate limiting as its only nonlinear *** was found that the isola(1)contains only rate-limited responses,(2)merges with the main branch when the forcing amplitude is sufficiently large,and(3)includes stable solutions that create a second attractor in regions where rate limiting is not ***,the isola is composed of two solutions for a given forcing *** solutions have the same amplitudes in the state(pitch rate)projection;however,they have distinct phases,and their amplitudes are also distinct when projected onto the integrator state in the *** rich dynamics observed in such a simple example underlines the impact of rate limiting on feedback ***,the combination of feedback and rate limiting can create detrimental dynamics that is hard to predict and requires careful analysis.
